Phylogenetic study of Ostertagia species

BACKGROUND: Ostertagia species are the main domestic ruminants abomasum worms, which cause parasitic gastritis. Parasitic gastritis is associated with disruption of the endocrine structure of abomasum, and influence the function and pH of abomasums. This can lead to weight loss, reduced milk and wool production with potentially mal-digestion of proteins. These cause the economic loss to animal husbandry industry. So, the national research priority in veterinary parasitology is identification of different native species of Ostertagia in Iran. OBJECTIVES: The main purpose of this study is identification and differentiation of Ostertagia species based on the molecular characterization. METHODS: During carcasses inspection at different abattoirs in Rey, Tehran, Mashhad and Bandar Abbas, 180 adult male worms of Ostertagia species from sheep and goat were collected. Morphological analysis was performed based on the morphometric characteristics of spicules. The ITS1, 5.8s and ITS2 ribosomal DNA was ampilified from individual worms by polymerase chain reaction (PCR) and then purified PCR product were sequenced and analyzed by Blast tool. RESULTS: 946 bp PCR products for all sequenced samples were compared with the released sequences of Ostertagia isolates available from GenBank and showed 2-3% differences and 97-98% similarity. CONCLUSIONS: The three species included in the present study are different in terms of molecular property (with 2-3% difference) and it is necessary to determine transmittance pattern and host affinity rules from each one to be used by program managers and evaluators for Prevention and Control programs.

Anthelmintic efficacy of levamisole against induced infections with 7- and 21-day-old Haemonchus contortus, Ostertagia circumcincta, Trichostrongylus axei, and T colubriformis was evaluated as an oral drench in goats. Group 1 (n = 8) was not treated, group 2 (n = 8) was given 3.96 mg of levamisole/kg of body weight, group 3 (n = 8) was given 7.92 mg of levamisole/kg, and group 3 (n = 7) was given 11.88 mg of levamisole/kg. Efficacy against all worms was low in goats given 3.96 mg of levamisole/kg, but was high against adult H contortus (99%) and adult T colubriformis (99.7%) in goats given 7.92 mg of levamisole/kg. Although efficacy against adults of all species was high in goats given 11.88 mg of levamisole/kg, some immature worms of all species remained in the abomasa of goats.

Occurrence of gastrointestinal parasites in camels in the Tianshan Mountains pastoral area in China

Gastrointestinal parasites are some of the most common pathogens which are seriously harmful to the camel’s health. The infection status of gastrointestinal parasites in camels (Camelus bactrianus) in the Tianshan Mountains pastoral area in China is still unclear. The aim of this study was to investigate the species and infection intensity of gastrointestinal tract parasites in local camels. A total of 362 fresh faecal samples were collected and examined for parasite eggs using the saturated saline floating and natural sedimentation method. The parasite eggs were subjected to morphological and molecular examination and identification, and the infection rate and mean intensity of the parasites were analysed. A total of 15 gastrointestinal tract parasite species’ eggs were identified, with a detection rate of 100%. Ostertagia spp. (100%) and Trichostrongylus spp. (98.1%) were dominant. Camels were often coinfected by 5–14 species. The average number of eggs per gram of faeces was higher for Ostertagia spp. (298), Haemonchus contortus (176) and Nematodirus spp. (138). The number of species of parasites infecting young camels was significantly lower than that of adult camels, but the infection intensity in young camels was significantly higher. Gastrointestinal parasites were highly prevalent in camels from the Tianshan Mountains pastoral area in China. This finding provides important epidemiological data for the prevention and control of associated infections in camels.

Objective--To evaluate the nematocidal effectiveness of the ivermectin sustained-release bolus throughout its 135-day delivery period. Design--Twenty-four naturally infected calves were randomly allocated to 1 of 3 equivalent experimental groups: group-T1 calves were untreated controls, group-T2 calves each received a sustained-release bolus on trial day 0 and group-T3 calves were rendered nematode-free and used at 35-day intervals during the study as tracers. One contaminated pasture was used for all principal calves for the 135-day grazing interval of the study. Calves of groups T1 and T2 were also artificially administered mixed infective nematode larvae at intervals during the grazing period, after which, all calves were confined to concrete for 21 days prior to necropsy. Animals--All calves were approximately 6 months old on trial day 0, weighed from 136 to 216 kg, and were of mixed breeding and sex. Procedure--At intervals during the study, feces from all calves were analyzed for nematode egg counts, and all calves were weighed and examined for bolus retention (T2 calves only). For nematode recovery, all calves were necropsied 21 to 22 days after removal from the contaminated pasture. Results--Parasitic populations of Haemonchus, Ostertagia, Trichostrongylus, Cooperia, Bunostomum, and Oesophagostomum spp were significantly reduced in cattle treated with the ivermectin sustained-release bolus. Conclusion--The nematocidal activity of the ivermectin sustained-release bolus proved highly effective, with > 98% efficacy for all nematode species present.

