Determination of ability of a thymidine kinase-negative deletion mutant of bovine herpesvirus-1 to cause abortion in cattle

The Cooper isolate of bovine herpesvirus-1, which causes abortion in cattle, was used to construct a thymidine kinase-negative (TK-) deletion mutant virus. Twelve heifers were inoculated IV at 25 to 29 weeks of pregnancy with either TK- or thymidine kinase-positive (TK+) Cooper virus. All heifers developed fevers of 1 to 2 C during the first week after inoculation. Temperatures of TK+ inoculates were slightly higher and remained above normal a few days longer than in TK- inoculates. Viremia was detected in 5 of 6 TK+ inoculates and in all 6 TK- inoculates. More virus isolations were made from nasal and vaginal swab specimens of TK+ inoculates than from swab specimens of TK- inoculates. All heifers developed virus neutralizing antibody within 14 days after inoculation and antibody titers were similar between the 2 groups. None of the TK- inoculated heifers aborted and their calves did not have neutralizing antibody at birth. Abortion occurred in 5 of 6 heifers given TK+ virus. All aborted fetuses were infected with bovine herpesvirus-1, as demonstrated by virus isolation or detection of viral antigen in fetal tissues. These results indicate that inactivation of the TK gene reduces abortifacient activity of bovine herpesvirus-1.

Seventeen sows were fed 1,000 Toxoplasma gondii oocysts of isolates GT-1 or PT-1 at 32 to 92 days of gestation, and the products of conception were examined for T gondii antibodies and parasites. Twelve of these sows were euthanatized near term between 21 and 62 days after being fed T gondii; fetal body fluids or fetal sera were examined for agglutinating T gondii antibodies, and tissues were bioassayed in mice for T gondii parasites. Six sows produced pigs that had been transplacentally infected with T gondii; one of them aborted a T gondii-infected fetus 17 days after ingesting oocytes. Agglutinating antibodies were detected in fetuses infected in utero, but transplacental transfer of T gondii antibodies was not observed in noninfected fetuses. Transcolostrally acquired T gondii antibodies disappeared by 3 months of age. Diagnosis of transplacental toxoplasmosis was confirmed on the basis of detection of T gondii organisms in fetal tissues by use of histologic examination and bioassay in mice. In conclusion, finding of T gondii antibodies in body fluids could serve as a rapid screening test for transplacental T gondii infection in pigs.

Plasma glucocorticoid concentrations and blood gas values were determined for 6 days in 47 newborn calves that had been subjected to various obstetrical procedures at term. Concentrations of glucocorticoids were uniformly high at birth (70 to 103 ng/ml). Increasing degrees of acidosis were accompanied by increasing glucocorticoid concentrations in plasma. Plasma glucocorticoid concentrations decreased sharply during the first 6 hours after delivery and reached a plateau at 48 hours after birth (14 to 21 ng/ml). The latter was taken as an indication that adaptation had been achieved. Calves subjected to severe pulling had higher glucocorticoid concentrations at birth (110.4 ng/ml) than calves requiring no assistance (88.3 ng/ml), calves requiring only slight assistance (83.8 ng/ml), or calves that had been delivered by cesarean section (82.9 ng/ml).

A superovulatory and surgical protocol was developed for recovery of bovine zygotes. Holstein cows and heifers were given follicle-stimulating hormone and cloprostenol to induce superovulation. Surgical cannulation and lavage of the uterine tube was performed 40 to 48 hours after the start of standing estrus. In general, cows had more corpora hemorrhagica than did heifers, but a higher percentage (P < 0.05) of ova recovered from cows were infertile. Several heifers were subjected to the procedure twice, and embryo recovery rates were equivalent both times.

Spinal-evoked potentials were recorded from 2 litters of clinically normal mixed-breed dogs between 35 and 300 days of age. Summated responses to tibial nerve stimulation were recorded from percutaneous needle electrodes placed at L7-S1, L4-5, T13-L1, C7-T1, and the cisterna cerebellomedullaris. The ulnar nerve was stimulated with recordings at C7-T1 and the cisterna cerebellomedullaris. Amplitudes did not change significantly with age, but were significantly (P < 0.05) different between various recording sites. On day 35, segmental and overall (L7-cisterna cerebellomedullaris) conduction velocities were less than half of the adult values. Spinal cord conduction velocities increased with age, reaching adult values at approximately 9 months of age. It was determined that quadratic equations best predicted the conduction velocities during maturation.

The influence of cortisol on estrogen synthesis by the bovine placenta and the importance of the delta 4 and delta 5 pathway for estrogen production were investigated. For experiment 1, portions of fetal villi (200 mg) were incubated for 48 hours with 0, 10, 100, and 1,000 ng of cortisol/ml with [3H]androstenedione (3H-A) or [3H]pregnenolone (3H-P5). Villi were also incubated for 4, 28, and 52 hours with or without cortisol (500 ng/ml) and with 3H-A or 3H-P5 (experiment 2). The conversion of various [3H]steroid metabolites such as A, P5, 17 alpha-OH-pregnenolone (17 alpha-OH-P5), progesterone (P4), 17 alpha-OH-P4, cholesterol (chol), and chol plus lipoprotein (500 micrograms/ml) into estrogen was measured during a 4-hour incubation (experiment 3). In experiment 1, cortisol increased conversion of 3H-A and 3H-P5 into estrogen by 3 to 41% and 7 to 34%, respectively, in a dose-dependent manner (P < 0.05). In experiment 2, times of incubation did not influence conversion of 3H-A into estrogen, which, however, was increased significantly (P < 0.05) over all times of incubation by administration of 500 ng of cortisol/ml. Conversion of 3H-P5 into estrogen increased over time of incubation and was stimulated by cortisol (P < 0.05). However, there was no interaction between cortisol treatment and time of incubation. In experiment 3, conversion of 3H-A, 3H-P5, and 3H-17 alpha-OH-P5 into estrogen was greater than the conversion of the other precursors tested. Mean conversion of 3H-A, 3H-P5, 3H-17 alpha-OH-P5, 3H-P, 3H-17 alpha-OH-P4, 3H-chol, and 3H-chol plus lipoprotein was 23%, 10.6%, 11.0%, 1.8%, 1.8%, 0.3% and 0.7%, respectively. Our results suggest that, in cows, the delta 5 pathway is the preferred pathway for placental estrogen synthesis and that cortisol directly stimulates estrogen production, probably by activating enzymes involved in this pathway.

