Callus cultures were initiated from mature nodal segments of mulberry (Morus indica L.) cultivar S13 on Murashige and Skoog (MS) medium supplemented with 1 mg/l 2,4,5-trichlorophenoxy acetic acid (2,4,5-T). Protoplasts were isolated from 20 days old callus cultures incubated in digestion solution containing 0.25 / of macerozyme R-10 and 1 / of cellulase "Onozuka" R-10 after 4h of digestion. Furthermore, a new procedure is developed for isolation and purification of mulberry callus protoplasts
