Lumpy skin disease virus (LSDV) was isolated, from naturally infected cattle that have a history of previous vaccination with live attenuated sheep pox virus (SPV) vaccine. The virus was isolated on chorio-allantoic membrane (CAM) of specific pathogen free (SPF) embryonated chicken eggs (ECE) and identified by agar gel precipitation test (AGPT) and neutralization test using specific hyperimmune serum against LSDV and SPV. Characteristic intracytoplasmic inclusion bodies was detected in trypsenized cell of infected CAM stained with H&E. Laboratory studies for characterization of isolated LSDV revealed that it was stable at a wide range of pH, but it was inactivated by exposure to 56 0C for 15 minutes. Treatment of isolated LSDV with lipid solvents (20% ethyle ether and chloroform) reduced the virus titer 3.2 and 4.4 log respectively after 24 hrs at 4 0C .On cross neutralization testcomplete neutralization of isolated LSDV was obtained with both reference LSDV and SPV antisera. Cattle vaccinated with live attenuated SPV vaccine under experimental condition found to be protected against natural field infection with LSDV.

A total number of 80 nasal swabs collected from apparently normal cattle slaughtered in Basateen abattoir were screened for the presence of infectious bovine rhinotracheitis virus. Among 80 examined samples, 4 samples found positive after the 3rd passage on MDBK cell line with appearance of the specific cytopathic effect (grape like clusters). The isolated virus titers were 103.9, 104.2, 105, 105.6 TCID50 / 0.1 ml. The four positive isolates were identified by agar gel precipitation test (AGPT), virus neutralization test (VNT) and gave the intracytoplasmic granules by indirect fluorescent antibody technique (IFAT). Electrophoretic profile of IBR in sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) was described and visualized by Coomassie blue stain. The mobilities of electrophoretic bands were determined with molecular weight marker at approximate range from 206.39 to 22.14 kDa.

Thirty four apparently healthy cattle (9 males and 25 females) of mixed breed (Balady X Friesen) were selected for ultrasonographic investigations in this study. Ultrasonographic measurements of vertical and horizontal diameters of kidney, the diameters of the renal parenchyma and the diameter of renal sinus were determined in the middle of right and left kidneys. Twenty nine cattle (9 males and 20 females), had normal ultrasonographic appearance of both right and left kidney while the other five cows had some pathological affections including cystic kidney and renal dysplasia in their left kidneys . The vertical diameter of the right kidney was (4.84±1.18 cm) , the horizontal diameter (9.16 ± 1.35 cm) , and the vertical diameter of the renal sinus was ( 3.54 ± 1.02 cm ) . The thickness of the renal cortex and medulla ( renal parenchyma) was (2.16 ± 0.46 cm ). On the other hand ,the vertical diameter of the left kidney was (5.89 ±1.13 cm), and the vertical diameter of the renal sinus was ( 3.83 ± 1.12 cm ). The thickness of the renal cortex and medulla (renal parenchyma) was (2.46 ± 0.35 cm ). It was concluded that the ultrasonographic values determined in this study can be used as references for the diagnosis of morphologic changes in the kidney of domestic dairy cattle

Objective--To evaluate antimicrobial activity of bovine bactericidal permeability-increasing protein (bBPI)-derived synthetic peptides against mastitis-causing gram-negative bacteria. Sample Population--Bacterial isolates from the milk of cows with clinical mastitis. Procedures--3 peptides were synthesized with sequences corresponding to amino acids 65 to 99 (bBPI6599) or 142 to 169 (bBPI142169) or the combination of amino acids 90 to 99 and 148 to 161 (bBPI9099,148161) of bBPI. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of these peptides against bacterial isolates from cows with mastitis were determined by use of a standardized broth microdilution assay. The ability of these peptides to retain their antimicrobial activity in serum and milk was also evaluated. Finally, bacterial lipopolysaccharide (LPS)-neutralizing activity of these peptides was assayed with the Limulus amebocyte lysate test. Results--Of the 3 peptides tested, bBPI9099,148161 had the widest spectrum of antimicrobial activity, with MIC and MBC values ranging from 16 to 64 Mg/mL against Escherichia coli, Klebsiella pneumoniae, and Enterobacter spp and from 64 to 128 Mg/mL against Pseudomonas aeruginosa. None of the peptides had any growth-inhibitory effect on Serratia marcescens. The antimicrobial activity of bBPI9099,148161 was inhibited in milk, but preserved in serum. Finally, bBPI142169 and bBPI9099,148161 completely neutralized LPS. Conclusions and Clinical Relevance--bBPI9099,148161 is a potent neutralizer of the highly proinflammatory molecule bacterial LPS and has antimicrobial activity against a variety of gram-negative bacteria. The ability of bBPI9099,148161 to retain antimicrobial activity in serum suggests a potential therapeutic application for this peptide in the management of gram-negative septicemia.

This article summarizes the different types of free radicals, antioxidants and the effect of oxidative stress on the activities of glutathione peroxidase and glucose 6-phosphate dehydrogenase in bovine liver diseases. A growing body of evidence suggests that the formation of reactive oxygen species is a common occurrence associated with most if not all disease processes. The overall importance of reactive oxygen species to the progression and severity of various disease states varies greatly depending on the conditions and whether the disease is acute or chronic. Free radical researches in animals are in progress and further investigations are needed to establish the involvement of reactive oxygen species in diseases affecting different animal species and the pathology they produce.

We examined the effect of stroma-derived factors, hepatocyte growth factor (HGF) and leptin, on morphological differentiation of bovine mammary epithelial cells (BMEC) in collagen gel three-dimensional culture in vitro. BMEC treated with HGF, but not leptin, formed duct-like organoids. The formation of organoids by HGF was enhanced by treatment with a mixture of insulin, cortisol and prolactin, while BMEC treated with the mixture alone did not produce the organoid. In contrast, the formation of organoids by HGF was dose-dependently inhibited by simultaneous addition of leptin, regardless of the presence or absence of the hormone mixture. These results suggest that stroma-derived factors intricately regulate mammary epithelial morphogenesis.

An R664X nonsense mutant AE1 is responsible for dominant hereditary spherocytosis in cattle and is degraded by the proteasomal endoplasmic reticulum-associated degradation. The present study demonstrated that R664X AE1 translated in vitro had the trypsin-sensitve site identical to that of the wild-type AE1. The P661S/R664X mutant containing a possible N-glycosylation site at Asnsup(660) showed an increase in size by 3 kDa both in the cell-free translation system and in transfected HEK293 cells. Moreover, steady state levels of R664X and P661S/R664X in HEK293 cells were markedly increased in the presence of a proteasome inhibitior. These findings indicate that the truncated C-terminal region of R664X AE1 has lumenal localization in the endoplasmic reticulum and is not accessible to proteasomal machineries in the cytosol.
