Phylogenetic studies of Newcastle disease virus isolated from poultry flocks in South Sulawesi Province, Indonesia, in 2019 Полный текст

Objective: Indonesia is one of the Newcastle disease (ND) endemic countries in the world. An outbreak of the ND virus(NDV) was first reported in Indonesia in 1926. This study aimed to detect, isolate, and classify the NDV by molecular approaches from poultry farms in South Sulawesi Province of Indonesia in 2019. Materials and Methods: As many as 36 pooling samples from the cloacal swab, trachea swab, proventriculus, and spleen tissues obtained from ND-suspected chickens were isolated in 11-day old embryonated chicken eggs type-specific antibody-negative. The viruses were confirmed by reverse transcription-polymerase chain reaction (RT-PCR), followed by sequencing. Results: The results showed that 18 out of 36 pooling samples were NDV-positive based on the isolation result and RT-PCR test. The sequencing results showed that 10 NDV isolates had a motif 112R-R-Q-K-R-F117 in the fusion protein cleavage site region, which suggested that the NDV isolates were of virulent pathotype. The phylogenetic studies based on the F genes partial nucleotide sequence classified the study isolates into NDV virus genotype/subgenotype VII.2. Conclusion: These findings are expected to help provide the latest characteristic information of NDV in South Sulawesi Province to determine the seed vaccine for control strategies of ND. Objective: The objective of this study was to determine the location, number, and direction of the nutrient foramen in the humerus, radius, femur, and tibia bones of mixed breed dogs. Materials and Methods: The humerus, radius, femur, and tibia of both (left and right) limbs of mixed breed dogs were examined in this study. The number, location, and direction of the nutrient foramina were identified. Once identified, the diameter of each nutrient foramen was measured and the site index calculated. Results: Only one nutrient foramen was identified in the humerus, radius, tibia, and right femur, while the foramen numbers ranged from one to three in the left femurs examinated. The nutri¬ent foramen was localized on the caudal surface in the radii, femurs, tibias, and left humeri. Contrasting, however, 75% were located on the caudal surface of the right humeri and 25% on the lateral surface. The average diameter of the nutrient foramen of the humerus ranged from 0.88 to 1.00 mm, while it ranged from 1.13 to 1.25 mm in the radius. On the hind limb, the diameter of the nutrient foramen on the femur ranged from 1.2 to 1.3 mm and 0.751.25 mm on the tibia. The nutrient foramen was directed towards the corresponding joint in 100% of the humeri and tibias, 75% of the radii, and 60%80% of the femurs examined. Conclusion: The anatomical data on the nutrient foramen obtained in this study will be valuable for veterinarians when diagnosing pathological bone lesions and for orthopedic surgery. Objective: This study evaluated the different traits from day-old to age at first eggs of Fayoumi and White leghorn (WLH) F1 chickens and their reciprocal crossbreeds. Materials and Methods: In this study, 1,000 eggs were used to obtain experimental chicks. However, only 150 chicks were selected from each genotype for subsequent trial evaluation and reared on the same diet in a completely randomized design. The effects of genotype on each considered trait were analyzed by the statistical analysis system, and Duncans multiple range test made a significant difference. Results: Comparative evaluations made on all parameters illustrated significant differences (p < 0.05) throughout the study period. F1 acquired from WLH cocks and Fayoumi hens maximized weights almost at all ages during brooding, whereas pure line Fayoumi showed lower importance for the trait stages of growth. Offspring from the crossbreed of WLH cocks and Fayoumi hens had the highest weight gain than any other crossbreeds from 4 to 8 and 12 to 16 weeks of age. Pure line WLH and Fayoumi lay larger eggs and smaller eggs (41.67 and 34.00 g), respectively. The highest and smallest body weight at the age of first egg was 1,364.89 and 1,178.36 g in the main crossbreed and Fayoumi line. The main WLH cock and Fayoumi hens and reciprocal crossbreds advanced in age at first egg by 4 and 11 days, respectively. Conclusion: The leghorn hen and Fayoumi cock crosses could be used to develop potential egg-laying lines than their reciprocal crossbreeds. [J Adv Vet Anim Res 2021; 8(1.000): 1-6]

