Anthelmintic Resistance to Albendazole and Fenbendazole in Gastrointestinal Nematodes of Sheep in Saghez Municipality, Iran

BACKGROUND: During the last two decades, anthelmintic drugs have been increasingly applied against gastrointestinal parasites of sheep in Iran. OBJECTIVES: For this purpose, drug resistance to albendazole (Alb) and fenbendazole (Feb) in gastrointestinal nematodes in sheep of Saqez multiplicity was assessed. METHODS: In in-vivo experiment, a total number of 90 sheep in three groups (30 sheep/group) with EPG≤150 were examined for nematode resistance to Alb and Feb. They were treated with Alb and Feb or untreated (as a control group). RESULTS: There was significant difference between Alb and Feb treated groups and control group. The EPG in Alb, Feb and control groups was 59.8±1.93, 18.8±1.258 and 204.07±4.81, respectively. There was drug resistance against Alb in compassion with control group (R=71%). There was suspicion drug resistance for Feb in comparison with control group (R=90.66%). CONCLUSIONS: From the results of the present study, it was concluded that there was absolute and suspected drug resistance against Alb and Feb in sheep of Saqez municipality, Iran, respectively.

Dermatophytosis is a common skin disease in cats and dogs caused by Microsporum and Trichophyton fungi. Species identification and knowledge of their antifungal susceptibility are therapeutically and epidemiologically important. This study assessed the prevalence of feline and canine dermatophytosis in Iran, identified the aetiological agents molecularly and tested their antifungal susceptibility. A total of 308 companion animals (134 dogs and 174 cats) with skin lesions were examined from March 2015 to March 2018. Hair and skin samples were examined by microscopy with 20% KOH and cultured on Sabouraud dextrose agar with cycloheximide and chloramphenicol. Fungal isolates were confirmed by sequencing of the internal transcribed spacer (ITS) r-DNA region. The antifungal susceptibility of dermatophytes was tested by broth microdilution assay using standard drugs. Dermatophytes were found in 130 (42.2%) samples, 62 of them feline and 68 canine. Based on sequencing of all strains, M. canis (78.5%, P<0.05), M. gypseum (10.7%), and T. mentagrophytes (10.7%) were the dermatophytes isolated. The non-dermatophyte species Nannizziopsis vriesii was also isolated from two feline dermatomycosis cases. Dogs and cats younger than one year (61.5%) showed a statistically significantly higher prevalence of infection (P<0.05). Caspofungin produced the lowest geometric mean MIC at 0.0018 μg/mL, followed by ketoconazole, terbinafine, itraconazole, miconazole, griseofulvin, clotrimazole and fluconazole, in a 0.038–1.53 μg/mL range. This is the first molecular study to identify the causes of pet dermatophytosis in north-western Iran. ITS-PCR was shown to be a useful and reliable method for the identification of closely related species of dermatophytes in clinical and epidemiological settings. The lowest MIC of caspofungin indicated that this drug was the most potent in vitro.

Candida species are a natural component of the intestinal tract microflora, but in favourable conditions they can cause superficial, mucosal, or even systemic candidiasis. Poultry production might be a source of human drug-resistant yeast infections, including Candida spp. The limited data concerning the antifungal susceptibility of poultry Candida isolates prompted us to carry out research to determine the susceptibility of isolates from turkey intestinal tracts. The beak cavity, crop and cloaca were swabbed of 580 turkeys from 58 flocks in western Poland. The susceptibility tests were conducted using the E-test method with amphotericin B, fluconazole, itraconazole, and voriconazole on 52 isolates of C. albicans, C. catenulata, C. glabrata, C. palmioleophila, C. rugosa, C. krusei and C. lusitaniae. All isolates were susceptible to voriconazole. According to the MIC values obtained for amphotericin B and fluconazole, all Candida spp. isolates were classified as susceptible according to the described breakpoints except for C. krusei, which was the only isolate that was amphotericin B-, fluconazole- and itraconazole-resistant. The susceptibility to itraconazole varied: 11 of the Candida isolates were susceptible (21.1%), 29 were dose-dependently susceptible (55.8%), and 12 isolates were resistant (23.1%). There are few resistant strains of Candida in turkeys, and the drug resistance varies. When Candida passes from turkeys to humans, there is a wide range of antifungal treatment options.

