Effects of vincristine and prednisone on platelet numbers and function in clinically normal dogs

Effects of a single IV administered therapeutic dose of vincristine sulfate on platelet numbers and function were evaluated in 16 clinically normal dogs over the 2 weeks after drug administration. Results were statistically compared with those of a previous control study in which the same 16 dogs were administered saline solution (IV), instead of vincristine. Of the 16 dogs, 8 were orally administered daily immunosuppressive doses of prednisone concurrently throughout the saline-control and vincristine study periods. Platelet numbers and mean platelet volume were measured, using an automated hematology analyzer. Platelet function was evaluated by turbidimetric measurement of platelet aggregation in response to collagen, platelet-activating factor, and adenosine diphosphate (ADP), and by clot retraction (diluted whole-blood method) and buccal mucosa bleeding time. Vincristine had a significant (P < 0.05) effect on circulating platelet numbers. Vincristine induced a transient mild decrease in platelet numbers, followed by a moderate increase in numbers, with peak platelet count observed 8 days after drug administration. Mean platelet volume was not significantly affected by administration of vincristine. Vincristine had no significant effects on platelet aggregation in response to collagen, low or high doses of platelet-activating factor, and a high dose of ADP. The maximal degree of platelet aggregation attained in response to a low dose of ADP was not significantly affected by prior administration of vincristine. The maximal rate of platelet aggregation induced by a low dose of ADP after vincristine administration, however, was significantly (P < 0.05) lower than the rate of aggregation induced by a similar dose of ADP in the previous control study. Vincristine had no significant effects on clot retraction and bleeding time. Prednisone did not significantly affect platelet numbers and function, and did not modify vincristine's effects on the same variables.

Paired metacarpi obtained at necropsy from 100 horses ranging in age from term fetus to 35 years were examined to estimate the prevalence and sites of metacarpal fusion. Metacarpal fusion was seen in 192 of 200 metacarpi, and 78% of all horses 2 years or older had 2 or more fusions. Fusion of the second metacarpal bone to the third metacarpal bone was significantly (P < 0.001) more common than was fusion of the fourth to the third metacarpal bone. Fusions appeared for the most part in pairs and were bilaterally symmetric. Rooney-Prickett type-A carpometacarpal joint configurations (in which there is no measurable articulation between the third carpal and second metacarpal bones) were rare in this population, and Rooney-Prickett type-B configurations (in which there is a measurable articulation between the third carpal and second metacarpal bones) were observed in 98.5% of metacarpi. Medial metacarpal fusion was positively correlated with age, occupation, and proportion of the proximal projection of the carpometacarpal distal joint surface that was taken by the second metacarpal bone. Lateral metacarpal fusion was positively correlated with age and the proportion of the proximal projection of the carpometacarpal distal joint surface taken by the fourth metacarpal bone. Horses in performance careers (racing, race training, or show ring occupations) had an earlier development of the first 2 fusions than did horses in other or unknown occupations; development of the third and fourth fusions were not significantly different between occupation groups. The rate of metacarpal fusion per horse-year appeared to be at least 10 times higher than a clinically evident rate.

A radioimmunoassay test for tetracyclines (Charm II) was compared with high-pressure liquid chromatography (HPLC) for detection of oxytetracycline (OTC) residues in milk samples from individual lactating cows. Oxytetracycline was administered by 1 of 3 routes (IV, IM, or intrauterine) to 21 lactating dairy cows. A total of 292 duplicate milk samples were collected from milkings before and through 156 hours after OTC administration. Concentration of OTC in these samples was determined by use of the Charm II test and an HPLC method with a lower limit of quantitation, approximately 2 ng of OTC/ml. Samples were also classified with respect to presence of OTC residues relative to the FDA safe concentration (less than or equal to 30 ng/ml), using the Charm II (by control point determination) and HPLC methods. There was a significant (P less than or equal to 0.05) difference between test methods in classification of milk samples with respect to presence or absence of OTC at the FDA safe concentration. A total of 48 of the 292 test results (16.4%) did not agree. Using the HPLC test results as the standard with which Charm II test results were compared, 47 false presumptive-violative test results and 1 false presumptive-nonviolative Charm II test result (a sample containing 31 ng of OTC/ml, as evaluated by HPLC) were obtained. The samples with false presumptive-violative Charm II results contained (less than or equal to 30 ng of OTC/ml, as evaluated by HPLC. In some respects, the Charm II test performed appropriately as a screening test to detect OTC residues in milk samples from individual cows. However, the tendency for the test to yield presumptive-violative test results at OTC concentrations lower than the FDA safe concentration (as evaluated by HPLC), suggests that caution should be exercised in using the test as the sole basis on which a decision is made to reject milk. As indicated by the manufacturer, presumptive-violative Charm II test results should be confirmed by additional testing Although not specifically evaluated, the tendency for misclassification of milk samples as presumptive-violative by the Charm II test may or may not occur in commingled milk, compared with milk samples from individual cows.