Objectives: Wound healing is a complex and dynamic process that begins immediately following tissue injury and continues until the wound is completely healed and remodeled. Applying the most effective burn repair techniques is a constant challenge in medicine. Antiulcerogenic and wound healing properties of Areca palm leaves have been validated through various investigations and animal studies. This study aimed to determine the potential for A. palm hydroalcoholic extract to heal burn wounds in rats. Materials and Methods: For 14 days, we examined 40 male Wistar albino rats in 5 groups: those receiving 1% silver sulfadiazine cream (reference standard), those receiving eucerin (positive control), and those receiving 5% and 10% ointments of Areca catechu hydroalcoholic extract (treatment groups). No treatment was given to the negative control group. On the dorsal part of the animals necks, burn wounds were made. After the rats were sacrificed, the wound contraction rate (WCR) was determined, and the wound sites were histopathologically examined. Results: On the 14th day, the WCR was significantly higher in rats treated with A. palm 10% extract ointment than in rats treated with 5% extract, positive or negative control groups (p < 0.001), or rats treated with silver sulphfadiazine (p = 0.01). After applying a 10% extract ointment to burn wound sites, complete healing occurred with only mild tissue inflammation and edema. Conclusion: The studys findings indicate that the hydroalcoholic extract of A. palm L. has the abil¬ity to expedite the wound healing process. Additional research is necessary to identify the compounds responsible for their wound healing properties and comprehend their action mechanism. [J Adv Vet Anim Res 2021; 8(4.000): 619-628]

Objective: The objective of this study was to obtain an insight into the role of the vascular endothelial growth factor (VEGF) in pregnancy-associated mammary epithelial development and angiogenesis. However, we examined the primary VEGF receptor (VEGFR-2) in the mouse mammary cycle. Materials and Methods: The virgin (1012 weeks), days 10 and 18 of pregnancy (P-10 and P-18), days 0, 5, 10, and 20 of lactation (L-0, L-5, L-10, and L-20), and days 5 and 10 of post-weaning (W-5 and W-10) stage were all used in this study. Immunohistochemistry and Western blotting were carried out on mammary tissues taken from three mice at each stage. Results: VEGFR-2 was detected immunohistochemically in the cytoplasm of mammary epithelial and endothelial cells. Immunostaining for VEGFR-2 was consistently positive in mammary endothelial cells across all stages, whereas staining intensity in epithelial cells varied across the mammary cycle. Additionally, immunoblot analysis indicated a 220 kDa unique band of VEGFR-2 protein at all stages of the mammary cycle, with the maximum expression reported toward the end of pregnancy and gradually decreasing toward the end of lactation. Conclusion: In conclusion, the presence of VEGFR-2 in the mammary epithelium in addition to the endothelium suggests that VEGF plays an autocrine and paracrine role in the development, proliferation, and differentiation of the mammary epithelium during pregnancy. Objective: The goal of this study was to determine the levels of lead (Pb), cadmium (Cd), and aluminum (Al) in raw bovine milk. Milk consumption was used to calculate the estimated weekly intake (EWI), provisional tolerable weekly intake (PTWI), and target hazard quotient (THQ) for individuals. Metal distribution in dairy products and byproducts was studied as a result of artisanal processing. Material and Methods: Seventy-five raw bovine milk samples were collected from artisanal producers in Alexandria city, West Delta, Egypt, and analyzed using the atomic absorption spectrophotometer. The effect of artisanal dairy processing on metal distribution was studied. Results: The averages of Pb, Cd, and Al in milk samples were 45.06, 4.77, and 2.93 μg/l, with 13.33% and 1.33% of analyzed samples had Pb and Al levels higher than the permissible limits, respectively. The EWI values of Pb, Cd, and Al were 1.050, 0.111, and 0.068 μg/kg body weight, which contributed to about 4.20%, 1.59%, and 0.97% from the PTWI, respectively. Objective: Fast foods are often responsible for staphylococcal foodborne illness. The present study was carried out to isolate Staphylococcus spp. from various fast foods sold in Mymensingh and to determine their antibiogram. Materials and Methods: Overall, 60 samples of fast foods sold in different restaurants were screened by culture, biochemical tests, and polymerase chain reaction (PCR) to isolate and iden¬tify Staphylococcus spp., followed by employing of disk diffusion method to determine their anti¬biotic resistance patterns. Results: Among these 60 samples, 8 [13.33%, 95% confidence interval (CI): 6.91%24.17%] were positive for Staphylococcus spp. by cultural and biochemical properties. By PCR, four (6.67%, 95% CI: 2.62%15.93%) isolates were confirmed as Staphylococcus aureus by targeting nuc gene. Additionally, all the S. aureus isolates were coagulase-positive. By antibiogram profiles, all the Staphylococcus isolates exhibited resistance to azithromycin and erythromycin (95% CI: 67.56% 100.00%), and frequently resistance to cefixime (75%, 95% CI: 40.93%95.56%), ampicillin (50%, 95% CI: 21.52%78.48%), and amoxicillin (37.5%, 95% CI: 13.68%69.43%); moderate to lower resistance was found against cefotaxime, gentamicin, and doxycycline. In addition, all the isolates were sensitive to ciprofloxacin and chloramphenicol. Interestingly, 75% (6/8; 95% CI: 40.93% 95.56%) isolates were multidrug-resistant (MDR) in nature. Furthermore, the indices of multiple antibiotic resistance (MAR) were ranged from 0.2 to 0.6. Conclusion: This study revealed that fast foods sold in restaurants were contaminated with MDR and MAR Staphylococcus isolates, having potential public health significance. Objective: This study was carried out to estimate the sex ratio, maturity size, gonadosomatic index (GSI), and peak breeding season of mud crabs. Materials and Methods: Samples were collected randomly from the estuary and river of the study area. Sampling was carried out monthly from April to September at every full moon during one high tide. A total number of 240 specimens were sampled, where 53 individuals were hermaphrodite. The crabs were shifted alive to the biology and histology lab for detailed biological study. Sex was determined. Male and female sex ratio and breeding season were also investigated. Results: The male:female ratio was 1:0.96 and the ovarian development was categorized into five stages based on internal observations, viz. immature (stage I), underdeveloped (stage II), early developed (stage III), late developed (stage IV), and mature (stage V). The maturity percentages were 37%, 19%, 13%, 11%, and 20%, respectively. 50% maturation was estimated at 82.36 mm inter¬nal carapace width (ICW). The highest mean GSI value was 7.97 ± 3.03. The mature stage was found in all the working periods. This shows that females have activated ovaries in all the working months, and the species are continuous breeders. A higher frequency of vitellogenic ovary and higher GSI value were found in September. The maximum GSI value was found in the size group 7079 mm. Conclusion: The study shows that the capture from the wild sources of mud crabs without any regulation can threaten the population structure. The capture of female mud crabs should be more than 82.36 mm ICW, which will help conserve and protect young crabs. [J Adv Vet Anim Res 2021; 8(1.000): 44-50]

