The effect of substances affecting cAMP and cGMP metabolism on various types of activation and the spontaneous rhythmic activity of the isolated rat uterus [laboratory animals]

This paper investigates the effect of organic nitrates on the isolated rat uterus. The following was established: -Nitro-compounds(sodium azide, sodium nitroprusside and nitroglycerol) cause dose-dependant inhibition of the isolated uterus, which was actived by oxytocin, electrical stimulation and high KCl concentration. Nitro-compounds and isoprenaline inhibit more intensively the tonic, rather than phase component of the contraction caused by oxytocin. -Sodium nitrite did not change significantly the response to oxytocin, high KCl concentration and electrical stimulation. -In the isolated uterus of the animals proviously treated with nitroglycerol a low degree of tolerance was noticed only after the application of low nitroglycerol concentrations during electrical muscle stimulation. There was no tolerance to higher nitroglycerol concentrations during electrical stimulation and KCl activation. -Methylene blue does not antagonize the relaxing effect of nitro-compounds on the isolated rat uterus. -Isoprenaline causes inhibition of various types of activation of the plain muscle. -The strongest inhibition was also noticed in the tonic contraction component caused by oxytocin. -Aminophylline inhibits spontaneous rhythmical activity of the isolated rat uterus. -Sodium azide, sodium nitroprusside, nitroglycerol, isoprenaline and aminophylline inhibit spontaneous rhythmical activity of the isolated rat uterus. It was possible to antagonize the nitro-compounds effects by increasing calcium in the medium.
