Китайский. 以6个不同基因型的玉米自交系幼嫩叶段为外植体,研究其愈伤诱导、继代和分化过程。结果表明,3mg/L 2,4-D是诱导叶段愈伤组织较为适宜的,玉米愈伤组织的诱导及分化与基因型有密切关系。从供试的6个基因型中筛选出出愈率较高、愈伤组织形态较好的3个基因型齐319、Mo17、鲁原92,对它们的愈伤组织进行GUS瞬时表达检测表明,来源于幼苗叶段的愈伤组织GUS+转化率较高。
Показать больше [+] Меньше [-]Английский. The initiation, maintenance and differentiation of callus from seedling-derived leaf segments of six maize genotypes had been studied. It was revealed that the optimum concentration of 2, 4-D for the leaf segments-induced callus was 3 mg/L. Callus induction and regeneration were dependent on the genotypes. Due to their better performance in callus induction and morphology, embryogenic callus of Qi319, Mo17 and Luyuan 92 were transformed with a GUS construct. The transient expression of GUS was performed by the histochemical staining assay. The results showed that the GUS+ frequency of young leaf segments-derived callus was high. This study provides technological reference for selection of leaf segment as an efficient explant for maize regeneration and genetic transformation.
