Research on biodiversity is crucial in the current dynamics of environmental modification. To optimize results is necessary to apply multiple techniques and standardize them, to increase efficiency. The analysis of the exochorionic structure of Lepidoptera provides a useful information system for taxonomic discrimination at different hierarchical levels. In this study, three abdominal rehydration techniques are described and tested utilizing samples deposited in scientific collections. The effectiveness and costs of the techniques was tested by comparing the results with the morphology of preserved exochorion with freshly obtained samples. Using these techniques over museum specimens could be an advance in the study of this system of morphological characters in female butterflies since it would increase the knowledge about rare, cryptic, and/or littlesampled species. Besides, it would increase the amount of data on the geographical and temporal variation of the chorionic characters. Although the results show that fresh material is the best option of rehydration techniques allow recovering the exochorion in acceptable conditions for analysis and comparisons, but it needs more time to clean these samples. The best rehydration technique was the use of proteinase K, since it obtained the most conserved eggs, even with specimens collected 30 years ago.
