Quantitative CT Analysis of Pulmonary Pattern in Dogs Affected by Pneumonia, Before and After Intravenous Contrast Medium Administration

BACKGROUND: Alveolar pattern of the lung field is usually caused by several lung involvements such as edema, hemorrhage and pneumonia, but differentiation using radiography is usually impossible. Objectives: Evaluation of HU and lung pattern of pneumonia in dogs using CT-scan technique and confirmation by CT assisted FNA as a safe diagnostic procedure. Methods: In this study, 10 dogs with respiratory distress suspected for pneumonia were selected and radiography and CBC were used for evaluation of lung pattern and cytology. Before and after contrast medium injection, CT-scans of the lung field were achieved. Finally, in transverse CT sections, FNA of involved lung lobes was taken for cytologic evaluation. After the confirmation of pneumonia, CT scans were reevaluated with more attention and pulmonary pattern was described and HU of involved regions was measured. Results: In all of the patients, ventral region of the lungs, especially cranial and middle lung lobes were involved and the involvement of the left lungs was prominent. The mean HU of the unhealthy lung parenchyma before contrast medium study was between 31 and 39, and after contrast study was between 46 and 70. It means the HU has been significantly increased. Also, the contrast of the involved lung regions was increased which helped to differentiate the unhealthy lung tissue from the adjacent soft tissues. Hematology tests in all of the patients were representative of infection. Conclusions: Both CT-scan and CT assisted FNA from the lung are beneficial and practical methods for the diagnosis of pulmonary disorders such as pneumonia

BACKGROUND: Annexins (including annexin A1 and annexin A2) are important proteins which have some roles in organisms such as intracellular signal conduction, membrane cellular skeletal connection, cellular proliferation and differentiation, especially inhibitory function in inflammation processing. Pasteurella multocida is the most common bacterial pathogen and has high prevalence in pneumonia. Objectives: This study was aimed to determine experimentally annexin A1 and annexin A2 in the serum of calves affected by Pasteurella multocida pneumonia. Methods: In this research, 10 male calves (2 - 4 months) were allocated to two equal groups, one group as the case: 300 ml in dilution 2×109 CFU Pasteurella multocida bacteria and the other as control group: 300 ml normal saline inoculated by special lavage catheter through oral to trachea. Clinical scores were recorded based on available tables. In treatment group, about 18 to 24 hours after inoculation and synchronous with observation clinical signs changes, bronchoalveolar lavage for cytology and bacteriology examination were done of fluids results from washing. Blood sampling was taken from calves jugular vein in both groups then blood serums were examined by using ELISA kits. Results: The rates of annexin A1 and annexin A2 in blood serum of treatment group showed significant increase (using independent t test) compared to control group (p<0.05). Conclusions: It seems these annexins (annexin A1 and annexin A2) can be used as important biomarkers in blood serum to diagnose inflammation processes such as pneumonia.

From 2 to 4.5 months of age, 80 crossbred gilts were reared in a conventional grower unit where they were naturally exposed to mycoplasmal and bacterial pathogens that cause pneumonia and atrophic rhinitis. At 4.5 months of age, gilts were moved to environmentally regulated rooms (4.9 X 7.3 m) and assigned at random to 1 of 2 treatment groups: low aerial concentration of ammonia (4 to 12 ppm; mean, 7 ppm) or moderate aerial concentration of ammonia (26 to 45 ppm, mean, 35 ppm). Low concentration of ammonia was obtained by flushing of manure pits weekly, whereas moderate concentration of ammonia was maintained by adding anhydrous ammonia to manure pits that were not flushed. Gilts were weighed biweekly. Mean daily gain (MDG) was less (P < 0.01) for gilts exposed to moderate concentration of ammonia than for gilts exposed to low concentration of ammonia after 2 weeks in their respective environments. By 4 and 6 weeks, however, MDG was similar between the 2 treatment groups. After 6 weeks in these environments, 20 gilts from each treatment group were slaughtered, and prevalence and severity of lung lesions and snout grades were determined. At slaughter, body weight was greater (P < 0.01) in gilts exposed to low, rather than moderate, ammonia concentration (94.5 vs 86.8 kg; SEM, 3.3 kg). Percentage of lung tissue containing lesions (18 vs 12) and snout grade (2.8 vs 3.1) were similar between gilts exposed to low or moderate concentration of ammonia. The remaining 20 gilts in each treatment group were maintained in their respective environments, exposed daily to mature boars and bred at first estrus. Age at puberty was similar between gilts exposed to low or moderate concentration of ammonia (208 vs 205 days; SEM, 1.3 days), even though weight at puberty was less (P < 0.03) for gilts exposed to low concentration of ammonia than for gilts exposed to moderate concentration of ammonia (109.7 vs 118.2 kg; SEM, 4.5 kg).

Six Rocky Mountain bighorn sheep were raised in captivity from birth (n = 5) or taken from the wild as a lamb (n = 1). After the bighorn sheep were in captivity for over a year, 6 clinically normal domestic sheep were placed on the 2 ha of pasture on which the bighorn sheep were kept. Nasal swab specimens were obtained from all sheep at the time the domestic sheep were introduced. Pasteurella haemolytica was isolated from swab specimens obtained from 4 of 6 domestic sheep, but not from specimens obtained from the bighorn sheep. All 6 bighorn sheep died of acute hemorrhagic pneumonia after exposure to domestic sheep. Death in the bighorn sheep occurred on days 4, 27, 27, 29, 36, or 71 after initial exposure to domestic sheep. Pasteurella haemolytica was isolated from respiratory tract tissue specimens of all bighorn sheep at the time of death. None of the domestic sheep were clinically ill during the study. At the end of the study, 3 of 6 domestic sheep were euthanatized, and at necropsy, P haemolytica was isolated from 2 of them. The most common serotypes in bighorn and domestic sheep were P haemolytica T-3 and A-2. Other serotypes isolated included P haemolytica A-1, A-9, and A-11 in bighorn sheep and A-1 in domestic sheep. On the basis of results of this study and of other reports, domestic sheep and bighorn sheep should not be managed in proximity to each other because of the potential fatal consequences in bighorn sheep.

A method of inducing Pasteurella haemolytica serotype 1 (Ph1) lung infection in goats, using low numbers of bacteria and without impairing host immunity, was developed. Two trials were conducted. Results of trial 1, using 10 principals (Ph1 agar beads) and 6 controls (agar beads alone), indicated that Ph1 organisms imbedded in agar beads could survive host lung defenses for 32 days. Results of trial 2 indicated that lung immunity in the inoculated goats (principals) was high and they were more protected than controls against a transthoracic challenge of Ph1 (1.18 X 10(7) colony-forming units) injected into lung of each goat on posttreatment day 35. When comparing challenge-exposed principals with controls, the controls developed rectal temperatures above normal for a longer time, duration of anorexia was longer, and sign of depression were seen. The controls developed large are of consolidated lung tissue, more Ph1 isolates were recovered from nasal turbinates and lung tissue, and higher Ph1 concentrations were found in the lungs. The serum Ph1 indirect hemagglutination antibody titers in the principals of both trials increased, compared with titer in controls. Principal goats in trial 2 had higher Ph1 indirect hemagglutination antibody titers after injection of Ph1-impregnated agar beads and less severe lung lesion after challenge exposure than did controls. The small pneumonic consolidated lesions in the principals, compared with extensive lesions in controls after Ph1 challenge exposure, indicated a high degree of immunity after exposure to Ph1 organisms imbedded in agar beads.