The efficacy, safety, and compatibility of fenbendazole (FBZ) and clorsulon (CLN) were tested after oral administration of label recommended and of higher (5 x) dosage rates to calves naturally infected with gastrointestinal nematodes and Fasciola hepatica. Results for 42 calves allotted to 4 treatment groups indicated a similar efficacy against mature F hepatica by FBZ (5 mg/kg of body weight) and CLN (7 mg/kg) in a combined oral suspension, compared with CLN (7 mg/kg) alone (100 vs 99% reduction). A lesser efficacy was observed against immature flukes (88.6 and 84.9% reduction, respectively). Calves given 25 mg of FBZ/kg and 35 mg of CLN/kg had nearly complete reduction of both mature (99.6%) and immature flukes (99.1%). Fasciola egg counts were reduced by > 99.5% in all treated groups. Against Ostertagia ostertagi, the percentage of efficacy of the combined FBZ (5 mg/kg) and CLN (7 mg/kg) treatment was 94.3% against adults and 81.3% against inhibited larvae. Efficacy against all other nematodes was 100%, except against Cooperia spp adults (98.3%) and immature Oesaphagostomum radiatum (88.0%). At 5 x dosage rates for FBZ and CLN, percentage of removal of adults and inhibited larvae of O ostertagi was 99.3 and 99.0%, respectively, and 99 to 100% for other nematodes. Results indicate that FBZ and CLN are compatible when mixed together and administered as an oral suspension to cattle and that the efficacy is similar to that of the drugs individually. On the basis of further results, we suggest that summer treatment may be superior in preventive value for gastrointestinal nematodes and F hepatica, compared with spring treatment, because of seasonal infection dynamics of the major cattle parasites in Louisiana.

The possible development of type-1 hypersensitivity reactions in the abomasal mucosa caused by soluble L3 products of Ostertagia ostertagi was studied in 4-month-old calves sensitized by repeated exposure to L3 over a 50-day period followed by anthelmintic treatment. Four groups each of 4 calves were used. Group 1 served as nonsensitized controls and group 2 as sensitized controls, group 3 was challenge exposed at 2-week intervals beginning at week 10 with a soluble L3 product (OAG), and group 4 was challenge exposed at 2-week intervals with an oral dose of L3, followed by anthelmintic treatment 3 days later. All calves infected with L3 became sensitized, as indicated by a positive reaction to an intradermal skin test. However, a passive cutaneous anaphylaxis was only partly effective in indicating the presence of homocytotropic antibody in the infected calves. Sensitized calves had significantly (P < 0.05) higher eosinophil counts and plasma pepsinogen values for the entire 14 weeks than uninfected controls. Globule leukocyte and mast cell counts from the abomasal mucosa were also significantly (P < 0.05) higher. Studies for possible immunomodulation revealed that lymphocyte counts decreased between every 2-week challenge-exposure period for groups-3 and -4 calves. A transient depression of blood lymphocyte (BL) responses to phytohemagglutinin (PHA), a T-cell mitogen, was observed over the first 8 weeks in the infected calves. Increases in BL responses to OAG were also observed. Differences were not observed in BL responses to pokeweed mitogen, a T- and B-cell mitogen. Blood lymphocyte responses to PHA in group-3 calves were low following the initial challenge exposure with OAG. The sensitized calf lymphocytes did not have suppressive activity on the response of control calf lymphocytes to PHA. Differences were not observed in lymphocyte responses to PHA in a suppressive assay done on abomasal lymph node lymphocytes. Increases in abomasal lymph node mass and lymphocyte responses to PHA, pokeweed mitogen, and OAG were observed in all sensitized calves. Histologic examination of abomasal lymph node sections from challenge-exposed calves revealed increased mitotic activity in germinal centers. Plasma pepsinogen values in groups 3 and 4 increased between each challenge exposure, which further suggested that type-1 hypersensitivity reactions had occurred in the abomasal mucosa, resulting in increased permeability and leakage of macromolecules.

Twelve calves (mean weight, 175.5 kg) were used to confirm efficacy of ivermectin delivered from a prototype sustained-release bolus against naturally acquired gastrointestinal nematodes including early fourth-stage (inhibited) larvae of Ostertagia ostertagi. The calves were allocated by restricted randomization on weight to 1 of 2 groups: controls, to which a placebo bolus was given orally, and treated calves, to which a sustained-release bolus designed to deliver 8 mg of ivermectin/day at a steady rate was given orally. After treatment, the 2 groups were housed in separate pens with concrete flooring. Twenty-eight days after treatment, all calves were euthanatized and necropsied. The ivermectin-treated calves had no larval or adult Ostertagia spp and significantly (P < 0.01) fewer adult Trichostrongylus axei and adult Cooperia (C oncophora, C punctata and C surnabada) than control calves. Efficacy of ivermectin was > 99% for Cooperia spp, and 100% for other parasites. Drug-related adverse reactions were not observed.
