Detection and molecular characterization of Grapevine fanleaf virus and Grapevine leafroll-associated virus 3 in Jordan [Vitis vinifera L.]

Английский. Grapevine fanleaf virus (GFLV) and Grapevine leafroll-associated virus 3 (GLRaV-3) have been detected by DAS-ELISA in six locations where grapevines are commonly grown in Jordan. Using pairs of specific primers, fragments of the RNA-dependent RNA polymerase (RdRp) gene of GLRaV-3 and the coat protein (CP) gene of GFLV were amplified from symptomatic grapevine tissues by IC-RT-PCR. After cloning and sequencing, sequences of the amplified fragments were deposited in Genbank. Alignment analysis revealed that the amplified CP gene fragment of the Jordanian isolate of GFLV (GFLV-JOR) shared sequence identity between 82 and 88% with 11 GFLV isolates from different parts of the world. High sequence identity (95-99%) was observed between the amplified fragment of RdRp gene of GLRaV-3 from Jordan (GLRaV-3-JOR) and isolates from Brazil, USA, Czech Republic and China

[Il Virus della foglia a ventaglio della vite (GFLV) e il Virus 3 associato all'accartocciamento fogliare della vite (GLRaV-3) sono stati individuati mediante DAS-ELISA in sei località viticole della Giordania. Utilizzando paia di primer specifici, da tessuti di viti sintomatiche sono stati amplificati, mediante IC-RT-PCR, frammenti del gene della RNA polimerasi (RdRp) RNA-dipendente del GLRaV-3 e il gene della proteina di rivestimento (CP) del GFLV. Dopo la clonazione e il sequenziamento, le sequenze dei frammenti amplificati sono state depositate nella GenBank. L'analisi dell'allineamento ha messo in evidenza che il frammento amplificato del gene della CP dell'isolato giordano del GFLV (GFLV-JOR) condivideva l'identità della sequenza fra l'82 e l'88% con 11 isolati di GFLV provenienti da parti diverse del mondo. E' stata osservata un'elevata identità di sequenza (95-99%) fra il frammento amplificato del gene RdRp del GLRaV-3 giordano (GLRAV-3-JOR) e isolati provenienti da Brasile, USA, Repubblica Ceca e Cina]