Six horses were subjected to 3 hours of low-flow ischemia and 3 hours of reperfusion of the large colon. After induction of anesthesia, the large colon was exteriorized through a ventral midline celiotomy. Colonic blood flow was measured continuously, using Doppler ultrasonic flow probes placed on the colonic arteries supplying the dorsal and ventral colons and was allowed to stabilize for 15 to 30 minutes after instrumentation. Low-flow ischemia was induced by reducing colonic arterial blood flow to 20% of baseline (BL) flow. Colonic mucosal, seromuscular, and full-thickness blood flow were determined on a tissue-weight basis by injecting colored microspheres proximally into the colonic artery supplying the ventral colon. Reference blood samples were obtained at a known flow rate from the colonic artery and vein at a site more distal to the site of injection. Left ventral colon biopsy specimens were harvested at BL, 3 hours of ischemia, and 15 minutes of reperfusion. Blood and tissue samples were digested and filtered to collect the microspheres, and dimethylformamide was added to release the colored dyes. Dye concentration in blood and tissue samples was measured by use of spectrophotometry, and tissue-blood flow was calculated. Data were analyzed, using two-way ANOVA for repeated measures; statistical significance was set at P < 0.05. Doppler blood flow decreased to approximately 20% of BL, whereas microsphere blood flow ranged between 13.7 and 15.5% of BL at 3 hours of ischemia. Doppler-determined blood flow increased immediately on restoration of blood flow, reached 183% of BL at 15 minutes of reperfusion, and remained at or above BL throughout 3 hours of reperfusion. This reactive hyperemia was also detected, using the colored microspheres; blood flow increased to 242 and 327% of BL at 15 minutes of reperfusion in the mucosal and seromuscular layers, respectively. Mucosal blood flow was not different from seromuscular blood flow at any time, indicating relatively equal distribution of blood flow between these 2 layers. As determined from the venous reference samples, there was no evidence of arteriovenous anastomoses.

The effects of short-term restraint stress, by means of snaring, on plasma concentrations of catecholamines, beta-endorphin, and cortisol were studied in 6 gilts. A catheter was inserted into the jugular vein, and 2 blood samples were collected before onset of stress. Thereafter, a hog snare was applied, and blood samples were collected at 0.5, 2, and 3.5 minutes after the start of snaring. Plasma epinephrine and norepinephrine concentrations increased (P < 0.001) within 0.5 minute after start of restraint and decreased thereafter. Plasma concentration of beta-endorphin increased (P < 0.05) within 2 minutes after start of restraint, whereas that of cortisol increased (P < 0.05) 3.5 minutes after start of restraint. Taken together, short-term stress, such as snaring, may increase the plasma concentration of catecholamines, beta-endorphin, and cortisol in pigs.

