Identification of 'Bursa si̇yahi' fig cultivar grown in Bursa using SSR molecular markers

The purpose of this research was to prove orginality of the fig that is 'Bursa Siyahı' cultivated in Bursa, doing genetic screening via SSR (simple sequence repeat) molecular marker technique. In this study, 'Bursa Siyahı' samples were collected from 3 regions, 11 different locations in Bursa. Beside this, by supplying plant samples which were identified as 'Bursa Siyahı' from a collection of genetic resources in Fig Research Institute in Aydın, were included in this research. The DNA samples were isolated from fresh leaves of each plant picked from Bursa Region. In addition to this leaves' samples, also 'Bursa Siyahı'fruit samples were examined for using in various pomological analysis and the results were evaluated in terms of statistics. MFC3, MFC4, MFC5, MFC6 primers were utilized in SSR analysis. Polymorphism wasn't observed in the bands obtained from molecular analysis results. All samples collected from gardens were also genetically identified as kind of black fig known as 'Bursa Siyahı'. As a result of pomological analysis, these outcomes were determined; 'Bursa Siyahı' fruits average weight is 82,6 g, average width is 56,1 mm, average height is 48,0 mm, average neck height is 7,8 mm, average ostium aperture is 6,4 mm, fruit peel thickness is 4,0 mm, average value is 4,55 pH, average TSS (total soluble solids) value is 17,4 brix. In the analysis of color measurement, CIE L *, a * and b * color values were obtained as respectively 29,70 L*, 6,49 a* and -0,70 b* in the peel of fruit; as respectively 44,04 L*, 12,93 a* and 16,97 b* in the interior color of fruit. Consequently, the results of the pomological analysis of fruit were supportive of the molecular results.
