Pure separation of various leukocytes is required for the assessment of their roles in immunological and physiological function. In this study, pure separation of monocytes from canine peripheral blood was attempted. At first, mononuclear cells (PBMC) were separated by ficoll-hypaque gradient method and then monocytes were recovered from PBMC suspensions in sucrose gradient Sol. (PBMC-Sucrose), autologous plasma (PBMC-Plasma) and autologous serum (PBMC-Serum) incubated at 37deg C for 2 hours. In the separation of PBMC by ficoll-hypaque gradient method in canine blood, higher relative centrifugal force (RCF) was required, as high as more than 1,300xg RCF for 40 minutes, for clear formation of PBMC layer than that in human blood as usually used 400xg RCF for 40 minutes. In monocytes-separation from three PBMC suspensions following PBMC separation, recovery-, purity- and viability- rate of monocytes showed better results in PBMC-Plasma and PBMC-Serum than in PBMC-Sucrose suspension, particularly showing better results from PBMC suspensions performed by centrifugation at 1,500xg RCF for 40 minutes

This investigation was conducted on 249 Jindo dogs aging from 2 to 12 months to determine body type and external appearance of the dogs. The withers height of male dogs growed rapidly during the period between 2 months and 7 months of age, and was 22.94cm and 49.77cm, respectively. Afterward the growth rate slowed down up to 12 months, 53.33cm. The growth rate of female was similar to male, and the withers height was 22.91cm, 46.81cm, 48.70cm at 2, 6 and 12 months of age. For male and female the body length was 27.21, 27.23cm at 2 months, 51.40cm, 51.62cm at 6 months, 58.33, 52.15cm at 12 months, respectively. Growth rate of the body length was very rapid from 2 to 6 months of age, but afterwards the growth rate was slow and somewhat retarded. For male and female, the withers height to body length ratio was 100 : 109.4 and 100 : 107.1 in 12 months old group. Cross over between the sexes in growth rate occurred between 5.3 to 6.5 months of age for wither height, between 6.0 to 7.2 months for body length, respectively. The average chest girths of male and female were 31.46, 30.46cm at 2 months, 54.92, 52.20cm at 7 months, 60.25, 57.90cm at 12 months, respectively. The growth rate of chest girth was rapid between 2 to 7 months of age and gradual between 7 to 12 months. The average skull lengths of male and female were 11.76, 11.32cm at 2 months, 21.83, 19.60cm at 12 months, respectively. The average head widths of male and female were 8.37, 7.94cm at 2 months, 15.47, 12.46cm at 12 months. Erected type of ear was completed from oblique type at 6 months in male and 7 months in female. On our detailed examinations, we concluded that Jindo dogs completed their growth at 6 to 7 months of age

Nine strains of Yersinia species isolated from pigs and dogs in Korea, comprising 5 strains of Yersinia enterocolitica, 2 strains of Y. kristensenii and each strain of Y. pseudo-tuberculosis and Y. intermedia, were examined for the presence of virulence-associated plasmids, calcium dependency and provocation of guinea pig conjunctivitis (Sereny test). Three strains of Y. enterocolitica isolated from pigs were positive in calcium dependency and harbored one plasmid of about 45 megadalton, but negative in Sereny test

The aim of the present study was to test the efficiency of estrous induction in the premature, metestrous and anestrous bitches. The estrus was induced with prostaglandin F2 alpha, estradiol-17 beta, pregnant mare serum gonadotropin (PMSG) and human chorionic gonadotropin (HCG) in the treatment A, and with PMSG and HCG in the treatment B. Day 0 was the first day of estrone injection in the treatment A and the day of PMSG injection in the treatment B. Twenty three of the twenty six bitches laparotomized under general anesthesia between 11 and 18 days after onset of behavioral estrus, whereas three bitches were not laparotomized and remained until parturition. Ovarian responses were evaluated with the total number of corpora lutea or ovulation sites. The uterine horns were flushed with phosphate-buffered saline added heat treated canine serum (10 %), the flushing media was collected into watch glass and the ova were examined under stereomicroscope. Standing estrus was observed on the day 17.7 +- 1.5 after injection of estrone in the treatment A, but ovarian responses were not detectable. Standing estrus was observed on the day 12.2 +- 0.2 after injection of PMSG in the treatment B and 14 of 17 bitches showed ovarian responses. Ova were recovered in 9 of the 14 bitches. Ovarian responses were observed in one of the three premature bitches, two of the three metestrous bitches and all of the 11 anestrous bitches. The average number of the ova collected from 9 bitchs were 12.2 +- 1.4. Three bitches in the treatment B exhibited behavioural estrus and all of them were mated with fertile male dog, resulting the pregnancy in only one bitch. The pregnant bitch gave the birth of two pups

