BACKGROUND: Hydatid cyst is an infection with global distribution that is caused by the larval stage of the tapeworm of Echinococcus. The long-term survival of the hydatid in the host shows the parasite has advanced highly effective strategies for escaping the host defense. Deaths are caused by parasitic infections which are often due to tissue damages that result in host cell death, this is known as apoptosis. So it is important to know the process and the role of apoptosis that is created or controlled by a parasite. OBJECTIVES: Investigation of cytotoxicity effect, induction of apoptosis and mechanism of induction of apoptosis of cattle hydatid fluid on bovine lymphocyte cells as efficient cells of immunity were studied. METHODS: In this study, the cytotoxicity effect of bovine hydatid fluid (HF) on lymphocyte cells was investigated as effective immune cells against Echinococcus granulosus by MTS method. Then the mean of the expression of caspase-3, Bax and Bcl-2 genes in bovine lymphocytes treated/untreated was determined with fertile and infertile hydatid cyst fluid using Real Time PCR method. RESULTS: The viability mean of lymphocytes was significantly lower in the fertile HF treated lymphocytes compared to both infertile HF-treated lymphocytes and cell control. Bax gene expression was significantly (P=0.046) higher in the fertile HF-treated lymphocytes compared to both infertile HF-treated lymphocytes and cell control. Although Caspase 3 was higher in this group, the difference was not significant. Also, expression of Bcl-2 gene in fertile fluid treated lymphocytes was found to be lower than that of infertile and control. CONCLUSIONS: Present study indicates that hydatid cyst fluid molecules can probably induce apoptosis in immune cells in vitro and the parasite’s ability to stay alive for a long time in the host by controlling the host immune response from the apoptosis pathway.

Apoptosis plays a role in many events such as the remodeling of cells starting from embryo formation, the elimination of faulty or dysfunctional cells, and is important for cellular balance. Bcl-2 family and various mediators, of which caspases are the basis, play a role in maintaining this balance in apoptosis. These mediators are important in the initiation, continuation and arrest of apoptosis pathways, and in the regulation of cellular balance. Damages to these mechanisms can cause undesirable results and diseases. In this review, we tried to give information about the pathways of apoptosis and the basic molecules involved in the regulation of apoptosis.

The OLR system aims to eliminate these factors by housing pigeons from various lofts in equal conditions in one facility. This in turn, however, fosters inter-individual transmission of pathogens.

Ochratoxin A (OTA) is a mycotoxin notably produced by Aspergillus and Penicillium spp. Bacillus subtilis fermentation extract (BSFE) contains specific enzymes which hydrolyse OTA. This study evaluated the efficiency of BSFE in ameliorating the immunotoxic and nephrotoxic effects of OTA in broiler chickens. Day-old broiler chicks were divided equally into four groups of ten: control, OTA (0.5 mg/kg feed), BSFE product (1 mL/L water) and OTA + BSFE at the same concentrations. The chicks were vaccinated against avian influenza, Newcastle disease, and infectious bronchitis, and lymphoproliferation was induced in all birds by phytohaemagglutinin-P (PHA-P). Serum samples were taken before sacrifice and organ tissue samples were taken after, in which renal function biomarkers were assayed and the presence of OTA residue was evaluated by high-performance thin-layer chromatography. Protein markers of apoptosis were determined by qPCR, and tissue lesions were examined histopathologically. Exposure to OTA significantly decreased the antibody response to the vaccines and the lymphoproliferative response to PHA-P, and significantly elevated the renal function indicators: serum urea, uric acid and creatinine. It also induced oxidative stress (reduced catalase activity and glutathione concentration), lipid peroxidation (increased malondialdehyde content), apoptosis (increased Bax and Caspase-3 and decreased Bcl-2 gene levels) and pathological lesions in kidney, bursa of Fabricius, spleen and thymus tissue. Residues of OTA were detected in the serum and tissue. BSFE mitigated most of these toxic effects. BSFE counters OTA-induced immunotoxicity and nephrotoxicity because of its content of carboxypeptidase and protease enzymes.

