In order to determining of relative resistance of alfalfa cold region ecotypes to a fungal disease, alfalfa downy mildew, an experiment was carried out with four replications in RCBD with 15 treatments including 13 alfalfa cold ecotypes, one relative susceptible ecotype (Yazdi) and one relative resistant ecotype (Nikshahri) as controls at four locations including Karaj (Greenhouse test), Hamadan, Gorgan and Zanjan (Field test in Natural conditions) during 2008 and 2009. The results of the project showed that Yazdi, Hamadani and Siahe Boomie Lorestan ecotypes with %67.4, %55.3 and %43.9 Disease Severity (DS) averages respectively had the most susceptibility and Nikshahri, Malek-kandi and Ghargholoogh with %18.4, %26.8, %26.3 DS averages respectively had the most tolerance to disease. The results of analysis of variance showed that there is highly significant (á = %1) difference among ecotypes based on their responces to disease but there isn't any significant difference between locations. These results were relative similar to greenhouse test as Yazdi and Hamadani were the most susceptible and Ramandi and Nikshahri were the most tolerant ecotypes. Consequently using some ecotypes like Malek-kandi, Ghargholoogh and Ramandi is suggested for next researches in order to release the cultivars, because of their suitable yield and agronomic traits and resistance to downy mildew.
