Lateral cecal arterial blood flow, carotid arterial pressure, heart rate, and mechanical activity of the circular and longitudinal muscle layers of the cecal body were measured in 7 conscious healthy horses during IV infusion of physiologic saline solution for 60 minutes (control), during a 60-minute IV infusion of dopamine (at dosages of 1, 2.5, and 5 microgram/kg/min), and for 60 minutes after IV infusion of dopamine. The mean values for lateral cecal arterial blood flow during IV infusion of dopamine at a dosage of either 1 or 2.5 microgram/kg/min were not significantly different from the mean values for lateral cecal arterial blood flow during IV infusion of saline solution. The mean values for lateral cecal arterial blood flow, however, were significantly greater during IV infusion of dopamine at a dosage of 5 microgram/kg/min than the mean values for lateral cecal arterial blood flow during IV infusion of saline solution. Intravenous infusion of dopamine at 1 and 2.5 microgram/kg/min did not significantly change the mean values for carotid arterial pressure. In contrast, the mean values for carotid arterial pressure were significantly less during IV infusion of dopamine at dosages of 2.5 and 5 microgram/kg/min than during infusion of saline solution. The mean values for heart rate were not significantly altered by infusion of dopamine at a dosage of either 1 or 2.5 microgram/kg/min, but infusion of dopamine at a dosage of 5 microgram/kg/min significantly increased heart rate. Intravenous infusion of dopamine at dosages of either 1 or 5 microgram/kg/min did not significantly change the mechanical activity of the circular muscle layer of the cecal body, as measured by the area under the strain gauge deflection curve. Conversely, the mechanical avtivity of the circular muscle layer of the cecal body was significantly reduced by IV infusion of dopamine at a dosage of 2.5 microgram/kg/min. This reduction of circular muscle mechanical activity by dopamine infusion was attributable to a significant decrease in the total duration of contractions. The mechanical activity of the longitudinal muscle layer was not significantly altered by infusion of dopamine at any dose. These results suggest that IV infusion of dopamine at a dosage of 5 microgram/kg/min increased lateral cecal arterial blood flow by either increasing cardiac output or dilating the lateral cecal artery, an effect most likely mediated by dopaminergic or beta-adrenergic receptors. In addition, dopamine had a biphasic effect on contractile activity of the equine cecum.

The effects of butorphanol tartrate on arterial pressure, jejunal blood flow, vascular resistance, oxygen extraction, and oxygen uptake were determined in 10 anesthetized ponies ventilated with a mixture of halothane and 100% oxygen, using isolated autoperfused jejunal segments. Physiologic saline solution or butorphanol tartrate (0.2 mg/kg of body weight) was administered as a single bolus into the left jugular vein. By 2 minutes, butorphanol decreased arterial blood pressure and intestinal blood flow, and increased intestinal oxygen extraction. However, intestinal vascular resistance and oxygen uptake were unaffected. Results of this study indicate that butorphanol tartrate induces a hypotension that secondarily decreases intestinal blood flow, but intestinal vascular resistance and metabolism are not adversely affected. We conclude that butorphanol tartrate does not compromise intestinal viability in halothane-anesthetized ponies and, therefore, may be a good analgesic choice for the equid destined for abdominal surgery.

Lateral cecal arterial blood flow, carotid arterial pressure, heart rate, and mechanical activity in the duodenum, right ventral colon, cecal body, and cecal apex were measured in 6 conscious healthy horses for 60 minutes during and for 120 minutes after IV infusion of 0.9% NaCl solution (control) or fenoldopam. There were no significant changes in these measurements during or after infusion of 0.9% NaCl (saline) solution. Fenoldopam, a selective dopamine- 1 receptor agonist, was administered in saline solution at dosages of 0.01, 0.05, and 0.1 ug/kg/min. Intravenous infusion of fenoldopam at 0.01 ug/kg/min significantly increased heart rate, but did not change average carotid arterial pressure or lateral cecal arterial blood flow. Intravenous infusion of fenoldopam at both 0.05 and 0.1 ug/kg/min significantly increased heart rate, significantly decreased average carotid arterial pressure, and significantly increased lateral cecal arterial blood flow. Intravenous infusion of fenoldopam at 0.01, 0.05 and 0.1 ug/kg/min did not significantly change the mechanical activity measured by the area under the strain gauge defection curve for the duodenum, right ventral colon, cecal body, or cecal apex. These results suggest that dopaminergic-1 receptors are present on the colonic vascudopaminergic-1 receptors exist on the visceral smooth muscle of the duodenum, right ventral colon, cecal body, or cecal apex of horses.

To investigate the potential role of sympathetic nerves in preventing pronounced increases in cerebral blood flow, we evaluated the effects of abrupt hypertension on the cerebral circulation of newborn pigs with intact cerebral sympathetic innervation and after cerebral sympathetic denervation. Epinephrine infusion was used to induce abrupt increases in mean (+/- SEM) arterial pressure (innervated pigs, 62 +/- 3 mm of Hg to 115 +/- 3 mm of Hg; denervated pigs, 71 +/- 4 mm of Hg to 132 +/- 4 mm of Hg) that remained increased for the 3 minutes of the study. Abrupt hypertension increased blood flow to all brain regions. In denervated pigs, the increased flow to the cerebrum was prolonged, compared with that in pigs with intact sympathetic innervation. Differences between pigs of the innervated and denervated groups were not apparent, with respect to blood flow to any other region (caudate region, brain stem, cerebellum). In newborn pigs, sympathetic nerves may attenuate hypertension-induced increases in blood flow to the cerebrum, but do not appear to affect flow to the rest of the brain.

