Genetic relatedness of ampicillin resistant Streptococcuspneumoniae clones from Saudi Arabia to wild strains

In the present work studies of 150 local Streptococcus pneumoniaeisolates from different hospitals in Riyadh, by polymerase chainreaction (PCR) techniques revealed 18 clones to be ampicillin resistant.The genetic relatedness of 18 (12%) of the 150 clones ampicillinresistant isolates to 9 wild type ampicillin resistant clones wasdetermined through Random Amplified Polymorphic DNA (RAPD).Similarity matrix and phylogenetic tree of the RAPD types showed thatthe ampicillin resistant wild type were not closely related to ampicillinresistant local isolates. The cluster analysis by unweighed-pairedgroup of arithmetic mean (UPGMA) also does not indicated geneticproximity between wild type and the local isolates. The present studyshows that there are no new clones identified. The local clones werenot very closely genetically related to each other. The local sp. 29and local sp. 36 were the two most related strains among the 18strains with value (0.32) in the similarity range. While differentinternationally widespread ampicillin resistant clones exist in theKingdom, in this study there is no common clone spread in the areawas found. This study will help in the development of awarenessabout the hazards caused by the resistant clones.
