BACKGROUND: Brucellosis is one of the most important zoonosis that is prevalent among human and animal. Today, a large percentage of animal and human population suffer from its side effects. OBJECTIVES: The purpose of the present study was to estimate the prevalence of anti-Brucella antibodies in flock- and animal-level in districts of south of Kerman province. METHODS: In this cross-sectional study, 300 herds of 7 districts in the area were selected randomly; 10 samples of sheep and goats in each flock were randomly selected. Out of 3000 samples, 2952 samples were examined using Rose-Bengal test; Wright and 2-ME tests were done on positive samples. Descriptive statistics and logistic regression in Stata 11.2 were used to analyze the data. RESULTS: The seroprevalence of anti-Brucella antibodies in animal- and flock-level was 6.4 and 25.3 percent, respectively. The chance of being infected in sheep was 2.12 times of goats. CONCLUSIONS: The prevalence of Brucella was considerably high in animal- and herd-level in this area. It is necessary to empower Iran Veterinary Organization in financial aspects to control this infection.

BACKGROUND: Adiponectin is one of the most important adipocytokines that regulate male and female fertility via AdipoR1 and AdipoR2 receptors. Recently expression of adiponectin system and its negative regulatory role on hypothalamus-pituitary axis have been confirmed. Objectives: No information is available about the expression pattern of adiponectin and its receptors in hypothalamus-pituitary axis in domestic animals. Here for the first time, we studied hypothalamus-pituitary adiponectin system gene expression in different stages of bovine estrous cycle. Methods: Anterior pituitary and hypothalamus were collected from Holstein cow at the local abattoir. The estrous cycle was classified to four phases (proestrous, metstrous, early luteal and late luteal) based on macroscopic examination of ovaries and uteri. Gene expression analysis of adiponectin and its receptors was done using quantitative real time PCR (qPCR Probe MasteKit) and according to the comparative 2-ΔΔCt method. E2 and P4 levels were measured using ELISA method. Results: Our results demonstrated that adiponectin and its two receptors were expressed in pituitary and hypothalamus of cyclic cow. Maximal expression of adiponectin was observed in early luteal phase, while it was expressed at minimal level during the proestrous stage. We observed no significant changes in the expression of AdipoR1 in both tissues at different stages of estrous cycle. The highest expression of AdipoRII in both tissues was detected during the proestrous stage, while it expressed at minimal level during the late luteal phase. E2 and P4 had respectively negative and positive correlations with adiponectin expression levels in hypothalamus and pituitary. Conclusions: Based on our results that demonstrated adiponectin was minimally expressed at proestrous stage and other data about the negative action of adiponectin on LH secretion from pituitary, we concluded that adiponectin may has role in the hormonal function of this axis during the estrous cycle.

BACKGROUND: Fibroblasts are one of the important cells in wound healing. These cells create a proper bed for keratinocytes migration and wound contraction.Wound healing in distal limb of horses has complications, such as formation of exuberant granulation tissue (EGT). The main factor in this problem is overgrowing of fibroblasts. ObjectiveS: The purpose of the present study was to compare fibroblast growth curve in isolated skin from horses’ neck and distal limb. Methods: 5 horses with normal hematological and clinical signs were selected. Two samples of full thickness of skin were taken from the neck and lateral metacarpal region of each horse asseptically. Then the samples were washed with PBS minced and placed in ventilated flask 25 cm2. After attaching samples to flask, 5 ml culture medium(RPMI-1640 with 10% FBS) were added and the flask was placed in an incubator at 37°c in 5% CO2. After leaving a sufficient number of cells from tissues adhered to the bottom of the flask, the cells were passaged to a new ventilated flask. After growth and proliferation of cells, they were passaged again and a suspension of cells in culture medium (10000 cells/ml) was maked. To each cell of a 24-well plate, one ml of this suspension was added. After 48 hour, cells of 3 well were detached with tripsin daily, counted and viability determinted within 8 days. Results: There was no significant difference between viable cells number but there was significant difference in viability percent of cells in neck and distal limb. The mean of population doubling time (PDT) for fibroblasts of neck is 31.73 hours and for fibroblasts of distal limb is 26.4 hours. This difference was not significant. ConclusionS: With regard to different viability percentage, it seems that the appoptosis in fibroblasts of neck skin is more regular than distal limb skin.

