Trace element bioaccumulation in edible red seaweeds (Rhodophyta): A risk assessment for consumers

As a precursor to risk assessment and risk management through consuming contaminated seafood, food safety needs to be quantified and assured. Seaweed is an increasing dietary component, especially in developing countries, but there are few studies assessing uptake rates of contaminants from this route. As such, the present study determined likely human uptake due to the trace elemental (Fe, Mn, Ni, Cu, Zn, Se, Hg, and As) concentrations in the edible red seaweeds (Rhodophyta) Gelidium pusillum and Hypnea musciformis, growing in the industrialised Cox's Bazar coastal area of Bangladesh. Metal and metalloid concentrations in G. pusillum were in the order (mg/kg): Fe (797 ± 67) > Mn (69 ± 4) > Ni (12 ± 5) > Zn (9 ± 4) > Cu (9 ± 4) >Se (0.1 ± 0.1) > Hg (0.1 ± 0.01), and in H. musciformis: Fe (668 ± 58) > Mn (28 ± 5) > Ni (14 ± 2) > Zn (11 ± 5) > Cu (6 ± 4) >Se (0.2 ± 0.03) > Hg (0.04 ± 0.01). Despite the industrial activities in the area, and based on 10 g. day⁻¹ seaweed consumption, it is concluded that these concentrations pose no risk to human health as part of a normal diet according to the targeted hazard quotient and hazard index (THQ and HI) (values < 1). In addition, and as a novel aspect for seaweeds, Selenium Health Benefit Values (Se-HBV) were determined and found to have positive values. Seaweed can be used as an absorber of inorganic metals for removing contamination in coastal waters. The results are a precursor to further research regarding the efficiency and rate at which seaweeds can sequester metal contamination in water. In addition, management techniques need to be developed thereby to control the contaminant inputs.

Antibiotics used in global aquaculture production cause various side effects, which impair fish health. However, the use of dietary composition such as carbohydrate, which is one of the dominant components in fish diets to attenuate the side effects induced by antibiotics, remains unclear. We determined the ability of high carbohydrate diet to protect Nile tilapia (Oreochromis niloticus) from oxytetracycline-induced side effects. Triplicate groups of thirty O. niloticus (9.50 ± 0.08 g) were fed on medium carbohydrate (MC; 335 g/kg) and high carbohydrate (HC; 455 g/kg) diets without and with 2.00 g/kg diet of oxytetracycline (80 mg/kg body weight/day) hereafter, MCO and HCO for 35 days. Thereafter, we assessed growth performance, hepatic nutrients composition and metabolism, microbiota abundance, immunity, oxidative and cellular stress, hepatotoxicity, lipid peroxidation and apoptosis. To understand the possible mechanism of carbohydrate protection on oxytetracycline, we assessed the binding effects and efficiencies of mixtures of medium and high starch with oxytetracycline as well as the MCO and HCO diets. The O. niloticus fed on the MCO and HCO diets had lower growth rate, nutrients utilization and survival rate than those fed on the MC and HC diets, respectively. Dietary HCO increased hepatosomatic index and hepatic protein content of O. niloticus than MCO diet. The O. niloticus fed on the HCO diet had lower mRNA expression of genes related to protein, glycogen and lipid metabolism compared to those fed on the MCO diet. Feeding O. niloticus on the HCO diet increased innate immunity and reduced pathogenic bacteria, pro-inflammation, hepatotoxicity, cellular stress and apoptosis than the MCO diet. The high starch with oxytetracycline and HCO diet had higher-oxytetracycline binding effects and efficiencies than the medium starch with oxytetracyline and MCO diet, respectively. Our study demonstrates that, high carbohydrate partially protects O. niloticus from oxytetracycline-induced side effects by binding the antibiotic. Incorporating high carbohydrate in diet formulation for omnivorous fish species alleviates some of the side effects caused by antibiotics.