Enteropathogenic Escherichia coli (EPEC) is one of the main pathotypes causing gastroenteritis, particularly in young immunocompromised hosts. The study reports the prevalence, characterisation, and molecular epidemiology of EPEC from piglets in northeastern India. A total of 457 faecal samples were collected, from which 1,286 E. coli strains were isolated and screened by PCR. The resultant EPEC strains were serotyped and phenotypically characterised for resistance against 15 antimicrobials. Also, the phylogenetic sequence was analysed for 11 selected strains. A total of 42 strains (3.26%) belonged to atypical EPEC, of which, 15 (35.71%, and 2.29% of the 654 strains from this farm type) were isolated from organised and 27 (64.29%, and 4.27% of the 632 strains from this farm type) from unorganised farms; further, 5 (11.90% of the EPEC strains and 1.51% of the 330 strains from this breed) were isolated from the indigenous breeds and 37 (88.10%, and 3.87% of the 956 strains from this breed) from crossbred piglets. Serogroups O111 (11.9%) and O118 (7.14%) were the most prevalent of the 10 present. Sequence analysis of a length of the eaeA gene of 11 isolates of the region showed them to have 100% homology with each other and their identity ranged from 99.4% to 99.7% with GenBank reference sequences. All the EPEC isolates were multi-drug resistant, showing the highest resistance to amoxicillin (80.9%) and cephalexin (76.19%). The study highlighted the association of EPEC with piglet’s diarrhoea in northeastern India. EPEC isolates belonged to many serotypes and phenotypically all were multi-drug resistant with close genetic homology.

Introduction: Tuberculosis is a highly infectious disease affecting humans and animals. It is caused by the Mycobacterium tuberculosis complex (MTBC) – Mycobacterium bovis and Mycobacterium caprae, which are aetiological factors of bovine tuberculosis (bTB). In Poland, the bTB eradication programme exists. Animals diagnosed with tuberculosis are in the majority of cases not treated, but removed from their herd and then sanitary slaughtered. Material and Methods: In total, 134 MTBC strains isolated from cattle in Poland were subjected to microbiological analysis. The resistance phenotype was tested for first-line antimycobacterial drugs used in tuberculosis treatment in humans: streptomycin, isoniazid, rifampicin, ethambutol, and pyrazinamide. The strains were isolated from tissues collected post mortem, so the test for drug resistance fulfilled only epidemiological criterion. Results: The analysis of drug-resistance of MTBC strains revealed that strains classified as M. bovis were susceptible to 4 antimycobacterial drugs: isoniazid, rifampicin, streptomycin, and ethambutol, and resistant to pyrazynamide. The strains classified as M. caprae were sensitive to all tested drugs. Conclusion: The results indicate that despite enormously dynamic changes in mycobacterial phenotype, Polish strains of MTBC isolated from cattle have not acquired environmental resistance. The strains classified as M. bovis are characterised by natural resistance to pyrazinamide, which is typical for this species.

This study aimed at investigation of the prevalence and antimicrobial susceptibility of Pseudomonas spp., in two marine water fish, namely Pagrus, and saurus intended for human consumption. In the current study, Pseudomonas spp., was isolated and identified from two marine water fishes. Fifty Pagrus fish and 50 Saurus (Saurida undosquamis) fish were sampled from fish markets at Sharkia governorate, Egypt. The prevalence rates of Pseudomonas spp. were 84%, and 40% in Pagrus fish, and Saurus respectively. The identified Pseudomonas spp., from Saurus was Pseudomonas aeruginosa (P. aeruginosa), P. fluorescens, P. fragi, P. cepacia, at 20% for each. P. alcaligenes and P. lundensis were recovered at 10%. In Pagrus, the prevalence rates were 31.3%, 56.3%, and 6.2% for P. aeruginosa, P. fluorescens, P. fragi and P. stutzeri. Seven serotypes of P. aeruginosa were identified with the serotypes O11 at the top prevalence (42.8%), and O6, O5, O1, O8 at 19.3% for each. The virulence-associated genes were lasB (Elastase B gene), exoS (Exoenzyme S) and pilB (pili gene) were detected in the recovered P. aeruginosa at 100%, 71.4%, and 28.6%, respectively. The recovered Pseudomonas species had high antimicrobial resistance to erythromycin, amoxicillin, ampicillin and gentamycin at 100%, 88.5%, 65.4%, and 50%, respectively. In conclusion, Pagrus and Saurus fish species are considered as potential sources of multidrug resistant Pseudomonas spp.

