Potential application of a cell-bound polysaccharide produced by the marine bacterium Zoogloea sp.

Marine microorganism producing polysaccharides, was newly isolated from costal sea of Korea and was identified as Zoogloea sp. (KCCM 10036). It has been reported that Zoogloea sp. produce only one type of polysaccharide, however, this strain (KCCM 10036) could produce two different polysaccharides (WSP, Water soluble polysaccharide, CBP, Cell bound polysaccharide). These polysaccharides were new types as compared with previously reported ones. In this study, we examined the basic properties of these polysaccharides. Furthermore, for the application of these polysaccharides the solution properties of CBP was observed. When CBP was dissolved in distilled water, the solutions showed the most high value of viscosity and showed a property of pseudoplastic non-Newtonian fluid above the concentration of 0.4 percent (w/v). The antitumor activity of CBP was observed. CBP showed a direct cytotoxic effect and tumor growth inhibition on Meth-A cells and Sarcoma-180 cells. CBP was also used for the adsorbent of metals and a new support of enzyme immobilization. CBP gel beads showed a high effective adsorption in Cr, Pb, Fe ion solutions. The adsorption rates were above 95 percent pH 5.0, 25 deg /c in 10 mg/ml of each metal solutions. The gel beads formed by CBP were stable at the range of pH 4.0 - 7.0 and temperature 40 - 55 deg C. The immobilized glucoamylase produced 10.5 mg/ml of glucose from 10 mg/ml of soluble starch.