We quantified the effect of passive immune status on pre- and postweaning health and growth performance of calves raised in a beef production environment. Blood samples were collected at postpartum hour 24 from 263 crossbred calves for determination of plasma protein (PP) and serum IgG concentrations. Serum IgG concentration was classified as adequate (> 1,600 mg/dl), marginal (800 to 1,600 mg/dl), or inadequate (< 800 mg/dl). Plasma protein concentration was classified as adequate (greater than or equal to 4.8 g/dl) or inadequate (< 4.8 g/dl). Morbidity and mortality events in the study population were monitored from birth to weaning, and after weaning throughout the feeding period. The lowest concentrations of serum IgG and PP were observed among calves that experienced morbidity or mortality prior to weaning. Calves that experienced morbidity in the feedlot had lower 24-hour PP values, but had IgG concentration similar to that in calves that were not observed to be ill during the feeding period. Calves classified as having inadequate IgG concentration were at greater risk of preweaning mortality (odds ratio [OR] = 5.4), neonatal morbidity (OR = 6.4), and preweaning morbidity (OR = 3.2), compared with calves classified as having adequate IgG concentration at 24 hours. Calves classified as having inadequate PP concentration at 24 hours had a greater risk of morbidity (OR = 3.0) and respiratory tract morbidity (OR = 3.1) while in the feedlot, compared with calves classified as having adequate PP concentration. The effects of 24-hour passive immune status on calf growth were indirect through effects on morbidity outcomes. Morbidity during the first 28 days of life was associated with a 16-kg lower expected weaning weight. Respiratory morbidity in the feedlot resulted in a 0.04-kg lower expected mean daily gain. Thus, passive immune status at postpartum hour 24 was an important determinant of health before and after weaning, and was indirectly associated with calf growth during the same periods.

Ocular examination, including gonioscopy and ultrasonographic biometry (biological measurement), was performed in healthy, purebred Samoyeds. When the dogs were subclassified according to the degree of kinship with cases of primary angle-closure glaucoma, it was found that the opening of the ciliary cleft was wider in dogs less related to dogs with primary angle-closure glaucoma than in more closely related dogs. Furthermore, multivariate analysis of the material indicated that parentage of a dog has a substantial effect on the intraocular distances studied in this investigation (relative depth of the opening of the ciliary cleft, relative corneal thickness, relative anterior chamber depth, relative lens thickness, and relative length of the vitreous body). Finally, heritability of the relative depth of the opening of the ciliary cleft was estimated at 56% and that of the common environmental factor at 19%. The results are indicative of a hereditary component in primary angle-closure glaucoma in the Samoyed breed.

In a 5-year prospective study, computed tomographic (CT) morphometry of the lumbosacral vertebral canal was performed on 42 large-breed dogs (21 controls and 21 dogs with lumbosacral stenosis). Dogs were allotted to 4 groups. Group 1 (n = 13) consisted of cadaver specimens obtained from dogs that died or were euthanatized for reasons unrelated to the spine; group 2 (n = 8) consisted of live dogs with no history of clinical signs related to the spine and with normal neurologic examination findings; group 3 (n = 10) consisted of dogs with surgically confirmed lumbosacral stenosis; and group 4 (n = 11) consisted of dogs with suspected lumbosacral stenosis that were managed conservatively. The CT scans were performed, using 5-mm contiguous slices obtained perpendicular to the vertebral canal, from the midbody of the 5th lumbar vertebra through the caudal endplate of the sacrum (L5-S3). Lumbosacral lordosis was minimized in all dogs by positioning them in dorsal recumbency with the hind limbs flexed. A tuberculin syringe calibration phantom was placed within the scanning field of view, parallel to the axis of the spine. In each dog, 11 CT slice locations within the lumbosacral spine were evaluated. At each slice location, sagittal plane diameter, dorsal plane diameter, and transverse area of the vertebral canal, vertebral body, and calibration phantom were measured, using the CT computer's software programs for distance and area calculation. Window/level settings were constant, and all measurements were made by the same operator (JCJ). Accuracy of calibration phantom CT measurements was 100% for sagittal and dorsal plane diameter and was 85% for transverse area. In control dogs (groups 1 and 2), vertebral canal dimensions were significantly (r greater than or equal to 0.50, P less than or equal to 0.0001) correlated with vertebral body dimensions, but not with dog weight or age. There were no significant differences between group 1 vs group 2, and group 3 vs group 4 for all absolute vertebral canal dimensions and for 5 ratios of vertebral canal to correlated vertebral body dimensions (general linear model for ANOVA). Pooled control dogs (n = 21) and those with lumbosacral stenosis (n = 21) were compared, and significant differences were not identified for absolute canal dimensions. Significant differences between control dogs and those with lumbosacral stenosis were identified in the ratios of vertebral canal transverse area to vertebral body sagittal diameter (P less than or equal to 0.01) and vertebral canal transverse area to vertebral body transverse area (P less than or equal to 0.001). For both these ratios, analysis by slice location identified significant differences (P < 0.05) between pooled groups at the caudal pedicles of L5 and L6. For the ratio of transverse canal area to sagittal vertebral body diameter, differences (P less than or equal to 0.05) also were found at the cranial pedicle of L7. These results indicate that: CT is an accurate method for performing morphometry of the canine lumbosacral spine; vertebral canal dimensions can be corrected for differences in dog size by calculating ratios of vertebral canal to vertebral body dimensions; statistical comparisons, using such corrected vertebral canal dimensions, may reveal differences not evident when absolute vertebral canal dimensions are used; and corrected transverse area of the vertebral canal differs in large-breed dogs with lumbosacral stenosis vs normal controls. Morphometric differences identified at more than 1 vertebral level support a theory that multilevel congenital or developmental stenosis of the lumbosacral vertebral canal may be a predisposing or contributing factor in large-breed dogs with acquired lumbosacral stenosis.

