Simple staining method of chicken embryonic skeleton

A new method of staining of rat fetal skeleton was modified and applied to staining of chicken embryonic skeleton. This method was simple and able to prepare chicken skeletal specimens with good stain. The method was as follows. The chicken embryo was fixed in 99% ethanol for at least one week, the skin of the head and thoracoabdominal region was removed, and the yellow fat and viscera were removed. The specimen was cleared and stained in a mixture, consisting of 1.5% KOH, 85% ethanol and 0.004% alizarin red S, for 4-5 days, followed by the second clearing in 1% KOH for 2-3 days. Next, the specimen was transferred to a 50% glycerin aqueous solution to destain overstained portions, and the specimen was used for observation. In this method, removal of the skin of the leg region is not necessary and almost no removal of the muscle is necessary. This method is thought to be clearly simpler and more efficient than the conventional method
