Accuracy of noninvasive, single-plane fluoroscopic analysis for measurement of three-dimensional femorotibial joint poses in dogs treated by tibial plateau leveling osteotomy

Objective- To compare accuracy of a noninvasive single-plane fluoroscopic analysis technique with radiostereometric analysis (RSA) for determining 3-D femorotibial poses in a canine cadaver stifle joint treated by tibial-plateau-leveling osteotomy (TPLO). Sample- Left pelvic limb from a 25-kg adult mixed-breed dog. Procedures- A CT scan of the left pelvic limb was performed. The left cranial cruciate ligament was transected, and a TPLO was performed. Radiopaque beads were implanted into the left femur and tibia, and the CT scan was repeated. Orthogonal fluoroscopic images of the left stifle joint were acquired at 5 stifle joint flexion angles ranging from 110° to 150° to simulate a gait cycle; 5 gait cycles were completed. Joint poses were calculated from the biplanar images by use of a digitally modified RSA and were compared with measurements obtained by use of hybrid implant-bone models matched to lateral-view fluoroscopic images. Single-plane measurements were performed by 2 observers and repeated 3 times by the primary observer. Results- Mean absolute differences between results of the single-plane fluoroscopic analysis and modified RSA were 0.34, 1.05, and 0.48 mm for craniocaudal, proximodistal, and mediolateral translations, respectively, and 0.56°, 0.85°, and 1.08° for flexion-extension, abduction-adduction, and internal-external rotations, respectively. Intraobserver and interobserver mean SDs did not exceed 0.59 mm for all translations and 0.93° for all rotations. Conclusions and Clinical Relevance- Results suggested that single-plane fluoroscopic analysis by use of hybrid implant-bone models may be a valid, noninvasive technique for accurately measuring 3-D femorotibial poses in dogs treated with TPLO.

Objective- To evaluate the effects of tylosin on C-reactive protein concentration, carriage of Salmonella enterica, and antimicrobial resistance genes in commercial pigs. Animals-120 pigs on 2 commercial farms. Procedures- A cohort of sixty 10-week-old pigs in 4 pens/farm (15 pigs/pen) was randomly selected. Equal numbers of pigs were given feed containing tylosin (40 μg/g of feed) for 0, 6, or 12 weeks. C-reactive protein concentrations were measured, microbial culture for S enterica in feces was performed, and antimicrobial resistance genes in feces were quantified. Results- No significant associations were detected between C-reactive protein concentration or S enterica status and tylosin treatment. During the 12 weeks of tylosin administration, increased levels of 6 antimicrobial resistance genes did not occur. Conclusions and Clinical Relevance-Treatment of pigs with tylosin did not affect C-reactive protein concentration or reduce carriage or load of S enterica. There was no evidence that pigs receiving tylosin had increased carriage of the 6 antimicrobial resistance genes measured. Impact for Human Medicine-S enterica is a public health concern. Use of the antimicrobial growth promoter tylosin did not pose a public health risk by means of increased carriage of S enterica.

Objective- To assess the accuracy of an ultrasound velocity dilution cardiac output (UDCO) method, compared with that of the lithium dilution cardiac output (LiDCO) method, for determination of cardiac output (CO) in juvenile horses with experimentally induced hypovolemia. Animals- 12 anesthetized 2- to 6-month-old horses. Procedures- For each anesthetized horse, CO was determined by the LiDCO and UDCO methods prior to any intervention (baseline state), after withdrawal of approximately 40% of the horse's blood volume (low CO state), after maintenance of hypovolemia and infusion of norepinephrine until mean arterial blood pressure was equal to baseline value (high CO state), and after further infusion of norepinephrine and back-transfusion of withdrawn blood (posttransfusion state). For each of the 4 hemodynamic situations, CO and calculated cardiac index (CI) values were obtained by each method in duplicate (8 pairs of measurements/horse); mean values for each horse and overall mean values across all horses were calculated. Agreement between CI determined by each method (96 paired values) was assessed by Bland-Altman analysis. Results- For the UDCO method–derived CI measurements among the 12 horses, mean ± SD bias was −4 ± 11.3 mL/kg/min (95% limits of agreement, −26.1 to 18.2 mL/kg/min) and mean relative bias was −10.4 ± 21.5% (95% limits of agreement, −52.6% to 31.8%). Conclusions and Clinical Relevance- Results indicated that, compared with the LiDCO method, the UDCO method has acceptable clinical usefulness for determination of CO in foals.

