Potency of spirulina (Spirulina platensis) on arsenic-induced lipid peroxidation in rat

Objective: Natural substances found in dietary sources and medicinal plants have attracted con¬siderable attention in recent years as chemopreventive agents. Spirulina is a blue-green alga that possesses chemopreventive properties. The purpose of this study was to determine the effect of spirulina on rats with inorganic arsenic (As) [sodium arsenite (NaAsO2)]-induced lipid peroxidation. Materials and Methods: 120 rats were randomly assigned to 10 groups and designated T0, T1, T2, T3, T4, T5, T6, T7, T8, and T9. One group was kept as a control (T0) that received no treatment. The seven groups received 3.0 mg of NaAsO2/kg body weight in drinking water and were given spirulina ad libitum. T1 was treated with NaAsO2 but not with spirulina. Two groups of rats (T2 and T3), on the other hand, were treated with spirulina without receiving any As (NaAsO2). T2 received agro-based spirulina (Ab-Sp; grown in 1.5% soybean meal media and harvested on day 12 of seed inoculation) at 2.0 gm/kg feed, whereas T3 received commercially available spirulina (Com-Sp) at 2.0 gm/kg feed. T4, T5, and T6 were concurrently treated with Ab-Sp at 1.0, 1.5, and 2.0 gm/ kg of feed. On the other hand, T7, T8, and T9 induced by NaAsO2 were concurrently treated with Com-Sp at 1.0, 1.5, and 2.0 gm/kg feed. All groups received treatment for 90 days. Results: The efficacy of both spirulina in preventing lipid peroxidation caused by As was deter¬mined quantitatively by measuring the rats serum malondialdehyde (MDA). The results indicated that As supplementation increased serum MDA levels, whereas both types of spirulina signifi¬cantly decreased them. The highest dose of Ab-Sp (2.0 gm/kg feed) was found to be the most effective in preventing lipid peroxidation in rats treated with inorganic As. Conclusion: Ab-Sp could be a natural, cost-effective, and safe measure to mitigate As toxicity. [J Adv Vet Anim Res 2021; 8(2.000): 330-338]

Objective: The objective of the study was to assess the effect of A,D3,E (I/M) and oregano oil extract 15% on some clinicopathological parameters during lamb bacterial enteritis treatment. Materials and Methods: Sixty Barki lambs, 20 apparently healthy (control group) and, 40 suf¬fered from bacterial enteritis [enteric group (EG)], were subdivided into four treated groups (TGs): antibiotic group (AG), antibiotic + A,D3,E group (A + A,D3,E), antibiotic + oregano oil (AOG), and oregano group (OG). Fecal swabs were collected from EG then aseptically cultured, isolated, phe¬notypically identified, genotypically confirmed, and sequenced by PCR 16srRNA. Paper disk dif¬fusion test was used for estimation of oregano oil extract 15% antibacterial activity. After blood sample aspiration from all animals, they were clinicopathologically and statistically analyzed. Results: Escherichia coli, followed by Salmonella species and then Klebsiella species, was the main causative agents of lamb diarrhea and were susceptible to oregano oil extract 15%. A + A,D3,E and AOG showed significant (p < 0.05) enhancement of some clinicopathological parameters more than AG or OG. Matrix metalloproteinases (MMP-2 and MMP-9) and total antioxidant capacity (TAC), yielded area under the curve, sensitivity, negative predictive value as 1, 100% and 100% respectively, were determined in both EG and TGs. Conclusion: Oregano oil extract 15% has good antibacterial properties against enteric bacteria in vitro and in vivo. The combination between antibiotic and antioxidant vitamins or oregano plant extract of 15% has a good impact on some clinicopathological alterations in lamb bacterial enteri¬tis treatment. TAC, MMP-9, and MMP-2 may be good markers for the disease and its treatment follow-up. [J Adv Vet Anim Res 2021; 8(2.000): 291-299]

