Preliminary study on classification of raw bovine’s milk using ATR-FTIR coupled with PCA from Peninsular Malaysia

A classification of raw bovine’s milk samples according to theirgeographical origin in Peninsular Malaysia by Makmal Kesihatan Awam Veterinar, Department of Veterinary Services Malaysia (DVS). Six hundred bovine milk samples were collected from Perlis, Kedah, Perak, Selangor, Pahang, Negeri Sembilan, Melaka and Johor states by 26 milkcollecting centres under DVS. This study was carried out directly using attenuated total reflectance Fourier transform infrared(ATR-FTIR) spectroscopy method coupled with a multivariate principal component analysis (PCA). The spectra generated by ATR-FTIR were analysed and regions of interest were found in between 3851.651cm-1 until 2700.819 cm-1 and 2419.173 cm-1 until 977.368 cm-1. The absorbance and wavenumber data of the regions were then analysed using PCA and the results show presence of clustering towards theirgeographical origin. ATR-FTIR coupled with multivariate PCA has potential for classifying the geographical origin of raw milk produced within Peninsular Malaysia. This method provides a rapid and nondestructive secondary methodology in milk classification without further sample preparation.

Identification of microorganisms, including bacteria, are widely used especially in environmental studies, biotechnology, clinical microbiology, microbial forensics, and in research study. The conventional method of bacteria identification is based on phenotypic observation techniques by profiling an organism’s metabolic attributes or some aspect of its chemical composition. Then, interpretation of test results involves substantial subjective judgement. Currently, general 16S rRNA sequencing and specific PCR play an important role in the accurate and faster identification of bacteria. The aim of this study is to compare the identification of the genus or species of bacteria from agricultural waste using conventional microbiology biochemical test and molecular techniques PCR 16S rRNA universal amplified ribosomal region (UARR) sequencing. A total of 72 agricultural waste samples and 2 ATCC culture as positive control were tested. Out of two ATCC bacteria and fifteen bacteria isolates identified by the biochemical test, twelve species (71%) of bacteria gave exactly the same bacteria genus as the 16S rRNA sequencing results. Aeromonas hydrophilia, Alcaligenes faecalis and Acinetobacter calcoaceticus was revealed as Pseudomonas sp. from the sequencing results. As for Alcaligenes sp., the results from the sequencing is Stenotrophomonas maltophilia. Previous reports also showed different results of the same isolate which were from similar classification, and closely related to each other. The limited number of biochemical tests available in a laboratory will contribute to misidentification of a proposed specie.

Recently, Fowl Adenovirus (FAdV) cases have been reported in many countries worldwide. FAdV is a contagious agent associated with inclusion body hepatitis (IBH) and hydropericardium syndrome (HPS) in chickens. It belongs to the Aviadenovirus genus of the family Adenoviridae. The virus is classified into five species (A to E) and further divided into 12 serotypes. Depending on the serotypes, they have diverse characteristics of virus that can either be pathogenic or nonpathogenic strain. From the viewpoint of epidemiological as well as vaccine development, it is very important to detect FAdV strains. Previous studies have been conducted on molecular research, but the continuity of this study in Malaysia has been limited. This study aims to identify the serotype classification of five Malaysian FAdV isolates obtained from field outbreaks during 2017-2019. In this study, polymerase chain reactions (PCR) were conducted based on Hexon gene. Results from the nucleotide sequence analysis discovered that the five isolates showed high similarity with FAdV-8b strains. High bootstrap values in phylogenetic analysis supported the clustering of the Malaysian FAdVs isolates into FAdVs species E. Consequently, the result of this study contributed important information on the epidemiology and culminated in the importance of control strategies against FAdV infection in Malaysia.
