Evaluation of polyherbal formulation in broilers fed high energy diet: Implications on zootechnical parameters, fat accretion, and serum L-carnitine levels

Objective: The current broiler trial was planned to assess the effects of Kolin Plus™, a polyherbal formulation (PHF), on performance, protein and fat accretion, and serum L-carnitine (LC) levels in broilers fed a high-energy diet (HED). Materials and Methods: A total of 500 1-day-old Cobb 430 male chicks were assigned to 5 treat¬ment groups consisting of 10 replicates, with 10 birds in each replicate (n = 100). Group G1 was a negative control fed HED, and group G2, a positive control supplemented with synthetic choline chloride (SCC) 1,500 gm/ton in HED. Groups G3, G4, and G5 were supplemented with PHF in HED at 400, 500, and 750 gm/ton feed, respectively (PHF400, PHF500, and PHF750). Results: The PHF produced a dose-dependent numerical improvement in body weight, feed con¬version ratio, livability, and the European Production Index. There were no changes in carcass nitrogen and protein accretion, whereas a statistically significant decrease (p < 0.05) in carcass fat and fat accretion was observed in the SCC and PHF groups. Moreover, PHF showed a significant increase in serum LC levels. Conclusion: Kolin Plus™ improves performance parameters akin to SCC by improving fat metabo¬lism and mobilization by enhancing serum LC levels and restoring normal fat accretion. Objective: Despite the development of several vaccines against severe acute respiratory syndrome coronavirus-2, the need for an additional prophylactic agent is evident. In recent in silico studies, isovitexin exhibited a higher binding affinity against the human angiotensin converting-enzyme 2 (hACE2) receptor than existing antiviral drugs. The research aimed to find out the point specificity of isovitexin for the hACE2 receptor and to assess its therapeutic potential, depending on the stability of the isovitexin–hACE2 complex. Materials and Methods: The pharmacokinetic profile of isovitexin was analyzed. The crystal structure of the hACE2 receptor and the ligand isovitexin were docked to form a ligand-protein complex following molecular optimization. To determine the isovitexin–hACE2 complex stability, their binding affinity, hydrogen bonding, and hydrophobic interactions were studied. Lastly, the root mean square deviation (RMSD), root mean square fluctuation, solvent accessible surface area, molecular surface area, radius of gyration (Rg), polar surface area, and principal component analysis values were found by simulating the complex with molecular dynamic (MD). Results: The predicted Lethal dose50 for isovitexin was 2.56 mol/kg, with an acceptable maximum tolerated dose and no hepatotoxicity or AMES toxicity. Interactions with the amino acid residues Thr371, Asp367, Glu406, Pro346, His345, Phe274, Tyr515, Glu375, Thr347, Glu402, and His374 of the hACE2 protein were required for the high binding affinity and specificity of isovitexin. Based on what was learned from the MD simulation, the hACE2 receptor-blocking properties of isovi¬texin were looked at. Conclusions: Isovitexin is a phytochemical with a reasonable bioactivity and safety profile for use in humans, and it can potentially be used as a hACE2-specific therapeutic to inhibit COVID-19 infection. Objective: This study aimed to determine acaricide resistance in Rhipicephalus decoloratus ticks collected from grazing cattle between November 2018 and May 2019 in Elundini, Senqu, and Walter Sisulu Local Municipalities in the northeastern region of the Eastern Cape Province. Materials and Methods: A sample of 20–30 adult engorged female R. decoloratus ticks were collected from at least 10 randomly selected cattle (highly tick-infested) at each dip tank and placed into the labelled plastic collection bottles containing absorbent paper and with a perforated lid at a constant room temperature of ±28°C and >70% relative humidity until resistance testing commenced. The Shaw larval immersion test method was used to determine R. decoloratus larvae resistance to various acaricide concentration levels [amidines, organophosphate (OP), and synthetic pyrethroids (SPs)]. Results: This study