Objective: This study was carried out to estimate the sex ratio, maturity size, gonadosomatic index (GSI), and peak breeding season of mud crabs. Materials and Methods: Samples were collected randomly from the estuary and river of the study area. Sampling was carried out monthly from April to September at every full moon during one high tide. A total number of 240 specimens were sampled, where 53 individuals were hermaphrodite. The crabs were shifted alive to the biology and histology lab for detailed biological study. Sex was determined. Male and female sex ratio and breeding season were also investigated. Results: The male:female ratio was 1:0.96 and the ovarian development was categorized into five stages based on internal observations, viz. immature (stage I), underdeveloped (stage II), early developed (stage III), late developed (stage IV), and mature (stage V). The maturity percentages were 37%, 19%, 13%, 11%, and 20%, respectively. 50% maturation was estimated at 82.36 mm inter¬nal carapace width (ICW). The highest mean GSI value was 7.97 ± 3.03. The mature stage was found in all the working periods. This shows that females have activated ovaries in all the working months, and the species are continuous breeders. A higher frequency of vitellogenic ovary and higher GSI value were found in September. The maximum GSI value was found in the size group 7079 mm. Conclusion: The study shows that the capture from the wild sources of mud crabs without any regulation can threaten the population structure. The capture of female mud crabs should be more than 82.36 mm ICW, which will help conserve and protect young crabs. [J Adv Vet Anim Res 2021; 8(1.000): 44-50]

Objective: The effect of seasonality needs to be considered in designing future studies because global warming has caused a rise in ambient temperatures. The objective of the present study is to investigate the effect of high ambient temperatures on fecal score and fecal microflora in dairy cows during summer. Materials and Methods: During the 7 days before the sampling of feces, the daily mean tempera¬tures were 19.9°C in early summer and more than 27.5°C in late summer. Fecal samples were collected from the rectum of cows and the fecal score was evaluated on a 4-point scale. The equalized samples were used to extract the genomic deoxyribonucleic acid (DNA) of the bacteria (Escherichia coli, Salmonella, Lactobacillus, and Bifidobacterium). Results: There was no significant difference in fecal scores between the sampling times in early and late summer. In the populations of the bacteria, there was no significant difference between sampling days in the DNA level of Salmonella, and E. coli in late summer increased to more than three times the level in early summer. However, both levels of Lactobacillus and Bifidobacterium in early summer significantly decreased after 2 months. Conclusion: These data suggest that the increase in temperature in late summer may adversely affect the populations of bacteria in the intestinal environment of dairy cows. In addition, the method used in the present study was sufficient to evaluate the changes in internal and external environmental conditions of dairy cattle. Objective: This study aimed to explore the seroprevalence of Brucella spp. in goats in some selected areas of Bangladesh. Materials and Methods: This study was conducted in different goat-populated regions of Bangladesh from July 2017 to June 2018. A total of 208 serum samples were randomly collected from goats in Jashore (n = 50), Jhenidah (n = 22), Tangail (n = 40), Savar (n = 46), Thakurgaon (n = 18), and Bandarban (n = 32) areas. The samples were subjected to determine the presence of antibodies against Brucella spp. by rose bengal plate test (RBPT) and competitive enzyme-linked immunosorbent assay (c-ELISA). Results: Overall, the seroprevalence of Brucellosis in goats was 4.33% (n = 9/208) by RBPT and 2.40% (n = 5/208) by c-ELISA. The seroprevalence of brucellosis on the basis of RBPT was 6% (buck: 0%, doe: 6%) in Jashore, 4.5% (buck: 0%, doe: 4.5%) in Jhenidah, 2.5% (buck: 0%, doe: 2.5%) in Tangail, 4.35% (buck: 0%, doe: 4.35%) in Savar, 6.25% (buck: 0%, doe: 6.25%) in Bandarban, and 5.56% (buck: 0%, doe: 5.56%) in Thakurgaon. On the other hand, the seroprevalence of brucel¬losis by c-ELISA was 4% (buck: 0%, doe: 4%) in Jashore, 4.5% (buck: 0%, doe: 4.5%) in Jhenidah, 3.13% (buck: 0%, doe: 3.13%) in Bandarban, and 5.56% (buck: 0%, doe: 5.56%) in Thakurgaon. Brucellosis was more prevalent (p > 0.001) in does aging 34 years. Conclusion: Goats from different areas of Bangladesh are caring antibodies against Brucella organisms. Objective: This study investigated and compared the chemical composition, cholesterol content, fatty acid (FA), and amino acid (AA) profiles of doi and rasomalai made from buffalo milk. Materials and