[The adventitious regeneration in peach and other stone fruit species [Prunus persica (L.) Batsch - Prunus avium L. - Prunus domestica L.]]

An efficient regeneration protocol has been defined to induce organogenesis from micropropagated shoots of the plum rootstock Ferdor (Prunus insititia x domestica), some peach [P. persica (L.) Batsch] genotypes (cvs Babygold 6, Yumyeong, San Giorgio, Marylin, White Globe and the selection 842 Standard) and the cherry cultivar Lapins (P. avium L.). Apices were excised from in vitro shoot cultures and cultured 21 days in dark on a LP medium supplemented with BA and NAA, and then transferred to an auxin free medium in the light for 21 days. The first apical leaves were excised from these apices and cultured in the same way. During dark incubation, leaves developed a callus at the petiolar base. Adventitious shoots appeared on this callus after transfer to auxin free medium in the light. A shorter protocol was applied to the shoot apices of cherry cv Lapins in order to evaluate the regeneration response. Adventitious shoots were obtained also from pre-conditioned apices of Ferdor and Lapins

[E' stato messo a punto un protocollo efficiente di rigenerazione per indurre l'organogenesi da germogli micropropagati del portinnesto di susino Ferdor (Prunus insititia x domestica), alcuni genotipi (cv Babygold 6, Yumyong, San Giorgio, Marylin, White Globe e la selezione 842 Standard) di pesco [Prunus persica (L.) Batsch]. Apici prelevati dalle colture di germogli in vitro sono stati coltivati per 21 giorni al buio su un terreno LP addizionato di BA e NAA, successivamente sono stati trasferiti in un substrato senza auxine alla luce, per 21 giorni. Le prime foglie apicali sono state tagliate da questi apici e coltivate secondo il medesimo protocollo. Nel corso dell'incubazione al buio, le foglie hanno sviluppato un callo alla base del picciolo. Dopo il trasferimento al substrato senza auxine alla luce, sul callo sono comparse radici avventizie. Agli apici dei germogli della cv di ciliegio Lapins e' stato applicato un protocollo abbreviato per valutate la risposta in termini di capacita' di rigenerazione. Sono stati ottenute radici avventizie anche dagli apici pre-condizionati di Ferdor e Lapins]