Microalgae have been used as live food in aquatic species. In recent years, the interest in microalgae has considerably increased, thanks to the evolution of production techniques that have identified them as an ecologically attractive aquafeed ingredient. The present study provides the first data about the effects of dietary inclusion of a microalgae consortium grown in a high-rate algal pond system on zootechnical performance, morphometric indices, and dietary nutrient digestibility as well as morphology and functionality of the digestive system of European sea bass, Dicentrarchus labrax. A dietary treatment including a commercial mono-cultured microalgae (Nannochloropsis sp.) biomass was used for comparison. Six hundred and thirty-six European sea bass juveniles (18 ± 0.28 g) were randomly allotted into 12 experimental groups and fed 4 different diets for 10 weeks: a control diet based on fish meal, fish oil, and plant protein sources; a diet including 10% of Nannochloropsis spp. biomass (100 g/kg diet); and two diets including two levels (10% and 20%) of the microalgal consortium (100 and 200 g/kg diet). Even at the highest dietary inclusion level, the microalgal consortium (200 g/kg diet) did not affect feed palatability and fish growth performance. A significant decrease in the apparent digestibility of dry matter, protein, and energy was observed in diets including 10 and 20% of the microalgal consortium, but all fish exhibited a well-preserved intestinal histomorphology. Moreover, dietary inclusion with the microalgal consortium significantly increased the enzymatic activity of maltase, sucrase-isomaltase, and ɤ-glutamil transpeptidase in the distal intestine of the treated European sea bass. Algal consortium grown using fish farm effluents represents an attempt to enhance the utilization of natural biomasses in aquafeeds when used at 10 % as substitute of vegetable ingredients in diet for European sea bass.

Dietary composition and husbandry practices largely determine the essential trace element status and the toxic metal exposure of livestock and consequently their concentrations in animal products. The main objective of this study was to determine the real contamination of selected areas of Slovakia compared with existing Environmental regionalization of the Slovak Republic (SR) created by the Ministry of the Environment SR. Another aim of this study was to determine the content of essential and toxic metals in feed and milk of dairy cows on selected farms in Slovakia referred to the suitability of the use of milk from these areas to other food processing. This article deals with the analysis of the content of selected elements in feed and milk of dairy cows in the area of Novoť (Northern Slovakia; area with undisturbed environment) and Čečejovce (Eastern Slovakia; highly disturbed environment). Eleven elements have been analyzed (essential elements: calcium, zinc, magnesium, selenium, iron, copper; toxic elements: arsenic, cadmium, mercury, lead, nickel). Samples of feed and milk were collected five times during the spring season and five times during the autumn season in 2016. Analysis of samples was performed in Eurofins Bel/Novamann (Nové Zámky, Slovak Republic). Analyses were performed by atomic absorption spectroscopy and atomic emission spectrometry. The obtained values of the individual elements in the feed and milk were evaluated by Student’s t test. Significantly higher contents of essential elements Ca, Zn, Fe, Mg, and Cu were found in the feed of dairy cows in Čečejovce (P < 0.001). Significantly higher (P < 0.001) As and Ni content in feed in Čečejovce in autumn season was also recorded. However, the content of these elements in feed did not affect their milk content. The content of Ca, Zn, and Mg in milk was significantly (P < 0.001) higher in Novoť in both investigated seasons. Some essential elements and toxic elements in feed and milk were below the LOQ (limit of quantification). Reduced content of Ca and Zn in milk in both farms compared with optimal limits, decreased Ca and Mg content in feed in Novoť, and a significant increase of Ca and Mg in feed in Čečejovce in autumn season were found. It can be concluded, due to the low content of toxic elements on these farms, that the use of milk of dairy cows from these areas for direct use or for dairy product processing is appropriate and poses no health risk to the consumers.

Terpenes in citrus oil must be removed to stabilize the products and to dissolve it in aqueous solution. Supercritical fluid extraction has been investigated for the terpeneless citrus oil processing as a lower temperature process. In order to achieve higher yield and higher separation selectivity, a continuous countercurrent extraction with reflux was studied at a temperature of 333 K and a pressure of 8.8 MPa. Cold-pressed orange oil from Brazil and a model mixture of 80% limonene and 20% linalool, where limonene and linalool are principal constituent of terpenes and oxygenated compounds in orange oil, were used as feed and carbon dioxide was used as solvent. Operation at total reflux was carried out to calculate the minimum number of plates required to achieve a separation between limonene and linalool. Effects of the solvent-to-feed ratio, reflux ratio, and feed inlet position on the yield and selectivity were investigated for continuous operation. The selectivity increased with the increase in the solvent-to-feed ratio. Terpeneless citrus oil was obtained on the operation at the higher solvent-to-feed ratio and longer stripping section.

