In our investigation we prepared two tests: enzyme-linked immunosorbent assay (ELISA), and immunoblot, based on local Borrelia burgdorferi strains, and compared to indirect immunofluorescence assay and commercially available Behring ELISA test. The immunoblot was the most specific assay, with specificity of 96%. The specificity of ELISA was 93%, and 89% of immunofluorescence assay. The sensitivity of tests varied with the stage of disease. ELISA showed higher specificity and sensitivity compared to immunofluorescence assay, and should be a screen assay for serodiagnosis of Lyme disease. Comparison of home made and Behring ELISA tests showed that the sensitivity of serological assay could be increased when the test is based on local Borrellia burgdorferi strains. Analysis of the test specificity and sensitivity showed that the immunoblot based on local Borrelia afzelli strain was the most sensitive and specific assay ad should be a confirmatory test for immunodiagnosis of Lyme disease.
