Hematological parameters and morphological characteristics of blood cells in turtle and tortoise species within captivity in Sri Lanka

Objective: This study was conducted to determine hematological reference ranges and morpho¬logical characteristics blood cells in healthy captive Sri Lankan fresh water turtle and tortoise species. Materials and Methods: Following turtle species, namely, Melanochelys trijuga parkeri (n = 06), M. trijuga thermalis (n = 06), and Lissemys punctata punctata (n = 06) and a tortoise species Geochelone elegans (n = 06) were evaluated. Blood smears were stained using LeishmanGram staining protocol. The differential white blood cell counts and morphology were assessed using the standard protocols. Results: Both red blood cells and their nuclei were irregular in M. trijuga parkeri, whereas M. trijuga thermalis had oval-shaped and the nuclei were irregular in shape. L. punctata punctata and G. elegans had oval-shaped red blood cells and their nuclei were round in shape. In terms of differential counts, heterophils were the most abundant leukocyte type in all the species. Melanochelys trijuga parkeri had the highest heterophils value of 55% and the lowest value of 48.5% was recorded with G. elegans. Lymphocytes count was significantly high in G. elegans. Characteristic morphological features were observed in different leukocytes. In terms of thrombo¬cytes, M. trijuga thermalis and L. punctata punctata had single platelets, whereas M. trijuga park¬eri and G. elegans had platelets as clumps. Thrombocytes are ellipsoidal with centrally located dark-stained nuclei and their cytoplasm is clear. Conclusion: The findings in this study can be used as the reference values in the assessment of health of the above species and also will be useful in future hematological studies related to these species. Objective: The main objective of this study is to isolate, identify, and quantify the active antimicrobial compounds present in the crude aqueous stem bark extract of B. dalzielii using some common pathogenic microorganisms as well as toxicological profile. Material and Methods: Crude aqueous stem bark extract of Boswellia dalzielii (CASEB) was par¬titioned by preparative thin layer chromatography (PTLC) using chloroformmethanolwater, 8:2:1 (v/v). The resulting bands were extracted using chloroformmethanol (50:50). The extract of each band was evaluated for antimicrobial activity on Streptococcus pyogenes, Staphylococcus aureus, Escherichia coli, Enterococcus faecalis, Klebsiella pneumonia, Pseudomonas aeruginosa, Proteus mirabilis, Salmonella typhi, and Candida albicans by disc diffusion. Compounds in the most antimicrobially bioactive fraction (MAAF) were identified by high performance liquid chro¬matography (HPLC), Fourier transform infrared spectrophotometry (FT-IR), and gas chromatogra¬phy-mass spectrometry (GC-MS). Toxicological profile of the CASEB was evaluated by studying its effect in albino Wister rats. Results: PTLC produced five bands/fractions of which the MAAF was identified as RF2-fraction being active against all the isolates except E. coli and K. pneumoniae. HPLC of the MAAF revealed seven components; FT-IR revealed 17 functional groups; GC-MS revealed five compounds of which 93.18% are Oleic acid (44.88%), Squalene (34.16%), and n-Hexadecanoic acid (14.14%). The acute toxicity showed LD50 > 3,000 mg/kg. Sub-chronic toxicity showed that higher doses of the CASEB caused significant changes in liver function indices and a fatty change with lymphocytic infiltration (sign of acute hepatitis) in the liver tissues, but none of these changes were observed in the kidneys. Conclusion: The antimicrobially active compounds in CASEB were Oleic acid, Squalene, and n-Hexadecanoic acid. These can be further purified and used as precursors of new antimicrobial agents for treating infections especially those due to fungi and Pseudomonas spp. that are known to resist wide array of antimicrobial agents. The LD50 of CASEB is >3,000 mg/kg in rats. However, long-term consumption of CASEB is associated with significant liver damage. [J Adv Vet Anim Res 2019; 6(2.000): 183-192]

Objective: Vibrio spp., particularly, Vibrio cholerae is a major etiology of diarrhea in humans worldwide. In this study, we isolated and identified V. cholerae from the human stool of suspected cases along with antibiogram. Materials and Methods: In total, 25 stool samples from cholera suspected patients were ana¬lyzed. Isolation and molecular detection of Vibrio species were performed based on staining, motility, cultural and biochemical characteristics followed by polymerase chain reaction (PCR) using groEL gene-specific primers. Results: Among the 25 samples, seven showed growth of yellow color colonies on Thiosulfate- Citrate-Bile salts-Sucrose agar plates. The isolates were Gram-negative, curved shaped, and motile. Biochemically, they were found positive for indole and Methyl Red tests and negative for VogesProskauer test. Out of the seven positive samples, only three isolates were confirmed as Vibrio spp. using genus-specific primers. Subsequently, these three isolates were confirmed as V. cholerae by PCR using V. cholerae groEL gene-specific primers. Antibiotic sensitivity test revealed these three isolates as highly sensitive to azithromycin, chloramphenicol, gentamicin, and norfloxacillin while resistant to streptomycin, tetracycline, and oxacillin. Conclusion: Vibrio cholerae were isolated from the stool of diarrheic human patients and confirmed by PCR targeting the groEL gene. The isolates were found resistant to streptomycin, tetracycline and oxacillin, and need further characterization to reveal the molecular basis of their origin and resistance. Objective: An in vivo study was carried out to evaluate the possible anti-trypanosomal activity of Leptadenia hastata crude root extract with also its associated hematological changes particularly the packed cell volume (PCV) in experimental Trypanosoma brucei brucei infection using Wistar rats. Materials and Methods: Thirty Wistar rats comprising of both males and females were categorized into six separate groups starting from A to F. Wistar placed in Group A and Group B were inoculated with T. brucei brucei and administered crude root extract of L. hastata at 100 and 200 mg/kg, respectively, as the treatment. Group C was infected with the parasite but untreated, while Group D was not infected with the parasite and was not treated. The remaining groups E and F were inoculated with the parasite using diminazene diaceturate at 3.5 and 7.0 mg/kg, respectively. The extract was administered enterally when parasitemia was detected. Standard laboratory techniques were employed to determine parasitemia and PCV after collection of blood samples every 2 days via the tail vein. Results: Infected groups (A, B, C, E, and F) showed a pre-patent period 2 days post infection (P.I) with mean parasitic counts of 3.93 ± 2.38, 2.46 ± 2.20, 0.67 ± 0.77, 4.60 ± 4.45, and 1.53 ± 1.44, respectively, which continued unabated in groups treated with the extract.The pack cell volume did not decline significantly in the in groups A and B. Acute toxicity study revealed the absence of any clinical or behavioral changes suggesting toxicity. Conclusion: There was no effect on parasitemia of Wistar rats infected with the parasite after adminis¬tration of 100 and 200 mg, respectively, using the extract as the treatment. PCV of the groups infected remained fairly constant with the control groups throughout the study with the extract being non-toxic. [J Adv Vet Anim Res 2019; 6(2.000): 241-246]

Objective: To explore the effect of glycerol at different concentrations using different extenders on DNA fragmentation and motility of frozen-thawed Kintamani Bali dog spermatozoa. Materials and Methods: Sample was collected from four mature Kintamani Bali dogs. Each ejacu¬late was prepared for cryopreservation with two different semen extenders; egg yolk Tris extender and coconut water-based extender. For each extender, three different glycerol concentrations were used; 4%, 6%, and 8%. Each of the six aliquots was loaded into 0.5 ml cryotube, placed on a styrofoam box 5 cm over liquid nitrogen for 10 min, and immersed in liquid nitrogen up to 8 min. Then, the frozen cryotubes were transferred into liquid nitrogen container. The cryotubes were thawed in a water bath at 38.5°C for 120 sec. After equilibration and thawing, each sample was assessed for motility parameters and for DNA fragmentation. Results: The addition of 6% glycerol to extenders revealed the most effective addition of glycerol on motility and sperm DNA fragmentation after equilibrium and post-thawing. Conclusion: It is concluded that both extenders with the addition of 6% glycerol are safe to be used as an extender in Kintamani Bali dog semen preservation, and DNA fragmentation of Kintamani Bali dog spermatozoa was not influenced by the freezing procedure. Objective: This study aims to determine the ameliorative effect of vitamin E (vit E) on histological features and androgen binding protein (ABP) levels in rats induced by allethrin. Materials and Methods: Thirty sexually mature male Wistar rats weighing between 200 and 300 gm, and aging 3 months were taken for this study and were divided into three groups: negative control (NC), positive control (PC), and treatment (T) groups. The PC and T groups were induced by allethrin 12 h per day for 31 days; however, only the T group was given vit E orally at 1 ml/gm body weight (BW) each day for 14 days. The paraffin block method was used to measure tubules diameter, thickness of the seminiferous epithelial layer, and Sertoli cell number. The ABP levels were measured by enzyme-linked immunosorbent assay. Results: The results showed that vit E gave significant effect (p < 0.05) on tubular diameter at NC 123.67 ± 12.77, PC 147.16 ± 10.64, and T 130.08 ± 10.00; tubular epithelial thickness at NC 33.55 ± 3.21, PC 30.02 ± 1.53, and T 32.96 ± 2.81; Sertoli cells number at NC 55.48 ± 5.9, PC 43.84 ± 3.77, and T 53.44 ± 4.26; and ABP levels at NC 72.35 ± 39.06, PC 38, 48 ± 18.78, and T 86.10 ± 35.77, respectively. Conclusion: This study concludes that vit E has an ameliorative effect against the toxic effects of allethrin at testicular histological features and ABP levels. Objective: The study was aimed to isolate, identify, and characterize common indicator bacteria, including Escherichia coli, Salmonella spp., and Staphylococcus spp. in manure and bio-slurry sam¬ples of different livestock farms and biogas plants of Bangladesh. Materials and Methods: A total of 114 samples of manure and bio-slurry were collected from different livestock farms and biogas plants in Bangladesh. The total viable count (TVC), E. coli, Salmonella spp., and Staphylococcus spp. counts were determined by the spread