Methods: Bangladesh Agricultural University Dairy Farm, Mymensingh-2202, Bangladesh was the source of raw buffalo milk. Then, doi and rasomalai were produced and ana¬lyzed. Prior to the production of doi and rasomalai, the gross composition and AAs of milk were evaluated. Milk and dairy products were evaluated for gross composition using an automated milk analyzer and the Association of Agricultural Chemists techniques, respectively. At the Bangladesh Council for Scientific and Industrial Research, Dhaka, Bangladesh, the cholesterol, FA, and AA lev¬els of doi and rasomalai were determined. Additionally, atherogenic and thrombogenic indices were determined using established equations. Results: The results indicated that the majority of the proximate components were significantly greater (p < 0.05) in rasomalai than in doi. Rasomalai had 3.64 mg more cholesterol (p > 0.05) than doi. The FA profile was identical across doi and rasomalai with the exception of oleic acid (C18:1cis-9), which was 1.50% greater (p < 0.05) in rasomalai. The atherogenicity index was found to be statistically higher in doi than in rasomalai (p > 0.05). Similarly, the thrombogenic index was found to be significantly higher (p > 0.05) in doi (1.98) when compared to the rasomalai (1.92). The concentrations of all AAs were found to be quantitatively higher in doi than in rasomalai (p > 0.05). Objectives: The objective of this study was to perform a comparative analysis of allelic diversity to reveal population-genetic characteristics of animal breeds, namely Shami (SH), Holstein (HLS), and Aberdeen-Angus (A-A). Materials and Methods: The genetic materials of SH breed animals represented by wool with hair follicles were collected from 39 SH heads in Syria. Also, genetic materials of HLS breed of American selection (n = 55, HLS) and bulls and cows of A-A breed bred at breeding enterprises in Russia (n = 30, A-A) were collected. Genetic differences between the cattle groups were studied using 11 microsatellite markers. Results: The cattle breed in Syria was characterized by high genetic diversity, 107 alleles, while the average number of alleles per microsatellite locus was 9.23, which is significantly higher than that in the animals of HLS (6.18) and A-A (5.00). When analyzing the genetic equilibrium for indi¬vidual locus in SH breed, a deviation from equilibrium at four loci was revealed: TGLA227, SPS115, TGLA122, and ETH225; at one locus in HLS breed: SPS115, for A-A breed: at two loci, i.e., TGLA122 and ETH225. When assessing the level of genetic consolidation, a deficiency of heterozygotes was observed in two of the three studied breeds: 4.8% for SH and 8.0% for A-A. A slight excess of heterozygotes was found in the HLS breed at the level of 0.2%. The average comparative measure¬ment of genetic variation in different populations value for 11 loci for all breeds was 0.069, which indicates that 93.1% of the total variability is due to the intra-breed diversity, and only 6.9% is due to the differences between breeds. Conclusion: The analysis of the animals belonging to their breed has shown a 100% genetic con¬solidation and the compliance of individual animals with the respective breeds. The study of genetic distances, adjusted for small samples, revealed the smallest genetic distance between the SH breed and HLS breed, equaling 0.107. The A-A breed, which has its separate origin and has never been imported into the Syrian Arab Republic, adjoins this cluster as an independent branch. Microsatellites can be used as an essential criterion for assessing the population-genetic charac¬teristics of groups of cattle of various breeds (degree of polymorphism, level of heterozygosity, fixation indices, genetic group membership). Objective: Previously, we have shown that predicted zymogen granule protein 16 homolog B (P-G3MZ19) existed in Bali cattle (Bos javanicus) saliva. It was suggested that P-G3MZ19 is a mem¬ber of the mannose-binding lectin family that plays an essential role in innate immunity. In the present study, we aimed to analyze the structure and ligand-binding of P-3MZ19 in Bali cattle saliva. Materials and Methods: Saliva of four adult healthy Bali cattle was collected, lyophilized, and subjected to two-dimensional (2-D) gel electrophoresis. The target spot of around 17 kDa related to P-G3MZ19 was excised