BACKGROUND: Probiotics, in form of microbial supplements, are known to be a suitable alternative for antibiotics and can affect the health indicators of host. Objectives: The present study was conducted to assess the combined effects of dietary autochthonous Saccharomyces cerevisiae and Aspergillus niger on haematological and serumbiochemicalparameters of beluga sturgeon (Huso huso) juveniles. Methods: This study was based on a completely randomized design with 4 treatments and 3 replicates on beluga juveniles with average weight of (mean ±SE) 31.8±2.81g. Beluga Juveniles were divided randomly into 12 fiber glassy tanks with density of 30 fish per tank and were fed with diet contain dietary probiotic with density of 2×106 (Cells/g) for the first treatment, 4×106 (Cells/g) for the second treatment, 6×106 (Cells/g) for the third treatment and basal diet without probiotic for the control group for 8 weeks. Results: Diet supplementing with concentration of 6×106 (Cells/g), significantly improved serum biochemical parameters (p<0.05), however hematological parameters were affected by supplemented diet with probiotics that showed no significant difference in comparison with the control group (p>0.05). Also results indicate that growth factors were improved in experimental treatments in comparison with the control group. Conclusions: The results showed that the use of combination of these species with studied concentrations can improve the performance of some biochemical parameters such metabolites factors, immune, enzymes and serum electrolytes of belugajuveniles. It is recommended that the concentration of A. niger and S. cerevisiae, used for third treatment be used as an immune stimulator for beluga juveniles.

BACKGROUND: Toxocara canis is a zoonotic disease that commonly infects canids. Mammals and birds are sometimes infected with this disease as paratenic hosts. It can also cause accidental infection in humans. The increase in the number of stray dogs, the expansion of urban gardens, and the proximity of dogs to humans increase the risk of human infection with Toxocara canis.OBJECTIVES: This study aims to determine the prevalence of Toxocara canis infection in dogs and foxes in Zanjan province, Iran.METHODS: A total of 484 fecal samples of stray dogs (n=355), rescue dogs (n=49), guard dogs (n=50), and foxes (n=30) in Zanjan were randomly collected from June 2021 to February 2022. The microscopic examination was done following formalin-ethyl acetate sedimentation procedures. Finally, the PCR method was used to confirm the presence of Toxocara canis in positive samples.RESULTS: Microscopic study revealed that, out of 484 samples, 21 (4.3%) were positive for Toxocara/ Toxascaris eggs. Between these positive samples of dogs and foxes, only 6 samples from dog feces were confirmed as a Toxocara canis infection by the PCR method.CONCLUSIONS: There is an increase in the prevalence of Toxocara canis infection in stray dogs in Zanjan, Iran. Given the presence of dogs in parks and residential areas, there is a risk of human infection with Toxocara canis, emphasizing the importance of adhering to treatment and prevention protocols in dealing with stray dogs.

BACKGROUND: Pre-purchase radiographic examinations of horses are very important for the diagnosis of possible disorders.OBJECTIVES: This study aims to determine the variation and frequency of common disorders of the tarsus in pre-purchase radiographic examinations of horses and assess the effect of age, sex, and type of hind limb (right or left) on the occurrence of these disorders.METHODS: In this study, we used the radiographs taken from the tarsal joints of the right and left hind limbs in 110 horses for a two-year period which were requested for pre-purchase radiographic examinations. Among these radiographs, the cases that included the standard position of the tarsal joint were evaluated for the existing disorders and the effect of age, sex, and type of involved hind limb.RESULTS: The most frequent disorders were osteoarthrosis (32.27 %), osteochondritis dissecans (5.91 %), and soft tissue swelling (1.36 %). The least common disorders were calcification, desmopathy, and tenosynovitis, each with a prevalence of 0.45%. Also, most of the tarsal ​​joint disorders had a mild degree of severity (80 %). The prevalence of disorders was 100 % for horses aged <4 years (4 out of 4) and 49.1 % for horses aged >4 years (52 out of 106). No significant difference was observed in terms of gender (P=0.65) and type of involved hind limb (P=0.17).CONCLUSIONS: Considering the prevalence of tarsal joint disorders in horses, pre-purchase radiographic examination can be very important for predicting the future health and performance of the horse. Therefore, radiographic evaluation of the tarsal joint should always be a part of pre-purchase examinations in horses.