found that most ticks were susceptible to exposure to different acaricide field concentrations of amidines (49% at 250 ppm), OPs (33% and 47% at 300 ppm and 500 ppm, respectively), and SPs (44% and 23% at 150 ppm and 300 ppm, respectively). The resistance test¬ing results showed no resistance to amidines at any localities and no resistance to OP in the Senqu region. However, resistance development of the larvae to amines, OPs, and SPs was extensively observed in Senqu (18%, 6%, and 7%), Elundini (15%, 15%, and 17%), and Walter Sisulu (13%, 19%, and 9%) regions, respectively. Conclusions: The larvae's resistance is a cause for worry. Hence, the continuous monitoring of tick resistance to commonly used acaricides will help mitigate widespread acaricidal resistance and sustain livestock productivity. Objective: Peste des petits ruminants (PPR) virus is the main infectious cause of goat mortality in Bangladesh, and co-infection may make diseases more severe. This study aimed to detect PPR and co-infecting diseases in goats. Materials and Methods: One hundred goats suspected to be infected with the PPR virus were collected from various areas of Mymensingh district, Bangladesh. A systemic post-mortem examination was carried out on PPR-suspected goats. Lungs, spleen, and lymph nodes (pre-scapular) were used for ribonucleic acid extraction, whereas lungs and mesenteric lymph nodes were used for deoxyribonucleic acid extraction. Seven-pair primer sets were used for molecular detection of pathogens specific for PPR, goat pox, contagious ecthyma (Orf), foot and mouth disease (FMD) virus, Klebsiella sp., and Mycobacterium sp. Reverse transcriptase-polymerase chain reaction (RT-PCR) or polymerase chain reaction (PCR) were used to find the exact cause. Results: Out of 100 PPR-suspected goats examined, 55 goats were confirmed as PPR-detected by RT-PCR. Among the 55 PPR-positive goats, 2 were co-infected with goat pox, 2 with tuberculosis, 10 with Klebsiella sp. infection, and 6 with FMD as detected by PCR and RT-PCR. Moreover, 12 goats were co-infected with PPRV and fascioliasis. Conclusion: About 58% of PPR virus-infected goats were co-infected with other organisms. There is a need to design technology to detect the state of co-infectivity at its early onset and future preventive and therapeutic strategies for co-infecting diseases. Objective: The objective of this study was to determine the effect of adding various doses of green tea extract to the semen of Kacang goats during the sexing process on motility, viability, membrane integrity, malondialdehide (MDA), and deoxyribonucleic acid (DNA) fragmentation. Materials and Methods: It started with the containment of the semen of the Kacang goat, followed by macroscopic and microscopic examinations. If the semen was considered viable, a diluter that had been added with various doses of green tea extract would be added to the semen. After that, sexing was carried out using the percoll gradient density medium. Next, the sexed semen was cryopreserved in liquid nitrogen. Then, an examination of motility, viability, membrane integrity, MDA, and DNA fragmentation was conducted. Result: There was a significant difference between the control and treatment (p ≤ 0.05). The highest result was obtained in the treatment of adding 0.05 mg of green tea extract/100 ml of Andromed®. Conclusion: The addition of green tea extract can improve the quality of the sexed semen of the Kacang goat after it has been cryopreserved. [J Adv Vet Anim Res 2022; 9(3.000): 412-418]

Objective: The increase and prevalence of multidrug-resistant bacteria in livestock animals are serious public health concerns. This study aimed to identify the presence of the blaTEM gene in extended-spectrum beta-lactamase (ESBL)-producing Escherichia coli isolated from rectal swabs of apparently healthy pigs in Malang District, East Java, Indonesia. Materials and Methods: A total of 120 rectal swab samples were collected from the pigs. The rectal swabs were screened for the presence of E. coli using standard microbiological identifi¬cation procedures. The Kirby–Bauer disk diffusion method identified multidrug-resistant E. coli. Five different classes of antibiotics were