One hundred fifty Se-deficient, pregnant, crossbred beef cows were assigned to 1 of 4 treatment groups: group A, Se-deficient control; group B, 1 Se bolus at 0 and 119 days; group C, 1 Se bolus at 0 days; and group D, 2 Se pellets at 0 days. The Se bolus is an osmotic pump designed to release 3 mg of Se/d into the reticulorumen. The Se pellets weigh approximately 30 g and contain 10% elemental Se, which is liberated in the reticulorumen. The Se bolus is designed to provide Se supplementation for 120 days and the Se pellets provide supplementation for up to 18 months. Cattle were maintained on Se-deficient pasture or forages prepared from these pastures for the duration of the experiment. Blood samples were collected from cows prior to treatment (time 0) and at 28, 52, 119, and 220 days thereafter and analyzed for blood Se (BSe) concentration. Body weights were recorded at each sampling time. Blood Se concentration of cows from all supplemented groups were significantly (P < 0.01) higher than control values at all sample dates after treatments began. By the end of the 220-day study, treatment group-B cattle had significantly (P < 0.01) higher BSe concentrations than any other group. Body weights of treatment groups fluctuated throughout the study, but did not differ (P > 0.05) between groups. One cow and 6 calves born to cows during the experimental period died. Necropsy of 5 calves provided no evidence linking these deaths to treatments. A difference (P > 0.05) in mortality between groups was not detected. Blood samples were collected from calves prior to suckling, and were analyzed for BSe concentration. Colostrum samples were collected from dams and analyzed for total Se concentration. Additional blood samples were collected from calves 24 to 48 hours after suckling and analyzed for BSe concentration and serum creatine kinase activity. Birth weight, gender, and health were recorded for all calves. Calves from cows in Se-supplemented groups had significantly (P < 0.001) higher BSe concentrations, both before and after suckling, than did controls. Postsuckle BSe concentrations within the groups of calves were not significantly (P > 0.05) different than presuckle BSe concentrations for any of the groups. Selenium concentrations in colostrum from Se-supplemented cows were significantly (P < 0.001) higher than from control cows. A difference (P > 0.05) was not determined in serum creatine kinase activities or birth weights between groups.

Efficacy of a repositol formulation of melengestrol acetate (MGA) for inhibition of pregnancy was determined in one hundred 14- to 16-month-old beef heifers. Nonpregnant heifers were allocated on the basis of weight into 5 groups of 20 heifers each and were given 0, 30, 60, 90, or 120 mg of repositol MGA, SC, on day 0. Four bulls, determined to be satisfactory potential breeders, were pastured with the heifers from postinjection days (PID) 7 to 177. The day of gesation was estimated for each heifer by rectal palpation at PID 59, 91, 134, 177, and 225. Heifers not pregnant by PID 177 were assigned a day of conception of greater than 177. For heifers given 0, 30, 60, 90, and 120 mg of MGA, respectively, the percentage pregnant at PID 177 was 95, 95, 50, 15, and 15%, and the median day of estimated conception was PID 21, 87, 175, greater than 177, and greater than 177. Repositol MGA significantly (P less than 0.001) affected the distribution of conception times over all doses. Average daily gain (+/- SEM) for 178 days was 0.28 +/- 0.04 kg, 0.24 +/- 0.03 kg, 0.33 +/- 0.04 kg, 0.40 +/- 0.03 kg, and 0.35 +/- 0.03 kg for heifers given 0, 30, 60, 90, and 120 mg of MGA, respectively. Increased dose of repositol MGA was associated with increased average daily gain, but this effect was not apparent when days pregnant were taken into account. Repositol MGA was an effective contraceptive for pastured heifers and the duration of its effect was dose-dependent.

Serum protein electrophoresis was performed on 71 clinically healthy juvenile and adult llamas (6 juvenile males, 7 juvenile females 25 adult males, 13 adult females, and 20 pregnant females) to determine normal serum protein concentrations. Values were reported for each of the 5 groups because the groups were not homogeneous in all 8 peaks. Although the values reported here may serve as reference values for adults, they represent only a guideline for the juveniles because of the limited number of animals in each of these groups.

Fetal and neonatal lamb hemostasis were studied from the 60th day of pregnancy to birth. Platelet counts and blood coagulation, as assessed by tests such as recalcification time and thromboelastography, were similar in fetuses, neonates, and adult sheep. The values of coagulation factors were low, ie, vitamin K-dependent Factors II, VII, IX, and X remained unchanged (30 and 40% of adult reference values) until the last 10 days of gestation, and then increased until birth (40 to 60%). Values of fibrinogen and Factor V followed a similar pattern, although their activities became identical to adult values at birth. Also, we measured values of protein C and antithrombin III, which are synthesized by the liver. The importance of hepatic failure and fetal vitamin K deficiency were discussed. Factors VIII and XII activities increased gradually during pregnancy to reach adult values at birth. Fetal fibrinolytic activity increased. This could not be explained by the values or tissue-type plasminogen activator (it was not detectable) or by the presence of its fast-acting inhibitor, whose concentration did not decrease.