BACKGROUND: In free stalls, factors related to the surface and dimensions of the stall affect how the cows rest and comfort. The brisket board and the neck rail are the most controversial parts of the free stall in Iran's dairy farms, that can affect the stability of the stall and its lifespan, while improper use of these structures has led to significant discomfort for cows, causing substantial issues including lameness and hock, knee and withers lesions.OBJECTIVES: This study aims to investigate Brisket boards and neck rails usage and measures in freestall barns and assess its possible impact on some comfort indices in dairy cows.METHODS: Nine dairy farms with over 100 milk cows and freestall barns were selected using the Dairy Farmers of Canada protocols by a convenience sampling method. Horizontal, vertical, and diagonal distances of the neck rail, the presence or absence of brisket boards, and the brisket board height from the bedding were measured. The locomotion score based on a five-point scale as well as hygiene, knees, hocks, and withers scores were recorded. The correlation was evaluated using the Spearman correlation test and Pearson’s correlation test.RESULTS: In 68.3 % of the freestall barns, the brisket boards were at the bedding level or were not used at all; however, the mean brisket board height (11.2±10.8) was not significantly different from the standard height value of 10 cm (P>0.05). The vertical distance of the neck rail (120.4±10.4 cm) was significantly different from the standard values. The median of withers and locomotion scores were consistent among all farms. At the farm level, the median knee, hygiene, and hock scores did not show a significant correlation with the mean of neck rail measures and brisket board height (P>0.05). Also, the median locomotion score did not show a significant correlation with the mean horizontal distance of the neck rail at the individual freestall barn level (P>0.05). However, a significant correlation between the mean of knee scores and vertical distance of the neck rail at the farm level, and between the mean of locomotion score and horizontal distance of the neck rail at the individual freestall barn level were reported.CONCLUSIONS: An increase in the mean vertical distance of the neck rail is associated with an increase in the median knee scores, while an increase in the mean horizontal distance in each barn was associated with an increase in the median locomotion score, indicating the potential impact of these measurements on cow comfort. However, further research using a larger sample size is needed.

BACKGROUND: Based on the historical and recently published documents, it has been demonstrated that Mirabilis jalapa as a herbal medicine may be used as remedies for various health problems included wound healing purposes.OBJECTIVES: The present study aimed to investigate the effect of ethanolic extract of Laleh abbasi green leaf on healing open wounds induced by skin puncture in the back of rats.METHODS: Collecting and drying Laleh abbasi leaves, leaf extracting through Soxhlet procedure, analyzing the extract via gc / ms method, and preparing eucerin-based extract ointment were done according to recommended routine procedures. Herein, we recruited 40 male rats that were randomly divided into five groups of eight, namely the control, phenytoin treatment, eucerin, 5% eucerin extract, and 7.5% eucerin extract ointment treatment groups. Skin puncture and application of ointments on the induced wounds were carried out. Subsequently, tissue samples were taken on days 3, 7, 10, and 14, followed by which histological slides were prepared and stained with H&amp;E and Masson’s trichrome staining methods. In vitro mycological studies were conducted using opportunistic fungi, including Candida, Mucor, and Aspergillus species.RESULTS: Based on the macroscopic evaluations of the wound healing process and microscopic assessments of tissue samples stained with Harris H&amp;E and Masson’s trichrome methods, the groups treated with eucerin-based ointments containing ethanolic extract of Laleh abbasi leaf had statistically significant positive wound healing responses compared to the other groups. However, the 7.5% ointment group showed statistically better responses compared to the 5% ointment group. The data obtained in the present preliminary experiment on rat model indicated that the extract could facilitate the wound healing process in terms of healing parameters, such as accelerating epithelium repair, creating a favorable inflammatory reaction, angiogenesis, fibroplasia, and collagen precipitation. The antifungal effects of ethanolic extract of Laleh abbasi leaves on Aspergillus fumigatos, fusarium, Candida albicans and Candida cruzei were demonstrated in vitro using saboro dextrose agar medium.CONCLUSIONS: According to the findings of this experimental study and the findings of other researchers, it can be concluded that ethanolic extract from Laleh abbasi leaves is of healing and antifungal properties.

