Morphology and molecular study of Fascioloides magna – a growing threat to cervids (Cervidae) in Poland

Introduction: The giant liver fluke, Fascioloides magna, has spread across Europe over the years posing a serious threat to the Polish cervid population. Material and Methods: Macroscopic and histopathological studies of the liver of 22 roe deer (Capreolus capreolus), 10 red deer (Cervus elaphus), and 6 fallow deer (Dama dama) were performed. Species determination of the recovered liver flukes and eggs was performed by PCR protocol amplifying fragments of ribosomal DNA (ITS2), according to a standard method. Results: The presence of F. magna was confirmed in three (13.6%) roe deer, seven (70.0%) red deer, and two (33.3%) fallow deer. The fluke eggs were found only in the stools of five red deer and one fallow deer. Conclusion: This study presents detailed pathological and histopathological changes in the liver of wild Polish cervids, including roe deer, which were subjected to such study for the first time. The hepatic lesions typical for different stages of liver cirrhosis varied depending on the host species and stage of the disease.

The classification of the bone pieces excavated from Gasan-ri archaeological site Ⅰ in Jinju, presumed to be relics was investigated macroscopically. The remains of the animal bone were 3 classes (Mammalia, Aves, Amphibia), 5 orders (Carnivora, Artiodactyla, Passeriformes, Rodentia, Salientia), and 6 species (Sus scrofa, Cervidae sp., Nyctereutes procyonides, Passeriformes sp., Rattus norvegicus caraco, Rana nigromaculata). The total weight of the animal bone remains was 1,002.80 g, with the identified bones comprising 975.30 g and an identification rate of 97.26%. A total of 447 animal bone fragments were identified, including 204 bone pieces of S. scrofa (468.00 g, 47.99%), 102 bone pieces of Cervidae sp. (453.79 g, 46.53%), 68 bone pieces of R. nigromaculata (4.69 g, 0.48%), 59 bone pieces of N. procyonides (47.14 g, 4.83%), 9 bone pieces of Passeriformes sp. (0.98g, 0.10%), and 5 bone pieces of Rattus norvegicus caraco (0.70 g, 0.07%). The bone pieces of the animal relics consisted of 81 skull (18.12%), 161 axial skeleton (36.02%), 64 forelimb (14.32%), and 141 hindlimb (31.54%) fragments. The archaeological significance of the animal bones excavated in this investigation was that wild boars and deer were presumed to have been mainly used animals in the Gasan-ri area of Jinju during the Three Kingdoms period.

OBJECTIVE To characterize antimicrobial prescribing patterns of clinicians and clinical services at a large animal veterinary teaching hospital and identify factors associated with antimicrobial prescribing. ANIMALS All large animals (ie, equids, bovids, sheep, goats, camelids, swine, and cervids) evaluated at the New Bolton Center hospital at the University of Pennsylvania from 2013 through 2018. PROCEDURES In a cross-sectional study design, data on antimicrobial use by clinicians and clinical services were collected from administrative and billing records. Multivariable regression modeling was performed to identify factors associated with antimicrobial prescribing patterns. RESULTS Antimicrobials and critically important antimicrobials of the highest priority were dispensed in 42.1% (9,853/23,428) and 24.0% (2,360/9,853) of visits, respectively, and these proportions differed significantly among clinicians. Per visit, the median (interquartile [25th to 75th percentile] range) number of animal-defined daily doses dispensed was 3.6 (0.8 to 11.1) and the mean (SD) number of antimicrobial classes dispensed was 2.0 (1.3). Patient species, age, affected body system, and duration of hospitalization as well as submission of specimens for bacterial culture were significantly associated with prescribing patterns. CONCLUSIONS AND CLINICAL RELEVANCE The frequency and quantity of antimicrobials prescribed differed significantly among clinicians within and across services, even for animals with clinical signs affecting the same body system. Patient- and visit-level factors explained some but not all of the heterogeneity in prescribing patterns, suggesting that other clinician-specific factors drove such practices. More research is needed to better understand antimicrobial prescribing patterns of clinicians, particularly in situations for which no antimicrobial use guidelines have been established.