for matrix-assisted laser desorption ionization time-of-flight mass spec¬trometer/time-of-flight mass spectrometer mass spectrometry analysis and sequencing. The structure and the ligand-binding of P-3MZ19 were analyzed using bioinformatics software pro¬grams published elsewhere. Results: Based on Iterative Threading ASSEmbly Refinement the 3D model of P-G3MZ19 was suggested to have similarities to exo-alpha-sialidase (EC 3.2.1.18); while its ligand-binding sites consisted of seven residues, i.e., 25aa-26aa (Gly-Gly), 95aa (Phe), 138aa (Tyr), 140aa (Leu), 141aa (Gly), and 143aa (Thr). Conclusion: The structure of P-G3MZ19 of Bali cattle saliva and its ligand-binding sites have been successfully determined by using bioinformatics techniques. The biological and immunological roles of the peptide are currently under investigation based on P-G3MZ19 synthetic peptides. [J Adv Vet Anim Res 2021; 8(2.000): 224-229]

Objective: Present research aims to isolate, identify, and determine the virulence of the Streptococcus agalactiae (group B Streptococcus; GBS), isolated from popped eye disease affected Tilapia and Vietnamese Koi (V. Koi) fishes. Materials and Methods: A total of 330 fish samples were collected, of which Tilapia (n = 180) and V. Koi (n = 150), were collected from 35 affected ponds of four selected districts of Bangladesh. Isolation of the bacterium was done using different culture media (Nutrient broth, Plate count agar, Tryptic Soy Agar, and Blood agar), and identification by using various biochemical tests (con¬ventional and using API 20 Strep kit) and polymerase chain reaction (PCR) using primers against 16S rRNA gene of S. agalactiae. Antibiotic susceptibility of the bacteria was performed using seven different antibiotics disc (Tetracycline, Oxytetracycline, Chlortetracycline, Streptomycin, Ciprofloxacin, Gentamicin, and Neomycin). Virulence of the isolated S. agalactiae was determined by infecting healthy Tilapia and V. Koi fishes through experimental infection. Results: Isolated bacteria were found Gram-positive paired and chained cocci, β-hemolytic and non-motile. Findings of biochemical and serological tests indicate that the isolated bacterium belongs to Group B Streptococcus of Lancefield classification. PCR result also confirmed that the bacteria were S. agalactiae. The bacterial isolates possessed resistance property against all the seven antibiotics used in this study. The isolated GBS was found highly virulent and showed 80%90% mortality for Tilapia and V. Koi fishes in experimental infection within 16 days of post-infection. Conclusion: From the findings of this study, it may be concluded that isolated GBS from the Tilapia and V. Koi fishes were highly virulent and possessed multidrug-resistance properties. Objective: Collagen is a fibrous protein that is primarily used in the pharmaceutical and biomedical industries. This study isolates and characterizes type-1 collagen from Sardine longiceps (scales, skin, and muscle). Materials and Methods: Collagen was isolated from S. longiceps using two methods: acid-solubilized collagen and pepsin-solubilized collagen. Sodium dodecyl sulfatepolyacrylamide gel electrophoresis (SDSPAGE) was used to estimate the molecular weight of isolated collagen. Ultraviolet (UV)-visible spectrophotometry analysis was used to confirm the collagen extracted (type-I collagen). The functional groups of isolated collagens were identified using fourier trans¬form infrared (FTIR) analysis. The X-ray diffraction (XRD) technique was used to investigate the crystallinity of isolated collagen. The high-pressure liquid chromatography (HPLC) technique was used to study the amino acid composition. Results: SDSPAGE of S. longiceps revealed molecular weights ranging from 116 kDa for α-2 to 97 kDa for α-1. UV-visible spectra showed an absorbance value below 300 nm, and the results confirmed type-I collagen. FTIR showed major functional groups like amide A, B, I, II, and III. XRD determined the crystallinity of isolated collagen. The HPLC results showed the presence of higher glycine content, followed by proline and hydroxyl proline in the extracted collagen. Conclusion: The overall study confirmed that fish waste materials (scales, skin, and muscles) could be used as an alternative source for collagen. [J Adv Vet Anim Res 2021; 8(4.000): 679-686]
