Cultivation of wild mushroom: VI Hed Hu Hnu (Auricularia spp.)

Basidiocarps of Hed Hu Hnu (Auricularia spp.) were collected from different locations and isolated for dikaryotic mycelium by tissue culture. The morphological study revealed that isolates A1 and A2 were Auricularia polytricha (Mont.) Sacc., isolated A3, A4 and A5 were A. fuscosuccinea (Mont.) Farl., while isolate A6 was not identified. Mycelial growth of each isolates was tested on 5 agar media. The results showed that V-8 juice agar was the best in supporting mycelial growth. Isolate A2, A3 and A5 were used to study some physiological aspects and yield comparison. A. polytricha (A2) grew well on media containing cellulose, glucose, soluble starch or sticky rice flour as carbon source. In term of nitrogen source, the best yield was obtained on media containing pepton, ammomium sulfate or ammonium chloride. The optimum temperature and pH for mycelial growth in malt extract medium were 25-30 deg C and 5-9, respectively. Light slightly inhibited mycelial growth of A. polytricha. For cultivation, the method of growing mushroom in autoclavable plastic bags was applied. The higher yield was obtained on para rubber sawdust supplemented with 6 percent rice bran and 1 percent Ca (OH)2. The time required for spawn running was 55.8 days in 1 kg of substrate. Yield obtained was 301.3 gm/bag (B.E.=86.1 percent) in a period of 80 days. A. fuscosuccinea (A3 and A5) grew well on media containing glucose or sticky rice flour as carbon source. Fructose was also good carbon source for mycelial growth of isolate A5. Both two isolated utilized peptone better than other nitrogen source. The optimum temperature and pH for mycelial growth was 25-30 deg C and 6-8, respectively. Light strongly inhibited mycelial growth of A. fuscosuccinea. The time required for spawn running of isolate A3 was 95.0 days and everage yield obtained during 80 days of watering was 95.6 gm/bags (B.E.=27.3 percent). Isolate A5 needed shorter time for full colonized the substrate and produced higher yield. Time required for spawn running was 49.3 days and produced 256.5 gm of basidiocarps during 80 days of watering (B.E.=73.3 percent).