Objective- To evaluate physical methods for inducing death during the slaughter of American alligators (Alligator mississippiensis). Animals- 24 captive hatched-and-reared American alligators. Procedures- Baseline electroencephalograms (EEGs) were obtained for awake and anesthetized alligators. Corneal reflex, spontaneous blinking, and EEGs were evaluated after severance of the spinal cord, severance of the spinal cord followed by pithing of the brain, application of a penetrating captive bolt, or application of a nonpenetrating captive bolt (6 alligators/group). Results- Overall, alligators subjected to spinal cord severance alone differed from those subjected to the other techniques. Spinal cord severance alone resulted in postprocedure EEG power values greater than those in anesthetized alligators, whereas the postprocedure EEG power values were isoelectric for the other 3 techniques. Corneal reflex and spontaneous blinking were absent in all alligators immediately after application of a penetrating or nonpenetrating captive bolt. One of 6 alligators had a corneal reflex up to 1 minute after pithing, but all others within that group had immediate cessation of reflexes after pithing. Mean time to loss of spontaneous blinking and corneal reflex for alligators subjected to spinal cord severance alone was 18 minutes (range, 2 to 37 minutes) and 54 minutes (range, 34 to 99 minutes), respectively. Conclusions and Clinical Relevance- Spinal cord severance followed by pithing of the brain and application of a penetrating or nonpenetrating captive bolt appeared to be humane and effective techniques for inducing death in American alligators, whereas spinal cord severance alone was not found to be an appropriate method.

Objective—To evaluate the effects of a dexmedetomidine constant rate infusion (CRI) and atropine on changes in global perfusion variables induced by hemorrhage and volume replacement (VR) in isoflurane-anesthetized dogs. Animals—8 adult dogs. Procedures—Each dog was anesthetized twice, with a 2-week interval between anesthetic sessions. Anesthesia was maintained with 1.3 times the minimum alveolar concentration of isoflurane with and without dexmedetomidine (1.6 μg/kg, IV bolus, followed by 2 μg/kg/h, CRI). Dogs were mechanically ventilated and received an atracurium neuromuscular blockade during both sessions. During anesthesia with isoflurane and dexmedetomidine, atropine was administered 30 minutes before baseline measurements were obtained. After baseline data were recorded, 30% of the total blood volume was progressively withdrawn and VR was achieved with an equal proportion of autologous blood. Results—Following hemorrhage, cardiac index, oxygen delivery index, and mixed-venous oxygen saturation were significantly decreased and the oxygen extraction ratio was significantly increased from baseline. The anaerobic threshold was not achieved during either anesthetic session. When dogs were anesthetized with isoflurane and dexmedetomidine, they had a significantly lower heart rate, cardiac index, and mixed-venous oxygen saturation during VR than they did when anesthetized with isoflurane alone. Plasma lactate concentration, mixed venous-to-arterial carbon dioxide difference, base excess, and anion gap were unaltered by hemorrhage and VR and did not differ between anesthetic sessions. Conclusions and Clinical Relevance—Results indicated that the use of a dexmedetomidine CRI combined with atropine in isoflurane-anesthetized dogs that underwent volume-controlled hemorrhage followed by VR did not compromise global perfusion sufficiently to result in anaerobic metabolism.

Objective—To construct and optimize a fiducial marker suitable for both CT and MRI. Sample—Fiducial markers containing serial dilutions of iopamidol mixed with water. Procedures—IV tubing sets were infused with serial dilutions (0% to 100%; increments of 10%) of iopamidol. Tubing ends were sealed; additional seals were added to create an equilateral triangle. A reference point was created by placing a crimp in 1 side. Markers were fixed to a gelatin soft tissue–attenuating phantom and evaluated by use of CT and MRI. For CT, simple linear regression analysis was used to assess the relationship between the percentage of marker contrast medium and quantitative variables, including marker attenuation, attenuation changes in the phantom, and beam-hardening artifact length. A subjective grading scheme for artifact creation on CT images and marker visibility on MRI images was used. Measurements were obtained by investigators who were unaware of the contents of each marker. Results—Percentage of contrast medium in each marker was strongly correlated with marker attenuation (r2 = 0.96), artifact length (r2 = 0.765), and mean attenuation changes within the phantom (r2 = 0.826) for CT. Subjective CT scores indicated that concentrations of contrast medium > 50% resulted in excessive artifacts. Markers with concentrations of iopamidol > 50% had poor subjective MRI visibility scores. No artifacts were seen on MRI. Conclusions and Clinical Relevance—A marker containing a 10% solution of iodinated contrast medium mixed with water provided ideal contrast for both CT and MRI.

Objective—To develop and assess the short-term feasibility, maintenance, and complications associated with percutaneous endoscopic gastrostomy (PEG) tube placement in standing horses. Animals—6 adult horses. Procedures—Feasibility of the technique was evaluated in 2 horses. In each of 4 other horses, a PEG tube was maintained for 14 days and used to provide fluid requirements during the latter 7 days, before removal. Following air inflation of the stomach, each PEG tube was placed via a left intercostal approach; proper tube location was ascertained by percutaneous ultrasonography and gastroscopy. The horses underwent physical examinations, CBCs, and peritoneal fluid analyses before and at intervals after tube placement. Seven days after tube removal, horses were euthanized and necropsied. Results—Placement of a PEG tube was feasible in all 6 horses. The 4 horses assessed long term tolerated water administration through the PEG tube and remained clinically stable throughout the 21-day experiment. However, during the period PEG tubes were in place, significant increases in some peritoneal and hematologic variables were detected. Postmortem evaluation revealed localized peritonitis in 1 horse and body wall inflammation along the PEG tube tracks in 3 additional horses. Conclusions and Clinical Relevance—Placement and maintenance of a PEG tube were tolerated well by the study horses, although peritoneal and systemic inflammation were detectable. Fluid requirements were adequately met with this technique, which could provide an alternative method for managing chronically dysphagic horses. Nevertheless, further research is warranted to evaluate the feasibility of enteral feeding by use of this approach in horses.