Capsaicin (CPS, 8-methyl-N-vanillyl-trans-6-nonenamide), a pungent alkaloid from chili peppers, has contradictory effects in both experimental and human carcinogenesis. Thus, we evaluated the modifying effects of chronic CPS during the promotion and progression stages of rat colon carcinogenesis induced by 1,2-dimethylhydrazine (DMH). Male Wistar rats were given four subcutaneous injections of DMH (40 mg/body weight (b.w.)) twice a week, for 2 weeks. After DMH-induced tumor initiation, the animals were treated with CPS at 5 or 50 mg/kg b.w. by gavage for 24 weeks (three times a week). High-dose CPS reduced both cell proliferation in adjacent “normal-appearing” colonic crypts and the total number of preneoplastic aberrant crypt foci (ACF) but did not change the number of dysplastic ACF or ACF multiplicity. Although the proportion of adenomas was increased, and tubular adenocarcinomas decreased in high-dose CPS, both CPS interventions exerted no effects on total tumor incidence, volume, multiplicity, cell proliferation (Ki-67), and apoptosis (caspase-3). In accordance, high-dose CPS treatment had discrete effects on gene expression in colon tumors, as only 3/94 (3.19%) genes were significantly modified (downregulation of Cebpd and Fasl, and upregulation of Jag1). The findings of the present study show that CPS does not impact on the promotion/progression stages of rat colon carcinogenesis. Therefore, CPS at a high-dose intervention showed to be a safe food ingredient.

Endocrine disruptors have gained widespread attention owing to their severe adverse health impacts. These produce enormous burden of disease and are associated with high economic cost especially in developed countries. Environmental pollutants causing endocrine disruption include pesticides, industrial wastes, packaging materials, food constituents, plastics, and cosmetic products. Likewise, pharmaceutical drugs have the endocrine disrupting potential through a wide array of mechanisms. Antipsychotic, antiepileptic, antihypertensive, antiviral, antidiabetic, and anticancer drugs are among the foremost non-hormonal endocrine disruptors. Several drugs affect thyroid hormone synthesis via interaction with iodine uptake to the release of T3 and T4 by thyrocytes. Prolonged use of some drugs increase susceptibility to diabetes mellitus either by direct destruction of β cells or enhanced insulin resistance. Other drugs may cause serious developmental defects in male or female reproductive system. Appropriate understanding of the mechanisms of endocrine disruption associated with non-hormonal drugs will guide future drug development and help us prevent and cure endocrine-related toxicity of pharmaceuticals. Therefore, this review focuses on endocrine disruption by pharmaceutical drugs as their side effect.

The application of the SRK equation of state for the prediction of the phase behaviour of mixtures of vegetable oils with near critical solvents is studied. The use of binary interaction parameters in the combinatorial rules for both the co-volume and the energy parameter is discussed. Two different sets of binary interaction parameters are needed in order to correlate the vapor-liquid and liquid-liquid equilibria. This indicates a serious limitation of van der Waals type of equations of state for modeling the phase equilibria of this type of systems.

Supercritical CO2 extraction of oleoresin from portuguese rosemary (Rosmarinus officinalis L.) was carried out with a flow apparatus at temperatures of 35 and 40 degrees C and pressures of 100, 125 and 200 bar. The highest fraction of volatile compounds (oil) in the oleoresin was obtained at 100 bar and 40 degrees C. An unsteady mathematical model was able to give good representation of the supercritical extraction curves and a mass transfer coefficient was determined using the successive quadratic programming method. Values of this coefficient ranged between 4.49 and 15.07 kg/m3s and the shift to a diffusion-controlled regime occurred when 44% of the total oil was extracted.