The aim of the study was to describe the process of neuron death in the cerebral cortex caused by embryonic carbofuran exposure. 81 mouse foetuses from 27 breeding mice were used in the study. Carbofuran was administered by gavage from the 6ᵗʰ to the 15ᵗʰ day of gestation to two groups: one at 0.0208 and the other at 0.0417 mg/kg b.w. On the 17ᵗʰ day, the mice were sacrificed and the foetuses were taken to measure the ROS (malondialdehyde/MDA and superoxide dismutase/SOD) activity in brain tissue, the number of apoptotic embryonic cerebral cortex neurons using a TUNEL assay, and necrotic cells using HE staining. Examination of p53 and caspase 3 expression was done by immunohistochemistry. Data were analysed using analysis of variance (ANOVA) and Duncan’s test. Increased activity of cerebral ROS characterised by significant elevation of the MDA level (P < 0.05), decreased SOD (P < 0.01), increased p53 and caspase 3 expression, and cerebral cortical neuron death either by necrosis or apoptosis (P < 0.05) were found. At the low dose carbofuran increased expression of p53, caspase 3, and apoptosis. At the high dose it increased levels of MDA and necrosis. Increased expression of p53 and caspase 3 and apoptosis indicated that carbofuran may cause apoptosis through the intrinsic pathway. The increased apoptosis grants an opportunity to prevent and treat the effect of ROS due to gestational carbofuran exposure.

Introduction: Thyroid hormones play a major role in the regulation of testicular maturation and growth and in the control of Sertoli and Leydig cell functions in adulthood. When naturally occurring, hypothyroidism causes male hypogonadotropic hypogonadism and Sertoli cell function disorders, but when iatrogenic and methimazole-induced its influence on the pituitary-testicular axis function with respect to Sertoli cells is poorly known. Material and Methods: Male adult Wistar rats (n = 14) were divided into two groups: E – taking methimazole orally for 60 days, and C – control animals. After 60 d, the concentrations in serum of testosterone, follicle-stimulating and luteinising hormones, and inhibins A and B were measured. Testicles were examined morphologically: the apoptotic Sertoli cell percentage (ASC%) and number of these cells functional per tubular mm² (FSCN/Tmm²) were calculated. Results: In group E, inhibin A was higher while inhibin B was lower than in group C. ASC% was higher and FSCN/Tmm² lower in group E than in group C. Conclusion: A specific modulation of Sertoli cell function in the course of methimazole-induced hypothyroidism leads to a simultaneous concentration increase in inhibin A and decrease in B. Inhibin A might share responsibility for pituitary-testicular axis dysfunction and hypogonadotropic hypogonadism in this model of hypothyroidism.

Introduction: In the present study apoptosis was investigated in the cornu ammonis and cerebellum of 10 dogs naturally infected with rabies virus. Diagnosis of rabies was based on the results of fluorescent antibody staining and experimental inoculation.Material and Methods: The paraffin tissue sections were stained with haematoxylin and eosin, avidin-biotin complex peroxidase (ABC-P), and terminal deoxynucleotidyl transferase biotin-dUTP nick end-labelling (TUNEL) methods.Results: Histopathological examination revealed encephalomyelitis of varying severity and the presence of Negri bodies. Dense rabies antigens were determined in the motor neurons with ABC-P method. On the other hand, Bcl-2 protein and Bax protein gave positive reaction in seven and five cases, respectively. TUNEL staining demonstrated very marked apoptotic changes in the nuclei of neurons localised deep in the substantia alba of the cerebellum. Similar changes were also determined in perivascular mononuclear cells and glia cells within the substantia alba. No apoptopic changes were found in the motor neurons of the cornu ammonis.Conclusion: The absence of apoptotic changes in the neurons was considered to be the consequence of the necrotic changes that developed in these neurons.

Introduction: Due to the high heterogeneity of canine lymphoma, the aim of the present study was to test in vitro the chemosensitivity of canine high-grade primary lymphoma cells to various cytostatic drugs commonly used to treat dogs: 4-HO-cyclophosphamide, doxorubicin, dexamethasone, prednisolone, vincristine, etoposide, chlorambucil, lomustine, and cytosine arabinoside. Material and Methods: To determine the cell viability and drug ability to induce apoptosis two different tests were used: an MTT assay and annexin V/propidium iodide staining. Results: Both in vitro tests were found to be useful tools. Significant differences in the sensitivity, depending on the drug type, between B-, T- and mixed/null-type lymphoma cells were found for the majority of the tested drugs. B-type cells were the most sensitive in vitro, whereas T-type cells seemed to be the most resistant. Doxorubicin, chlorambucil, etoposide, and vincristine most strongly reduced the cell viability and induced apoptosis. Conclusion: In vitro assays, such as the MTT test and especially the annexin V/PI assay, may be useful tools for predicting a response to the treatment of high-grade lymphoma in dogs or improving the treatment outcomes in individual animals.
