Detection of bacterial phytopathogens based on nucleic acid technology

Detection systems, based on nucleic acid sequence recognition, were designed for the quarantine bacterial phytopathogens Xanthomonas campestris translucens and Erwinia amylovora. By comparative sequence analysis of ribosomal DNA regions of these bacteria and of taxonomically related phytobacteria, target specific oligonucleotides were defined. Used as primers in PCR, one oligonucleotide combination specifically recognized the Xanthomonas campestris pathovars belonging to the taxonomic groups translucens and graminis, producing a 139 bp PCR fragment. The pathogen was detected to the limit of 2000 bacteria per gram of contamined cereal seed. Another primer combination was found specific for the species Erwinia amylovora, amplifying a 565 bp fragment. This detection system was used in Erwinia amylovora migration experiments in the field and in the diagnosis of diseased plant sample material.