used to identify multidrug-resistant isolates, including Ciprofloxacin, Trimethoprim, Tetracycline, Streptomycin, and Aztreonam. Multidrug-resistant E. coli isolates were characterized for the presence of ESBL using double-disk synergy test methods. The presence of blaTEM genes was determined using polymerase chain reaction methods. Results: The results of this study indicated that 107 (89.2%) out of 120 samples analyzed were positive for E. coli isolates. A total of 32 (29.9%) E. coli isolates were identified to be multidrug-re¬sistant and further subjected to molecular testing. The molecular analysis revealed (5; 15.6%) E. coli isolates to harbor the blaTEM gene. Conclusion: The results of this study revealed that pigs and products of pork origin must be considered a source of transmission of ESBL-producing E. coli to public health important under the food chain. Objective: This study assessed the bacteriological quality and prevalence of foodborne bacteria in raw broiler meat sold in Mymensingh City. Materials and Methods: Thigh and breast meat samples (n = 80) from broiler chickens were randomly collected from four live bird markets (LBM) in Mymensingh city for bacteriological analysis. To determine the bacteriological quality, a 10-fold serial dilution of the thigh and breast homogenate was made. Then, total viable count (TVC), total coliform count (TCC), Staphylococci, and Salmonella spp. counts were determined using plate count agar, MacConkey agar, Mannitol salt agar, and Salmonella-Shigella agar. Gram stain, biochemical testing, PCR assays, and cultural properties were used to identify the bacterial isolates. Results: The TVC in the broiler meat sample ranged between log10 8.30 ± 0.54 colony forming unit (CFU)/gm and log10 9.04 ± 0.26 CFU/gm. TCC was found between log10 5.53 ± 0.38 CFU/gm and log10 6.66 ± 0.80 CFU/gm. The mean Staphylococcal count was recorded between log10 4.64 ± 0.61 CFU/gm and log10 6.42 ± 0.53 CFU/gm, and the total Salmonella count ranged between log10 4.75 ± 0.08 CFU/gm and log10 5.69 ± 0.58 CFU/gm. The prevalence of Escherichia coli was the highest (43.2%), followed by Staphylococcus aureus (36.8%) and Salmonella spp. (20%), respectively. Conclusions: Data from this study indicated that the TVC and TCC of raw broiler meat sold at LBM exceed the permissible limits and are contaminated with foodborne bacteria, which might cause public health hazards. Objective: There are three objectives in this study; 1) to measure the live weight of pre-and post-weaning Saanen kids; 2) to determine the growth curves between males and females pre-and post-weaning Saanen kids; and 3) to analyze the body condition score (BCS) between male and female pre-and post-weaning kids. Materials and Methods: This study included pre-weaning kids aged 1 month, as well as post-wean¬ing kids aged 3–6 months. 10 pre-weaning purebred Saanen kids (n = 5 male and n = 5 female) and 20 post-weaning Saanen kids (n = 10 male and n = 10 female) were employed in this investigation. Pre-and post-weaning kids' live weights were assessed weekly on a weighing scale, and BCS was calculated based on their body frame. In the data analysis, the two-sample t-test with Minitab Software was utilized. Results: The findings revealed that pre-weaning Saanen kids gained weight steadily from week 1 to week 6, with males being heavier than females. The p-value, on the other hand, suggested that there was no difference in live weight between pre-weaning male and female Saanen kids. Over 6 weeks of sampling, the male had a larger proportion of live weight gain (80%) than the female (75%). Meanwhile, the BCS of pre-weaning Saanen kids grew from week 1 to week 6. It is critical to account for the development of muscle mass while still evaluating the fat cover to determine whether the kids are maintaining an adequate BCS. However, the live weight of post-weaning kids was inconsequential because they were still in the growing phase. As a result, from the 1st to 6th week, post-weaning kids' body weight and BCS increased as their growth progressed. After 6 weeks of sampling, females had a higher percentage of live weight than males. This is because the kids raised on the farm do not