Objective--To determine the relative role of endogenous prostaglandin F2 alpha (PGF2 alpha) secretion in cloprostenol-induced abortion in mares that no longer require luteal progesterone secretion for maintenance of pregnancy, and to evaluate the ability of a prostaglandin cyclooxygenase inhibitor (flunixin meglumine) to prevent cloprostenol-induced abortion. Design--The effect of flunixin meglumine on PGF2 alpha secretion and outcome of pregnancy was compared between mares treated with cloprostenol only and mares treated with cloprostenol plus flunixin meglumine. Animals--Five pregnant mares, aged 4 to 15 years, of light-horse type. Procedure--Cloprostenol (250 micrograms) was administered at 24-hour intervals to 5 pregnant mares. Flunixin meglumine (500 mg, IV) was administered at 8-hour intervals starting 15 minutes before the first cloprostenol administration. Hourly blood samples were analyzed for 15-ketodihydro-PGF2 alpha, progesterone, and estrogen concentrations. Previously reported data on cloprostenol-induced abortion in 6 pregnant mares treated daily with cloprostenol only were used as historic controls. Results--The mean (+/- SEM) interval from first cloprostenol administration to fetal expulsion 56.4 (+/- 13.7) hours and number of cloprostenol administrations 3.2 (+/- 0.6) in the 5 flunixin meglumine-treated mares were not significantly different, compared with values for 6 pregnant mares treated daily with cloprostenol only, 48.6 (+/- 5.6) hours and 2.8 (+/- 0.2) cloprostenol administrations. Flunixin meglumine did not inhibit endogenous PGF2 alpha secretion. Prostaglandin F2 alpha secretion rates on the day before and day of fetal expulsion were similar in both groups. Conclusion--Flunixin meglumine at a dosage of 500 mg/animal, administered IV every 8 hours, is ineffective in modulating uterine PGF2 alpha secretion during cloprostenol-induced abortion. Clinical Relevance--Flunixin meglumine is ineffective in the modulation of prostaglandin-induced uterine PGF2 alpha secretion and, therefore, does not offer a viable alternative for the prevention of abortion in mares at risk of abortion because of systemic illness.

Neospora caninum-induced abortion is a major production problem in the daily cattle industry in the United States and worldwide. Abortions attributable to naturally acquired N caninum infection also have been observed in pygmy goats. We studied experimentally induced infections with N caninum in pregnant pygmy does to determine whether abortions attributable to N caninum infection would occur after inoculation. Seven pregnant pygmy does (1 control doe and 6 inoculated with N caninum) were studied. The control doe remained clinically normal throughout the study and delivered 2 healthy kids. Abortion, fetal death, and stillbirths were observed in some pregnant does inoculated with N caninum. Two pregnant pygmy does inoculated with N caninum early in gestation (day 51) had fetuses that died and were aborted, or died and were reabsorbed. Neospora caninum tachyzoites and lesions were observed in the brain, spinal cord, and heart of aborted fetuses; parasites also were isolated from the placenta. Four additional pregnant pygmy does (2 inoculated at mid-gestation [day 85], and 2 at late gestation [day 127]) did not abort after inoculation. However, 1 doe inoculated during mid-gestation delivered a stillborn fetus that had died about 1 week prior to parturition. This kid was congenitally infected with N caninum. Neospora caninum was isolated from the placentas of all inoculated does examined. Neonatal neosporosis was not observed in live-born kids, nor were stages of N caninum isolated from any live-born kid. Does did not undergo abortion or have congenitally infected kids when they were rebred and evaluated for neosporosis.