Staphylococcus aureus (S. aureus) is considered the most important cause of food borne intoxication, which occurs due to ingesting of food contaminated with enterotoxin of S. aureus. In this study, S. aureus from saurus, and pagrus fish species were isolated, then identified by morphological and biochemical examination. S. aureus coagulase, and D-Nase +ve were detected in 16 from 25 (64%), and 17 from 25(68%) of the examined samples of saurus, and pagrus, respectively. Total S. aureus counts were 5.14±0.06 in saurus, and 5.02±0.03 log 10 cfu/g in pagrus fish, respectively. Detection and typing of enterotoxin by Reverse Passive Latex Agglutination technique "RPLA" revealed the detection of staphylococcal enterotoxins (SEs) SEA, SEC, and SED at 18%, 6%, 0% in saurus and 5%, 0%, and 11%, at pagrus, respectively. S. aureus was tested for antimicrobial susceptibility. The recovered S. aureus coagulase, and D-Nase +ve in the current study showed resistance to kanamycin, clindamycin, nalidixic acid, and sulphamethoxazole at 100%, 87.9%, 84.8%, and 81.8%, respectively. The isolates showed sensitivity to amikacin, imipenem, meropenem, oxacillin at 90.9%, 87.9%, 84.8%, and 81.8% respectively. It could be concluded that multidrug resistant enterotoxigenic strains of S. aureus could be isolated from saurus and pagrus spp. Therefore, it is highly recommended to adopt strict hygienic measures and efficient cooking during before consumption of such fish species.

The present study aimed to investigate the prevalence of Salmonella and their antibiotic resistance pattern against two commonly used antibiotics: ciprofloxacin and ceftriaxone in marketed live chickens in a live birds market. Materials and methods: The study was conducted on live Sonali chickens (crossbred of Fayoumi female and Rhode Island Red male). Cloacal swabs were collected from 50 randomly selected live birds from 5 retail sellers (10 samples/seller) at Jhawtola live birds’ market, Chattogram, Bangladesh. Culture and biochemical tests were used to identify Salmonella. Positives samples for Salmonella were further tested for antibiogram by disc diffusion method.Salmonella was identified from 28 (56%) of samples by culture and biochemical test. Among the positive isolates, 27 (96.42%) samples exhibited resistant to ceftriaxone, and 20 (71.42%) samples were to ciprofloxacin.The result of this study indicates there is a high prevalence of Salmonella in marketed local chicken and are resistant to ceftriaxone and ciprofloxacin which has economic importance as well as a public health concern. Awareness should be increased to prevent random use of antibiotics and provision of strict biosecurity measures.

Objective-To evaluate various sampling strategies for potential use in measuring prevalence of antimicrobial susceptibility in cattle. Sample Population-500 isolates of non-type-specific Escherichia coli (NTSEC) isolated from the feces of 50 cows from 2 dairy farms (25 cows/farm and 10 isolates/cow). Procedures-Diameters of inhibition zones for 12 antimicrobials were analyzed to estimate variation among isolates, cows, and farms and then used to determine sampling distributions for a stochastic simulation model to evaluate 4 sampling strategies. These theoretic sampling strategies used a total of 100 isolates in 4 allocations (1 isolate from 100 cows, 2 isolates from 50 cows, 3 isolates from 33 cows, or 4 isolates from 25 cows). Results-Analysis of variance composition revealed that 74.2% of variation was attributable to isolates, 18.5% to cows, and 7.3% to farms. Analysis of results of simulations suggested that when most of the variance was attributable to differences among isolates within a cow, culturing 1 isolate from each of 100 cows underestimated overall prevalence, compared with results for culturing more isolates per cow from fewer cows. When variance was not primarily attributable to differences among isolates, all 4 sampling strategies yielded similar results. Conclusions and Clinical Relevance-It is not always possible to predict the hierarchical level at which clustering will have its greatest impact on observed susceptibility distributions. Results suggested that sampling strategies that use testing of 3 or 4 isolates/cow from a representative sample of all animals better characterize herd prevalence of antimicrobial resistance when impacted by clustering.
