Molecular study of Cysticercus tenuicollis from slaughtered sheep in Sulaymaniyah province, Iraq

Cysticercosis caused by the larval stage of Taenia hydatigena is economically the most important endemic parasitic disease in Iraq. Few data are available relating to the genetic divergence of this helminth. This study aimed to molecularly characterise Cysticercus tenuicollis isolates from sheep in Sulaymaniyah province, Iraq. DNA extraction and amplification of specimens of C. tenuicollis from 46 sheep were conducted by PCR for the mitochondrial 12S rRNA gene. The 19 amplicons were subjected to purification and partial sequencing. Five 12S rRNA nucleotide sequence haplotypes were found. The pairwise nucleotide difference between haplotypes of 12S rRNA gene ranged from 0.2% to 0.7%. Four out of the five haplotypes of C. tenuicollis contained one to two base mutations and were discovered in Iraq for the first time, and this may be a unique mutation globally which has not been recorded previously. Three newly recorded haplotypes contained only one single mutation, and the other one contained two mutations. Phylogenetic analysis showed that all isolated strains were closely related to Iranian sheep isolates. Four new strains of T. hydatigena were discovered for the first time in the study area.

The occurrence of veterinary drug residues in chicken meat originating from 320 small and medium scale chicken slaughterhouses in Peninsular Malaysia was determined. 637 chicken meat samples were examined for tetracycline (TCs), sulphonamide (SAs) and quinolone residues using a microbiological inhibition test and was further confirmed using liquid chromatography mass spectrometry (LCMS/MS). The presence of TC residues were confirmed in 10 (1.6%) samples, and 1 (0.2%)sample was confirmed in compliance to the established maximum residue limit (MRL) for residues of quinolone. A total of 6 (0.9%) samples were above the MRL for TC. The samples were from Pulau Pinang, Terengganu and Kelantan. Among those tested in compliance, the main analytes found for TC and quinolone werechlortetracyclines (CTC), enrofloxacin and mixture of chlortetracycline (CTC) and oxytetracycline (OTC). No samples were found to contain sulfonamides residues.

Lipopolysaccharide (LPS) fractions were obtained from smooth cultures of Brucella abortus strains 2308 and S-19 by butanol extraction procedures. The LPS from the initial butanol extraction contained 10 to 15% protein and was reduced to less than 1% protein by treatment with proteinase K. The LPS fractions were identified and characterized on the basis of the chemical analysis, sodium dodecyl sulfate gel electrophoresis, cesium chloride gradients, electron microscopy, and gel immunodiffusion. Results indicated that the butanol procedure is a reliable method in the extraction of LPS from Brucella abortus cells. Proteinase K-treated LPS containing less than 1% protein from strain 2308 was used to vaccinate BALB/cByJ mice. Immune and protective criteria for vaccinated and nonvaccinated mice were increased immunoglobulin (IgG and IgM) titers in sera of prechallenge-exposed mice, reduced colony-forming units/spleen, and splenomegaly in post-challenge-exposed mice. Results indicated that proteinase K-treated LPS was immunuogenic as well as protective for mice.

Shrubs represent the most affordable and accessible form of feed that livestock can rely on to acquire both essential and non-essential elements of life. In addition to their inherent toxins, they contain endogenous substances commonly referred to as 'antinutritive factors' (ANFs) that often interfere with the utilisation of nutrients. Their abundance may lead to severe clinical trauma. Hence, the objective of the study was to investigate the effects of different extraction techniques on Nerium oleander L. and animal feeds as well as to quantify oxalates. Organic (hexane, acetone and methanol) sequential and aqueous (infusion and decoction) extractions were explored. Qualitative and quantitative analyses were conducted to determine the presence of various phytochemicals and oxalate contents as putative ANFs, respectively. The results showed higher extraction yields of 22.6% and 43.1% in the decoction and infusion of N. oleander, respectively. The quantification methods were validated for linearity, accuracy and precision. Oxalate contents of 6.76 ± 0.245 (0.65%) mg/g and 5.74 ± 0.236 mg/g dry weight (0.55%) were obtained in N. oleander and feeds, respectively. This difference was statistically significant with p < 0.05. Percentage recoveries of 98.5 (percent relative standard deviation [% RSD] = 2.3), 85.7 (% RSD = 1.03) and 80.3 (% RSD = 1.22) at 76%, 95% and 112% fortifications were obtained, respectively. Relative standard deviation for precision was 0.99% and 1.13% at 0.33 mg and 0.39 mg fortifications, respectively, while reproducibility showed 2.21% RSD. Therefore, these methods can be used to provide a valuable basis for qualitative determination of ANFs, particularly in shrub foliage.

