The dynamic behaviour and kinetics of glucose oxidase immobilized packed column integrated with amperometric electrode

A high sensitive amperometric biosensor was developed for specific determination of D-glucose. Glucose oxidase (GOD) was immobilised by adsorption on weak basic ion exchange resin (Duolite A568). A probe type hydrogen peroxide electrode polarised at 650 mV was used. Electrode surface was covered with plain cellulose acetate membrane to eliminate interferences of electroactive compounds which may found in food samples. Different flow rates for packed column were tested. Km and Vmax values of free and immobilised GOD enzyme were determined. The Km of GOD enzyme was significantly increased upon immobilisation from 0.197 to 0.712 mM D-glucose. On the other hand Vmax value decrees from 1.55 to 1.09 mM per min. The response time of the electrode was in range of 15 s and 5 min (depend on the sensitivity). The maximum sensitivity and linearity were determined 83.6 nA/mM and 1.25 mM D-glucose respectively.