To elucidate the physiological characteristics of histamine receptor of the isolated renal artery in dog, effects of various receptor blockers and Ca++ channel blockers on the contractile responses of histamine were investigated in the isolated renal artery of dog. Histamine caused the contraction in the isolated renal artery of dog, and the contractile responses increased between the concentration of 5 x 10** (-6)M and 10** (-2)M in dose-dependent manner. The contractile response induced by histamine (10** (-3)M) was not blocked by pretreatment with atropine (10** (-6)M), phentolamine (10** (-6)M), propranolol (10** (-6)M) or cimetidine (10** (-6)M), but was completely blocked by pretreatment with pyrilamine (10** (-6)M). The contractility induced by histamine (10** (-3)M) was decreased in the Ca++ free medium. The contractile response induced by histamine (10** (-3)M) was decreased in the Ca++ free medium. The contractility induced by histamine (10** (-3)M) was markedly inhibited by pretreatment with papaverine (5x10** (-5)M) or verapamil (5x10** (-5)M). After the pre-contraction induced by norepinephrine (10** (-7)M) or prostaglandin F2 alpha (5x10** (-6)M), the contractile response induced by the additional treatment of histamine (10** (-3)M) was markedly increased compared with that of histamine (10** (-3)M) alone

Hematologic values were determined in blood samples obtained from 203 normal, healthy Jindo dogs of both sexes that ranged in age from 3 months to 10 years. Age-related differences were detected for the total erythrocyte count, hemoglobin content, PCV and the number of eosinophils. The persistent eosinophilia appears to be due to the dirofilariasis and flea-allergy dermatitis

The study was undertaken to obtain indirect values of volume of packed red cell (VPRC) without centrifugation, using the erythrocyte sedimentation rate (ESR) of diluted blood in dogs. ESRs of diluted blood using the diluent of autologous plasma in which formed numerous RBC-rouleau clumps, autologous serum in which formed a few RBC-rouleau clumps, and 5 % dextrose or 6 % sucrose solutions in which formed numerous RBC-aggregation clumps, were accelerated. But ESR of diluted blood using the 0.9 % saline, D-S, ACD-B, CDP or D-PBS solutions were sluggish, because erythrocytes were dispersed in these diluents. Reliable values of VPRC on the basis of the correlating regressive equation to the ESR could be derived from values of 60deg C-angled-micro-ESR/40 min in the mixture, four parts of 5 % dextrose solution and one part of whole blood. In the ESR values of diluted blood with low ratio, 1 : 1-3 : 1, 60deg C-micro-ESR was higher than 60deg C-Wintrobe-ESR. But, in the diluted blood with high ratio, 4 : 1-5 : 1, there was no different ESR values. For an aid of practical use, authors suggested a list of the 60deg C-micro-ESR/40 min in the diluted blood with equivalent VPRC of whole blood

Five hundred and forty-eight samples of pig heart muscle were collected from the abattoirs of many regions in Korea to reveal the frequency of Sarcocystis infections and to identify the species from June 1988 to April 1989. Heart muscle of the pigs was inspected for sarcocysts by the direct detection technique and for bradyzoites by the trypsin digestion technique. For examination of development of the parasites in the final host, 5 cross bred mature dogs, 5 puppies and 5 kittens were fed 100g, 50g and 50g of the infected meat respectively, four times in 2 days. Of 402 fattened and 146 older culled breeding pigs, 3 fattened pigs and 39 culled pigs were positive for Sarcocystis. Sarcocystis cysts from heart muscle measured an average of 425 x 169 micro m and bradyzoites an average of 15.6 x 3.5 micro m. Of 15 animals, only 2 puppies were infected with Sarcocystis. The prepatent period was 11 to 12 days and patent period was not examined since the puppies were infected with some another infections and one died on day 11 and another died on day 12 after ingestion of the meat. The sporulated oocysts were detected 11 days after ingestion of the meat and sporocysts 12 days from the puppy feces. The sporulated oocysts measured an average of 16.5 x 11.5 micro m and sporocysts an average of 12.6 x 7.9 micro m. On scaping examination of the intestinal mucosa, fully sporulated oocysts were detected in the tip of the intestinal villi. Considering above all descriptions, Sarcocystis in pig heart muscle in Korea was identified with Sarcocystis suicanis