Analysis of protein and antigen profiles of Tritrichomonas foetus isolates from cattle from 5 western states was accomplished sodium dodecyl sulfate polyacrylamide-gel electrophoresis, immunoblot, immunoprecipitation, and fluorography techniques. Total protein profiles of all isolates were compared by Coomassie brilliant blue staining of T foetus protein samples prepared by 4 protein-extraction methods. Antigenic tritrichomonas proteins were identified by immunoblot assay with polyclonal bovine or rabbit anti-T foetus serum. Additionally, [14C]glucosamine-labeled T foetus was used for total and antigenic glycoprotein analyses. Detectable differences in the composition of total proteins or antigenic tritrichomonal proteins were not observed among all isolates. However, intensity differences in some antigenic protein bands were apparent. Bovine and rabbit sera from immunized animals possessed antibodies to the same antigenic tritrichomonal proteins. Each T foetus isolate contained 4 to 7 molecular weight size classes of glycoprotein, which were labeled by [14C]glucosamine; however, only 3 to 4 glycoproteins were identified as antigens by bovine or rabbit antiserum.

The function of atrial natriuretic peptide (ANP) is claimed to be control of salt and water homeostasis, and thus, the hormone may be involved in the pathogenesis of certain diseases with impaired volume regulation. We, therefore, studied plasma ANP concentration in dogs with chronic renal failure, congestive heart failure, and hyperadrenocorticism. Dogs with chronic renal failure had twofold higher plasma ANP concentration (16.2 +/- 5.8 fmol/ml), compared with healthy dogs (8.3 +/- 3.5 fmol/ml). An even more distinct increase (sixfold) of plasma ANP concentration was found in dogs with congestive heart failure (52.9 +/- 29.7 fmol/ml). In contrast, dogs with hyperadrenocorticism did not have high ANP plasma concentration (5.5 +/- 2.0 fmol/ml). High-performance liquid chromatographic analysis of plasma from dogs with congestive heart failure indicated that, in addition to the normal circulating form of ANP (99-126), the unprocessed precursor ANP (1-126) is detectable in the circulation. These qualitative and quantitative alterations of plasma ANP concentration in dogs further suggest involvement of this peptide in the development and/or maintenance of diseases associated with impaired volume regulation.

A monitoring program for melamine in milk and feed was conducted in response to global melamine alertness in the year 2008. Two screening methods were adopted i.e., a liquid chromatography triple quadrupole tandem mass spectrometry (LC-MS/MS) and enzyme-linked
immunosorbent assay (ELISA). The liquid chromatography method developed by several international research centers was adapted. This method consisted of an initial extraction with 10%trichloroacetic acid (TCA) for milk samples or 60% methanol/water for feed samples, followed by a series of centrifugation, dilution and filtration steps. Melamine was analysed in the chromatographic program using a zwitterionic HILIC LC column. Electrospray ionisation in positive ion mode was used. The quantity of melamine
present was determined with a calibration curve consisting of sample extracts from milk or feed fortified from 25 to 50 ppb that were taken through the extraction procedure. The ranges of recovery from
fortified raw milk samples (n=20) and feed samples (n=21) was 70–80% and 68%, respectively. The limit of detection was estimated at 10 ppb for both matrixes. Milk samples were found negative for melamine,
however 4.5% of feed samples were found to contain the compound at concentrations between 1 to 5 ppm.

Objectives: This research aimed to determine the optimum type of solvent and extraction time to produce secondary metabolites (phenolics, flavonoids, tannins, and antioxidants) from mangosteen leaves (Garcinia mangostana L.) as feed additive candidates for poultry. Materials and Methods: This research used a completely randomized design with a 2 × 5 factorial design with three replications. Factor A used two types of distilled water as a solvent (ordinary distilled water and distilled water heated at 100°C), while Factor B encompassed various extraction times (15, 30, 45, 60, and 75 min). The parameters assessed included total phenolic content (TPC), total flavonoid content (TFC), total tannin content (TTC), and overall antioxidant activity. Results: The TPC, TFC, TTC, and total antioxidant activity all showed a highly significant interaction (p < 0.01) with the type of solvent and extraction duration. Conclusion: The best solvent and time for mangosteen leaf extract to produce secondary metabolites, which can be candidates for feed additives in poultry, is ordinary distilled water for 45 min. In this research, the phenol content was 81.03%, flavonoids 11.07%, tannins 1.01%, and antioxidants 77.61%. [J Adv Vet Anim Res 2024; 11(1.000): 139-145]

Studying of interaction of phytolectins with carbohydrate epitopes of cattle erythrocytes and alpha 1-4 D-glukan was realize in the conditions of the Republic of Belarus was realized by the example of seeds of soy (Glycine max) of Viliya variety, common bean (Phaseolus vulgaris) of Olga variety, spring barley (Hordeum vulgare) of VM-MGF variety, and wheat (Triticum aestivum) of Akogomugi varity. Lectins studies of pathological conditions (infringement of a cellular metabolism, transformation and cell destruction, etc.) were closely connected with research of structure and functions of cellular membranes that was important for carrying out of various biotechnological works. As a result of the realized research there was given the comparative characteristic of hemagglutinin activity of phytolectins of some bean and grain crops in relation to erythrocytes of cattle and precipitating with and 1-4 D-glukan. Intensive precipitation of alpha 1-4 D-glukan was observed in the conditions of its interaction with lectins of soya and wheat and less expressed in the conditions of interaction with lectins of bean and barley. It was established, that glucose monomers and its derivatives proved to be the basic structural element of glycocalyx of erythrocytes which detected the phytolectins of soya, beans, wheat and barley