BACKGROUND: Excretory urography is a method of imaging the kidneys, ureters, and urinary bladder which uses contrast medium containing the iodine compounds.OBJECTIVES: This study aimed to evaluate the anatomical structures of urinary tract in the nephrogram, pilogram, and cystogram phases, and determine the exact standard for the size of kidneys, ureters and urinary bladder in guinea pigs to be used to interpret the results, and clinical decisions.METHODS: This study was carried out on 10 guinea pigs with a mean age of 12±1.33 months and average weight of 1.12±0.18 kg. Before to the administration of contrast medium, each guinea pig was fast and Dimethicone 20 mg/kg was given orally. At the time of administration of contrast agent, each animal was sedated by using Ketamine 30 mg/kg and diazepam 5 mg/kg cocktail, and then 1500 mgI/kg of meglumine compound 60 % was injected subcutaneously over the shoulder area. Ventrodorsales and lateral abdominal X-rays were taken, thereafter every 5 minutes up to 60 minutes to complete the pylogram phase. In lateral radiographs of each guinea pig, the length of the body of the second lumbar vertebra was measured to be used as an indicator in determining the standard size of the kidneys.RESULTS: Based on the results of this study, the average length, width, and thickness of the right kidney compared to the length of the second lumbar vertebra were 2.19, 1.64, and 1.33 cm, and in the left kidney of 2.09, 1.53, and 1.41 cm and this average in right and left ureter was 6.41 and 6.22 cm, respectively.CONCLUSIONS: The exact standards can be used in the interpretation of results, and clinical decisions to determine the normal and abnormal size of kidneys, ureters and bladder in the guinea pigs.

BACKGROUND: Coronaviruses, which mainly cause gastrointestinal and respiratory infections, have been identified in various species. Among the extensive genomic data of disease-causing Coronaviruses in humans and animals, some similarities can be analyzed by in-silico methods.   OBJECTIVES: In the present study, comparative genome analysis of medical and veterinary medicine Coronaviruses was performed to obtain more accurate information about the genetic similarities and differences of different members of this family.METHODS: The genomic sequences were retrieved from NCBI and Virus Pathogen Resource databases. Using the NCBI database blast algorithm, all sequences were aligned with the SARS-CoV-2 genome sequence, and similarity was obtained. Amino acid sequences of structural and non-structural proteins associated with coding regions (CDS) were aligned separately with the SARS-CoV-2, and their similarities were calculated. The 3D structure from each protein was compared with the corresponding protein in SARS-CoV-2, and Template Modeling Scores (TM-Score) were obtained. A phylogenetic tree of different species of the Coronaviridae family was drawn based on nucleotide and amino acid sequence data.RESULTS: Nonstructural coding gene sequences detected the highest interspecies similarities in nucleotide, amino acid sequence, and 3D structure (nsp12, nsp13, nsp14, and nsp16). The ORF1ab, encoding non-structural proteins, carries essential functions for viral replication.CONCLUSIONS: This study showed that the transcription complex is highly conserved among human and animal Coronaviruses. A comparison and analysis of the Coronaviridae transcription complex can be considered a key target for diagnosing, developing antiviral therapies, and designing vaccines.
