Occurrence of enterococci in mastitic cow’s milk and their antimicrobial resistance

Introduction: The aim of the study was to evaluate the occurrence of enterococci in inflammatory secretions from mastitic bovine udders and to assess their antimicrobial resistance. Material and Methods: A total of 2,000 mastitic milk samples from cows were tested in 2014–2017. The isolation of enterococci was performed by precultivation in buffered peptone water, selective multiplication in a broth with sodium azide and cristal violet, and cultivation on Slanetz and Bartley agar. The identification of enterococci was carried out using Api rapid ID 32 strep kits. The antimicrobial susceptibility was evaluated using the MIC technique. Results: Enterococci were isolated from 426 samples (21.3%). Enterococcus faecalis was the predominant species (360 strains), followed by E. faecium (35 isolates), and small numbers of others. The highest level of resistance was observed to lincomycin, tetracycline, quinupristin/dalfopristin (Synercid), erythromycin, kanamycin, streptomycin, chloramphenicol, and tylosin. Single strains were resistant to vancomycin and ciprofloxacin. All isolates were sensitive to daptomycin. E. faecalis presented a higher level of resistance in comparison to E. faecium, except to nitrofurantoin. Conclusion: The results showed frequent occurrence of enterococci in mastitic cow’s milk and confirmed the high rate of their antimicrobial resistance.

In this study, it was aimed to investigate the presence and the prevalence of Enterococcus faecalis and Enterococcus faecium strains isolated from the urine and stool samples. A total of 500 routine urine and feces samples were used for testing as the study materials, and a total of 349 Enterococcus spp. were collected for investigation. For the isolation, blood agar and bile esculin agar were used. DNA isolations of the 24-hour growth cultures of possible enterococci were carried out using a DNA isolation kit.Out of 350 routine urine and 150 stool samples taken with the approval of the patients, 235 (67.1%) and 114 (76%) Enterococcus spp. were isolated respectively. Using the multiplex PCR method with species specific primers, 136 (57.8%) of urine and 22 (19.2%) of stool originated enterococcal strains were identified as Enterococcus faecalis; on the other hand, 17 (7.2%) of urine and 61 (53.5%) of stool originated enterococci were identified as Enterococcus faecium.As a result of the study in Van, Turkey, the isolation rate of Enterococcus faecalis and Enterococcus faecium strains were found to be lower than other regions.

The growth characteristics and the properties of the Enterococcus faecium (E. faecium) isolated from intestine of slaughting pig were examined in order to confirm whether it will be used practically as probiotics or not. E. faecium was identified by morphological and biochemical tests including of final confirming by API strep kit. Among 264 samples, 44 isolates (16.7%) of E. faecium were observed as an exellent growth as a range of 1×10∨7 CFU/ml~3.3×10∨9 CFU/ml in viable bacterial count on MRI medium.

Objective—To determine immunomodulatory effects of synbiotics administered in ovo on immune-related gene expression in adult chickens. Animals—30 Green-legged Partridgelike chickens. Procedures—On incubation day 12, eggs were injected with 3 synbiotics (Lactococcus lactis subsp lactis IBB SL1 with raffinose family oligosaccharides [RFOs; S1], Lactococcus lactis subsp cremoris IBB SC1 with RFOs [S2], and Lactobacillus acidophilus and Streptococcus faecium with lactose [S3]). Control eggs were injected with RFOs prebiotic or saline (0.9% NaCl) solution. Gene expression of 6 cytokines (interleukin [IL]-4, IL-6, IL-12p40, IL-18, interferon [IFN]-β, and IFN-γ) and 1 chemokine (IL-8) was analyzed in the cecal tonsils and spleen of 6-week-old chickens by means of reverse transcription quantitative PCR assays. Results—Gene expression for IL-4, IL-6, IFN-β, and IL-18 was significantly upregulated in the spleen of chickens in groups S2 and S3. In contrast, IL-12 expression was downregulated in group S2 and IFN-γ expression was downregulated in group S3. Expression of IL-8 did not change in chickens treated with synbiotics in ovo. Gene expression of all cytokines, except for IL-18, was downregulated in cecal tonsils. Conclusions and Clinical Relevance—In ovo administration of synbiotics activated the immune system in adult chickens. The intestinal immune system (cecal tonsils) had downregulation of expression for the cytokines evaluated, which indicated an increase in oral tolerance, whereas in the peripheral part of the immune system (spleen), expression of IL-4 and IL-6 was upregulated. Evaluation of immune-related gene expression patterns may be useful when monitoring the effectiveness of synbiotic selection with respect to immunobiotic properties.