Objective: Ivermectin (IVM) could be used effectively to treat bovine cutaneous papillomatosis, a widespread viral skin disease that causes major economic losses in cattle. This study aimed to evaluate the regression of bovine cutaneous papillomas induced by IVM by estimating oxidative stress markers, besides clinicopathological and hematological findings. Materials and Methods: Twenty naturally infected animals with cutaneous papillomatosis were chosen randomly and diagnosed clinically and histopathologically. All the infected animals were divided into groups: Group I (n = 10), which received no treatment and was considered the con¬trol group. In Group II (n = 10), the animals were subcutaneously injected at 0.2 mg/kg of IVM 2 weeks apart during the 90-day experimental period. Papilloma regression was tracked clinically, papilloma biopsies were taken for histopathological analysis, and blood samples were taken for hematological and oxidative parameter testing. Results: From the 15th to 45th day after receiving IVM, papillomas began to fade. Necrotic areas, ulcerations, and lymphocytic infiltration were found in the histopathological studies, besides a decrease in papilloma epidermal proliferation. total erythrocytes count, packed cell volume, total leucocytes count, and lymphocytes values were increased significantly, and a large decrease in glutathione peroxidase and glutathione reduced was identified as signs of IVM-induced oxidative stress. Conclusion: IVM has oxidative and immunostimulatory properties, and it can be used against cutaneous papillomatosis. [J Adv Vet Anim Res 2021; 8(3.000): 370-377]

Objective: This case report highlights the first detection of Trypanosoma lewisi, a blood protozoan parasite found in an urban rat in Kota Bharu, Kelantan. Materials and Methods: Rat trapping was carried out within the Kota Bharu vicinity near a local wet market. A total of 38 rats were captured and subjected to peripheral blood smearing using Giemsa stain. Positive rats were sent for histopathological analysis for the evaluation of the organ samples. Results: The presence of trypanosomes was found in one sample from a blood smear. This was connected to a histological lesion on kidney tissues, which revealed a high concentration of trypanosomes. Additionally, the positive sample was confirmed as T. lewisi based on molecular diagnosis via polymerase chain reaction and subsequent sequencing and phylogenetic analysis. Conclusions: This finding serves as a baseline for further surveillance on T. lewisi population among urban rats in Kelantan and possible zoonotic transmission to humans.
