Cytological evaluation of the influence of high and low doses of bisphenol A on an erythroblastic cell line of porcine bone marrow

Introduction: Bisphenol A (BPA) is a substance widely used in industry for the production of polycarbonates and epoxy resins used in packaging and containers for beverages, contact lenses, compact discs (CDs), window panes, and many other elements. This compound belongs to the group of polyphenols and xenoestrogens commonly found in the human environment. What we know about BPA is still insufficient to enable us to protect our health against its adverse effects, and current knowledge of the influence of BPA on erythroblastic cell lines in bone marrow is rather fragmentary. The aim of the experiment was to assess the effect of two doses of BPA (0.05 mg/kg and 0.5 mg/kg b.w. per day) on myeloid haematopoiesis. Material and Methods: During this experiment, the number of all types of cells in the erythroblastic cell line was evaluated in porcine bone marrow before and after BPA administration. Results: The obtained results clearly indicate changes in haematopoietic activity of the bone marrow, which was demonstrated by a decrease in erythroblastic cell line production in both experimental groups. The haematological effects of the bone marrow changes were anaemia, caused by a number of erythrocytes which was depressed due to their immaturity, and a significant decrease in mean cellular volume in both groups. Conclusion: The harmful effect of high and low doses of BPA on haematopoietic processes was proved.

Introduction: The purpose of this study was to investigate the therapeutic effect of hydrogen on the therapy of onion poisoned dogs. Material and Methods: A total of 16 adult beagle dogs were divided into two groups (control and hydrogen) and all were fed dehydrated onion powder at the dose of 10 g/kg for three days. The dogs of the experimental group were given subcutaneous injection of 0.2 mL/kg of hydrogen for 12 days after making the poisoned model successful. Blood samples were collected before feeding onions, one day before injecting hydrogen, and 2 h after the injection of hydrogen on days 1, 3, 5, 7, 9, and 12. Control dogs were not treated with hydrogen. Results: The levels of leukocyte production, anaemia, red blood cell degeneration which was reflected by the values of Heinz body count, haemolytic ratio, and oxidative products in hydrogen treated group were lower than in control dogs on some days. The capacity of medullary haematopoiesis that was based on reticulocyte counts, and the antioxidation in hydrogen group were higher compared with control group. However, the differences in renal function were not obvious in both groups. Conclusion: Accordingly, it was concluded that subcutaneous injection of hydrogen could alleviate the symptoms in onion poisoned dogs.

We investigated the potential of fucoidan for its ability to provide protection from gamma ray-induced damage. In our results, the fucoidan significantly improved the counts of endogenous colony forming unit to 9.5±1.5, from 5.5±2.5 compared with un-treated irradiated control group at 10 day after 7 Gy whole body irradiation. After 2 Gy irradiation, fucoidan treatment attenuated the percent of tail DNA of splenocytes, parameters of DNA damage, from 30.17±1.7% to 13.67±2.81% by comet assay and also accelerated the proliferation of splenocytes, compared with un-treated irradiated control group by ³H-thymidine incorporation assay. Furthermore, fucoidan decreased the number of apoptotic fragments per intestinal crypt by 31.8% at 1 days after 2 Gy irradiation. These results indicated that the fucoidan significantly improved the hematopoietic recovery, prevented the DNA damage in immune cells and enhanced their proliferation, which had been suppressed by ionizing radiation. in addition, fucoidan rescued intestinal cells from radiation-induced apoptosis. Thus, this study raises the possibility of using fucoidan as adjuvant therapeutic agent after radiotherapy.

Direct effects of equine infectious anemia virus (EIAV) on hematopoiesis in vitro were studied. Bone marrow mononuclear cells from clinically normal horses were incubated with 100 TCID50 of EIAV/10(7) cells. These cells were cultured to assay for colonies derived from erythroid progenitors, granulocyte/monocyte progenitors, and fibroblastic progenitors. The EIAV had a selective suppressive effect on the erythroid progenitors. Colony-forming units-erythroid were suppressed to 80% of that for medium controls (P = 0.011). Burst-forming units-erythroid were suppressed to 70% of that for medium controls (P = 0.003). Significant effect was not apparent on colony-forming units-granulocyte/macrophage or on colony-forming units-fibroblastic.