plate technique method. Isolation and identification were performed by colony characteristics, staining, bio¬chemical tests, and, finally, by using PCR. Antibiotic susceptibility test of the isolated bacteria was tested against commonly used antibiotics by using the disk diffusion method. Results: The mean TVC, E. coli, Salmonella spp., and Staphylococcus spp. counts were ranged from 8.1910.75, 5.26.96, 5.816.87, 5.687.68 in manure samples and 7.268.65, 3.825.2, 45.54, 3.145.9 log cfu/gm in bio-slurry, respectively. In anaerobic digester after 30 days digestion, the presence of E. coli, Salmonella spp., and Staphylococcus spp. varied from 05.11, 04.84, and 05.59 log cfu/gm at 25°C, 27°C, 29°C, and 45°C temperature. Above-mentioned bacteria were absent in bio-slurry collected from anaerobic digester after 60 days digestion at environmental temperature. Bacterial counts were reduced significantly in both household slurry pits and exper¬imental anaerobic digester. Antibiotic susceptibility results revealed that multidrug-resistant indi¬cator bacteria were present in the bio-slurry samples. Conclusion: Our findings conclude that the microbial load after treatment of animal manure via anaerobic digestion (Biogas plant) was grossly reduced and the reduction of bacterial pathogen depends on the duration and temperature of digestion. Objective: The current study aimed to determine the prevalence, infection burden, and risk factors associated with the occurrence of gastrointestinal (GI) parasites in different avian species in Ilorin, Nigeria. Materials and Methods: This study was conducted in Ilorin, involving 597 fecal samples and GI tracts from a variety of sold and slaughtered avian species. The study was conducted between September 2017 and February 2018. Fecal samples were examined using floatation technique, while the GI tracts were examined for gross helminths and its content were subjected to the direct wet mount examination. Data were analyzed using percentages (descriptive) and the Chi-square (χ2 ) test (inferential). p < 0.05 was considered significant for all analysis. Results: Ten GI parasites were detected with Eimeria species (32.83%), Ascaridia galli (30.15%) and Heterakis gallinarum (24.79%) as the most prevalent ones. Multiple parasites co-infection was recorded in all the avian species: broilers (77.78%), layers (33.33%), cockerels (45.16%), indigenous chickens (17.91%), ducks (69.70%), pigeons (94.12%), turkeys (47.83%), and guinea fowls (77.36%). Pigeons (100.00%) and turkeys (95.65%) were the most infected avian species. Age, sex, and avian types were significantly (p < 0.05) associated with the occurrence of GI parasites infection. Conclusion: This study gives a reflection of the GI parasites fauna of avian species in Nigeria. The GI parasites are endemic among different avian speciesin Ilorin, North Central Nigeria. Knowledge on the epidemiology of these parasites is important in instituting a good preventive and control measures against GI parasites, so as to have maximum production and reproduction effects in the poultry industry. Objective: We performed a randomized two-way crossover study to evaluate the pharmacokinetic profiles of two high-dose ascorbic acid (AA) after IV infusion in healthy beagle dogs. Materials and Methods: The dogs were administered IV AA at two doses of 1.5 and 3 gm/kg for 4 h, and the AA concentration in plasma and urine pH was measured before and after administration. Results: The plasma concentrations of AA in both groups peaked 3 h after administration. Among the two groups, the urine pH was not significantly different (p = 0.12990.7944). High-dose IV AA did not induce serious adverse events in dogs. Conclusion: The results of this study suggest that the high dose of AA which reaches the therapeutic dose for cancer and supports the safety of high-dose IV AA in dogs. Objective: The objective of this research work is to know the productive and reproductive perfor¬mances and problems of local, crossbred, Nilli, and Murrah buffalo cows in selected study areas in Bangladesh. Methodology: A total of 1,241 local, crossbred, Nilli, and Murrah buffalo cows were surveyed in the selected areas with a pre-set questionnaire. Among 1,241 buffalo cows, 112 buffalo cows were randomly selected at day 0 of the estrus cycle for studying ovarian features. Results: Results showed that the average age, body condition score, and body weight were sig¬nificantly (p < 0.05) different among the studied breeds. Milk production in Murrah and lactation length in Nilli cows were significantly (p < 0.05) higher than indigenous, crossbred, Nilli, and indig¬enous, crossbred, Murrah buffalo cows, respectively. Results also illustrated that sexual maturity, estrus cycle length, insemination time after the onset of estrus, and gestation length insignifi¬cantly (p > 0.05) varied among the surveyed breed. But, the fallout of the study denoted that estrus duration, first calving age, parity number, number of service per conception, calving inter¬val, and voluntary waiting period varied significantly (p < 0.05) in different breeds. Ovarian phys¬iological characteristics such as vaginal electrical resistance, average number of follicles in two ovaries, and largest follicular diameter, estrogen, and progesterone at day 0 of the estrus cycle of local, crossbred, Nilli, and Murrah buffalo cows showed insignificantly (p > 0.05) differences.