Pulsed-wave Doppler echocardiography was performed on 30 clinically normal 1- to 6-year-old racing Standardbreds. There were 13 females, 13 geldings, and 4 stallions. Cardiac disease was not detected with M-mode, 2-dimensional real-time or pulsed-wave Doppler echocardiography. Normal flow velocities for right and left atrial outflow, right and left ventricular outflow, the aorta, and pulmonary artery were determined. Peak flow velocities for right and left atrial outflow occurred during the rapid filling phase and were higher toward the mitral valve (mean, 0.70 +/- 0.24 m/s) than toward the tricuspid valve (mean, 0.49 +/- 0.17 m/s). Peak flow velocities in the right and left ventricular outflow tracts were similar (means, 0.81 +/- 0.10 m/s and 0.75 +/- 0.39 m/s, respectively). Peak flow velocities in the pulmonary artery (mean, 1.09 +/- 0.42 m/s) and aorta (mean, 1.01 +/- 0.29 m/s) were similar, although flow peaked earlier in systole in the aorta than in the pulmonary artery.

The blood supply patterns to the spinal cord were examined and compared in 15 dogs and 10 cats by use of dissection and radiographic visualization. The lowest percentages of radicular contributions and the smallest diameter vessels were found in the thoracic part of the spinal cord. The central arteries were fewest in number in the thoracic region and unilaterally or bilaterally supplied the gray matter. The percentage of bilaterally distributed central arteries increased from the cervical to the lumbar regions. The anastomotic plexus on the surface of the spinal cord was found to be most dense in the cervical and lumbar regions.

The in vitro reactivity of vasoconstrictive mediators that are implicated in acute laminitis was determined in palmar and plantar digital arteries and veins obtained from healthy horses and in palmar digital vessels of horses with early laminitis (Obel grade I). To obtain baseline reactivity data, 3 experiments were conducted, using healthy horses: (1) the reactivity of palmar and plantar digital arteries and veins to angiotensin II, norephinephrine, and 5-hydroxytryptamine (serotonin) were compared; (2) the direct effects of bacterial endotoxin on vascular reactivity were assessed; and (3) the reactivity of palmar digital arteries and veins to angiotensin II, norepinephrine, prostaglandin F2 alpha (PGF2 alpha), sertonin, and a thromboxane-endoperoxide analog (U46619) were determined. The vascular reactivity of these same 5 vasoconstrictors then was determined in horses with early laminitis and was compared with data from healthy (control) horses. Obel grade-I laminitis was experimentally induced in horses using carbohydrate overload. Dose responses were conducted for each agent at concentrations between 10(-8)M and 10(-4)M. The potency of a drug was defined as the mean effective concentration necessary to induce 50% of maximal contraction (EC50). There were no differences in EC50 concentrations and in maximal contractions between forelimb and hind limb arteries and veins for angiotensin II, norepinephrine, and serotonin. Incubation with endotoxin had no effect on the reactivity of arteries and veins to angiotension II, norepinephrine, and serotonin. In healthy horses, serotonin and U46619 were more potent arterial constrictors than were norepinephrine PGF2 alpha, and angiotensin II. In veins, serotonin, U46619, and angiotension II were similar in potency, and all were significantly (P less than 0.05) more potent than were norepinephrine and PGF2 alpha. Serotonin induced greater arterial constriction than did all other agents tested. There were no differences in the maximal venoconstriction induced by norepinephrine, PGF2 alpha, serotonin, and U46619. Angiotensin II induced the least amount of arterial and venous constriction. Maximal contractions were significantly (P less than 0.05) greater for veins than for arteries for all agents evaluated, except for angiotensin II. In horses with early laminitis, angiotensin II and serotonin were the most potent (smallest EC50 values) constricting agents for the arterial and venous segments and norepinephrine and PGF2 alpha were the least potent. Serotonin and norepinephrine induced significantly (P less than 0.05) greater venoconstriction than did the other agents. Angiotensin II induced the least arterial and venous contraction. For all agonists except angiotensin II, the mean EC50 values for vessels from horses with early laminitis were either similar or greater than those for vessels from control horses. The EC50 values for norepinephrine and the thromboxane analog were significantly (P less than 0.05) greater for vessels from horses with early laminitis, compared with those from control horses. The mean maximal contractions for all vasoconstrictors, except angiotensin II, were significantly (P less than 0.05) less for vessels from horses with early laminitis. Significantly greater venous-to-arterial maximal contraction ratios were found for norepinephrine and serotonin in horses with laminitis, compared with those ratios in control horses. These data suggested that the digital vasculature of horses with early laminitis was not more sensitive to the vasoconstrictor substances tested and that the vessels were significantly less repsonsive than was vasculature from the control horses. However, the venous-to-arterial contraction ratios were either the same or significantly (P less than 0.05) greater in horses with laminitis.