have complete control over the environmental effects. Over 6 weeks of sampling, female post-weaning Saanen kids grew a slightly higher percentage of live weight (88%) than males (85%). Conclusion: This study conducted a direct assessment study, which monitored and determined the live weight and BCS of pre-and post-weaning Saanen kids. Pre-weaning kids' average values of live weight were calculated as insignificant at the age of 1 month. The mean live weight is most affected by milk consumption from its mothers, the management of the farm, and the environment. For the post-weaning Saanen kids, the females have a slightly higher average live weight gained for 6 weeks than the males (p > 0.05). Objective: This research aims to examine the effect of different fermentation times on the results of scanning electron microscope energy dispersive X-ray (SEM-EDX), chemical, organoleptic, and microbiological quality of banana peel. Materials and Methods: The design in this study used a completely randomized design (four treatments and four replications). The treatments in this study were different durations of fer¬mentation of banana peels; T0 = no fermentation; T1 = 6 days; T2 = 12 days; and T3 = 18 days. The research parameters were chemical, organoleptic, microbiological quality, composition, and SEM-EDX images. Analysis of chemical, organoleptic, and microbiological quality data was done using analysis of variance, followed by Duncan's multiple range testing at the 5% significance level, while the elemental composition and SEM-EDX images were analyzed descriptively. Results: The results of the study showed that the length of fermentation had a significant effect (p < 0.05) on moisture content, ash, crude fiber, nitrogen-free extract, and all organoleptic quality variables of banana peels. Different fermentation durations had no effect (p > 0.05) on crude protein, ether extract, metabolic energy, total lactic acid bacteria, total bacteria, Gram positive or negative bacteria, and total fungi on banana peels. The analysis showed that the elemental composition of banana peels is C, Na2O, Cl, K2O, MgO, SO3, SiO2, and P2O5. Conclusion: Eighteen days of fermentation improved the chemical quality, organoleptic, microbi¬ological, elemental composition, and SEM-EDX image of fermented banana peel. Objective: The research was designed to assess the consequences of Azadirachta indica aqueous leaf extract (AILE) on neuropathic pain in Wister rats and the role of the ATP-dependent potassium channel (KATP) as an underlying mechanism. Materials and Methods: This experimental layout was conducted on Wistar rats (n = 120) having 150 to 200 gm of body weight. On the foundation of the experimental design, rats were divided into group I (normal saline, 5 ml/kg/body weight) and group II (sham surgery and treatment with NS), group III [chronic constriction injury (CCI) in the sciatic nerve; and treated with NS], group IV (CCI and treated with AILE 400 mg/kg body weight), Group V (CCI, pretreated with Glibenclamide 15 mg/kg followed by treated with AILE 400 mg/kg). All the treatments were given once daily for a consecutive 21 days via the oral route, except Glibenclamide. Glibenclamide was given once through the intraperitoneal route on the day of the experiment. Results: Based on the neuropathic pain evaluation test, all groups were again sub-divided into subgroup "a" (walking tract analysis), "b" (cold tail immersion test), "c" (Von Frey test), and "d" (hot plate test). AILE showed a significantly higher sciatic functional index (p < 0.05) in walking track analysis, tail flick latency (p ≤ 0.05) in the cold tail immersion test, and paw withdrawal threshold (p ≤ 0.05) in the Von Frey test compared to CCI control. In addition, a nonsignificant difference in all these above-mentioned variables between the rats with CCI plus AILE and the CCI plus AILE plus glibenclamide group indicated that the KATP channel was not involved in the beneficial analgesic effects of AILE. Conclusions: The outcome of the present study indicates that AILE prevented worsening of neu¬ropathic pain after chronic constriction injury in the sciatic nerve of Wistar rats in which the KATP channel was not involved.