A variety of in vitro cloning assays have been used for studying hematopoiesis in mice and human beings. However, these techniques have had limited use in dogs, a species used extensively as a model for hematopoietic research, particularly hematotoxicity. We have adopted cloning assays for in vitro growth of canine colony-forming unit-erythroid (CFU-E) and colony-forming unit-granulocyte/macrophage (CFU-GM) progenitor cells, using modified microplasma clot and soft agar culture systems respectively. Marrow mononuclear cells separated by density-gradient centrifugation were added to the aforementioned culture systems. Erythroid colonies were stimulated with sheep plasma erythropoietin and incubated at 37 C in 5% CO2 for 2 days. The CFU-E colonies were fixed with 5% glutaraldehyde, stained with benzidine, counted, and expressed as a mean of 8 replicates. The CFU-GM colonies were stimulated with pooled serum from endotoxin-treated dogs and incubated for 8 days at 37 C in 10% CO2. Using an inverted microscope, the CFU-GM colonies were counted and expressed as a mean of 6 replicates. The number of colonies was proportional to the plated cell concentrations. The addition of 10% autologous serum to CFU-GM cultures increased the number of colonies by 80 to 100%, but markedly reduced the size and number of CFU-E colonies. The marrow cloning capacity among dogs of comparable age was similar, and little variation was noticed when bone marrow cells from the same dogs were cultured repeatedly over a period of 3 to 4 months. We concluded that these cloning assays are fast, reliable, and reproducible and that they allow quantitative determination of canine hematopoietic progenitor cells. The assays may be useful in screening the hematotoxic potential of various therapeutic agents and are particularly suited for studying the pathogenetic mechanisms of drug-induced blood disorders and their reversibility.

Morphological and structural changes in bone marrow of piglets which were immunized with produced in the Republic of Belarus SPS vaccine (against salmonellosis (Salmonella), pasteurellosis (Pasteurella) and streptococcus) in combination with immunostimulants and without them were analyzed in the conditions of the Republic of Belarus. Piglets of the first group were vaccinated against salmonellosis, pasteurellosis and streptoccocusis by SPS vaccine. Piglets of the second group were vaccinated by SPS vaccine with vitamin C. Piglets of the third group were vaccinated by SPS vaccine with immunotstimulant sodium thiosulphate. The fourth (control) group of animals was administrated with normal saline. Animals were immunized twice, intramuscularly with 7 day interval in dose of 4 ml (primary), and 5 ml (secondary). Vitamin С was administrated in dose of 0,05g per head. Sodium thiosulfate was administrated in combination with the vaccine in 30% concentration. Alongside with myelogram there were derived formulas of different cell groups of bone marrow: leucoerythroblastic index; intramedullary index of neutrophil maturity; intramedullary index of eosinophile maturity. Detailed myelogram of piglets on the 14-th day after the second immunization was presented. Research results showed that in all groups of vaccinated animals there was noted the activation of myeloblastic hematogenesis and decreasing of erythropoeisis. Piglet immunization was accompanied by strengthening of myeloblastic hematogenesis, increased number of lymphocytes and plasma cells in bone marrow

Searching for new, effective and safe methods of treatment of hookworm disease (ancylostomatidosis) of dogs caused by Ancylostoma caninum and Uncinaria stenocephala was realized in the conditions of the Republic of Belarus on the basis of studying of spices composition and dissemination of Ankilostomatidae, age-specific and seasonal disease dynamics, testing of medical and preventive preparations Univerm and Fenbendazol. In the Republic of Belarus Ancylostoma caninum was stated 4,2% of studied cases and Uncinaria stenocephala – in 14,3%. The maximum intensity of the invasion was stated in summer and autumn periods: in cases of individual keeping – 19,4%; in cases of group keeping the disease transformed into enzootic form with 66,7% affect. In winter period there was the lowest invasion – up to 1,4%. The preparation Univerm administrated twofold with feed in dose of 0,1 mg/kg with 24 hours interval proved to be highly effective therapeutic agent with extensefficiency100%. Anthelmintic Fenbendazol administrated once with feed in dose of 0,005 g/kg proved to be highly effective with extensefficiency 80% and intensefficiency 98,84%. Hematological indexes of sick dogs showed the evidence of erythropenia (4,30-4,80 x 10E12/l), hemoglobinemia (113-119 g/l) and eosinophilia (up to 11%), as well as the increased quantity of leukocytes on 5,46% - 23,61 %, and the lowered quantity of lymphocytes on 15,84 % in comparison with healthy animals indexes