Information on steroid hormone receptor distribution in the uterus is essential to understand the roles of their ligands in pregnancy. This study examined the spatio-temporal localization of estrogen receptor alpha (ERalpha) and progesterone receptor (PR) in the uterus of sika deer (Cervus nippon) to determine the estrogen and progesterone action site during pregnancy. Ovaries and uteri were collected from 21 pregnant sika deer with single fetus and two corpora lutea, ranging from Day 20 to Day 207 of pregnancy. In addition, genital organs were also collected from three sika deer whose gestational status was unknown: one female had only one developing corpus luteum: =Day 4 (metestrus) and two females had two corpora lutea, one of which was at the developing stage equivalent to diestrus or early pregnancy: Day 7 (diestrus). Staining of ERalpha and PR was clear in all cell types during metestrus. During diestrus, the presence of ERalpha was also clear in deep glandular epithelium, stroma and myometrium, whereas it was suppressed in luminal epithelium and shallow glandular epithelium. Staining of PR was suppressed in luminal epithelium but was detectable in other cell types. Staining of ERalpha in all cell types and PR in luminal epithelium and glandular epithelium became undetectable by Day 28. PR was presented in stroma and myometrium throughout pregnancy. The distribution pattern of ERalpha and PR was different during diestrus from that in a ruminant. This could be attributed to estrogen secretion from the maturing and ovulating follicles in the presence of developed corpus luteum.

From 1985 to 1996, 42 deers (seven Mazama americana, 16 M. gouazoubira, 13 Ozotoceros bezoarticus and six Blastocerus dichotomus) were submitted to necropsy. From those animals, 14,426 Trichostrongyloidea nematodes were gathered, being 13,281 (92.05%) parasites of abomasum and 1,145 (7.95%) parasites of small intestine. Haemonchus contortus, H. similis, Trichostrongylus axei, T. colubriformis, Cooperia punctata and C. pectinata were identified in those organs. All of the animals had helminthic infections by one or more worm species, occurring a wide variation in the intensity of infection (from one to 4,345 nematodes). The results showed low intensity of infection values, less than 100 worms, in 25 (59.52%) of the animals. The higher results of mean intensity of infection were observed in M. gouazoubira (596.37) and in O. bezoarticus (331), and the lower results, in M. americana (17.57) and B. dichotomus (75.5). The most expressive values of intensity of infection, abundance and prevalence were observed for Haemonchus spp (fourth stage larvae - L4), H. contortus, H. similis and T. axei. Genus Haemonchus was verified in 35 animals, and, therefore, with a prevalence of 83.33%; the total intensity of infection of this genus was 11,616 specimens, representing 80.52% of the Trichostrongyloidea nematodes verified, being that the majority (8,903) of those worms were constituted of young forms. Otherwise, H. similis was the specie predominating in the infections and, therefore, the one that showed the highest value of abundance. Genus Trichostrongylus was verified in 24 (57.14%) animals, and the total intensity of infection was 2,444 specimens, being 1,655 specimens of T. axei, that represented 11.54% of the parasitic burden observed. The six worm species observed in deers are also common to domestic ruminants in the states of São Paulo and Mato Grosso do Sul, and, in this way, none species of Trichostrongyloidea exclusive to deers was observed. | No período compreendido entre 1985 e 1996 foram necropsiados, para pesquisa de helmintos, 42 cervídeos, sendo sete Mazama americana, 16 M. gouazoubira, 13 Ozotoceros bezoarticus e seis Blastocerus dichotomus. Desses animais, foram colhidos 14.426 nematódeos Trichostrongyloidea, sendo 13.281 (92,06%) parasitos de abomaso e 1.145 (7,94%), de intestino delgado. Nesses órgãos, foram identificadas seis espécies de nematódeos: Haemonchus contortus, H. similis, Trichostrongylus axei, T. colubriformis, Cooperia punctata e C. pectinata. Todos os animais apresentaram infecções helmínticas por uma ou mais espécies, ocorrendo grande variação na intensidade de infecção (1 a 4.345 nematódeos). Ainda com relação à intensidade de infecção, os dados expressavam valores menores que 100 parasitos em 25 (59,52%) animais. Os valores mais altos de intensidade média das infecções foram observados em M. gouazoubira (596,37 helmintos) e em O. bezoarticus (331), e os menores, em M. americana (17,57) e B. dichotomus (75,5). Os dados mais expressivos de intensidade de infecção, abundância e prevalência foram observados para Haemonchus (larvas de 4º estágio), H. contortus, H. similis e T. axei. O gênero Haemonchus foi constatado em 35 animais, com prevalência de 83,33%; apresentou carga parasitária de 11.616 exemplares, representando 80,52% dos nematódeos verificados, sendo a maioria (8.903) constituída por formas imaturas. Por outro lado, H. similis foi a espécie predominante nas infecções e, portanto, a que apresentou maiores valores de abundância. Verificou-se o gênero Trichostrongylus em 24 (57,14%) animais, com carga parasitária de 2.444 exemplares, sendo 1.665 espécimes de T. axei, que representou 11,54% da carga parasitária obtida. As seis espécies de vermes identificadas nos cervídeos são comuns aos ruminantes domésticos nos Estados de São Paulo e Mato Grosso do Sul e dessa maneira não se observou nenhuma espécie de Trichostrongyloidea exclusiva dos cervídeos.