Objective-To determine changes in Canine Brief Pain Inventory scores for dogs with osteoarthritis after administration of a monoclonal antibody (mAb) against nerve growth factor (NGF) that was modified by use of a proprietary process for administration to dogs. Animals- 11 adult dogs. Procedures- Dogs received the anti-NGF mAb (0.2 mg/kg, IV) at various evaluation times during the study period; at other evaluation times, dogs received an equivalent volume of PBS solution IV. Owners determined Canine Brief Pain Inventory pain severity (PS) and pain interference (PI) scores immediately before (baseline) and 2, 4, and 6 weeks after administration of the anti-NGF mAb; owners were unaware of the evaluation time at which the mAb had been administered. Results- Compared with baseline PS scores (median, 4.75; range, 0.75 to 8.5), dogs had significantly lower PS scores 2 weeks (median, 3; range, 1 to 5.5) and 4 weeks (median, 2.25; range, 0.25 to 7.25) after administration of anti-NGF mAb. Compared with baseline PI scores (median, 5.33; range, 1.17 to 9.33), dogs had significantly lower PI scores 2 weeks (median, 3; range, 0.67 to 6.83) and 4 weeks (median, 3.33; range, 0.67 to 6.67) after administration of anti-NGF mAb. The PS and PI scores 6 weeks after mAb administration were lower than baseline scores, although values were not significantly different. Conclusions and Clinical Relevance- Results of this study suggested the evaluated anti-NGF mAb decreased PS and PI scores for 4 weeks after administration. This treatment may be effective for alleviation of signs of pain in dogs with osteoarthritis for up to 4 weeks.

Objective—To evaluate the ability of small interfering RNAs (siRNAs) to inhibit in vitro viral replication and gene expression of feline coronavirus (FCoV). Sample—Cell cultures of Crandell-Rees feline kidney cells. Procedures—5 synthetic siRNAs that each targeted a different region of the FCoV genome were tested individually and in various combinations for their antiviral effects against 2 strains of FCoV (feline infectious peritonitis virus WSU 79-1146 and feline enteric coronavirus WSU 79-1683) in cell cultures. Tested combinations targeted the FCoV leader and 3′ untranslated region, FCoV leader region and nucleocapsid gene, and FCoV leader region, 3′ untranslated region, and nucleocapsid gene. For each test condition, assessments included relative quantification of the inhibition of intracellular viral genomic RNA synthesis by means of real-time, reverse-transcription PCR analysis; flow cytometric evaluation of the reduction of viral protein expression in infected cells; and assessment of virus replication inhibition via titration of extracellular virus with a TCID50 infectivity assay. Results—The 5 siRNAs had variable inhibitory effects on FCoV when used singly. Combinations of siRNAs that targeted different regions of the viral genome resulted in more effective viral inhibition than did individual siRNAs that targeted a single gene. The tested siRNA combinations resulted in approximately 95% reduction in viral replication (based on virus titration results), compared with findings in negative control, nontargeting siRNA–treated, FCoV-infected cells. Conclusions and Clinical Relevance—In vitro replication of FCoV was specifically inhibited by siRNAs that targeted coding and noncoding regions of the viral genome, suggesting a potential therapeutic application of RNA interference in treatment of feline infectious peritonitis.

Objective—To determine the effects of UVB radiation produced by artificial lights on serum 25-hydroxyvitamin D concentrations in domestic rabbits (Oryctolagus cuniculi) Animals—9 juvenile domestic rabbits. Procedures—After an acclimation period, rabbits were anesthetized with isoflurane, and an initial blood sample was collected for determination of serum 25-hydroxyvitamin D concentration. Rabbits were randomly assigned to receive 12-hour exposure to UVB radiation produced by 2 compact fluorescent lights daily (n = 5) or no UVB supplementation (4) commencing on day 1. The UVB radiation emitted into the cage was measured at 9 points approximately 34 cm from the surface of the UVB light sources (representing the position of the rabbits in the cage) after 10 hours of exposure on days 1, 8, and 14. On day 14, another blood sample was collected from anesthetized rabbits for determination of serum 25-hydroxyvitamin D concentration. Results—The UVB radiation level was 8.3 to 58.1 μW/cm2 for the exposed rabbits and consistently < 0.001 μW/cm2 for the control rabbits. Mean ± SD serum 25-hydroxyvitamin D concentrations in the rabbits that were or were not provided supplemental UVB radiation for 14 days differed significantly (66.4 ± 14.3 nmol/L and 31.7 ± 9.9 nmol/L, respectively). Conclusions and Clinical Relevance—Exposure to UVB radiation produced by artificial light significantly increased serum 25-hydroxyvitamin D concentration in juvenile rabbits. Because vitamin D is an essential hormone in vertebrates, these findings suggested that the provision of supplemental UVB radiation to captive rabbits may be important.
