Pharmacokinetics of combination antiparasitic drug preparation for dogs and cats in the form of spot-on solution Полный текст

Objective: The object of the study was to examine the major pharmacokinetic parameters after a single application of a complex drug preparation for veterinary use based on fipronil, praziquantel, moxidectin, and pyriproxyfen in cats and dogs. Materials and Methods: For dogs, the drug preparation was administered spot-on solution in the following dosage of active pharmaceutical substances: fipronil 27.0 mg/kg body weight (bwt), praziquantel 10.8 mg/kg bwt, moxidectin 6.75 mg/kg bwt, and pyriproxyfen 5.4 mg/kg bwt; for cats, the dosage was the following: fipronil 43.2 mg/kg bwt, praziquantel 17.28 mg/kg bwt, mox-idectin 4.32 mg/kg bwt, and pyriproxyfen 8.64 mg/kg bwt. The blood samples were taken from dogs and cats. The principle of the method for determining praziquantel, trans-4-hydroxyprazi-quantel, pyriproxyfen, and fipronil in serum samples was chromatographed in a high-pressure liquid chromatograph with detection by means of a mass-spectrometric detector. The moxidectin content of the blood was detected by high-performance liquid chromatography. Results: The drug preparation active substances: praziquantel, fipronil, and moxidectin are absorbed into the blood of dogs and cats. The penetration of praziquantel into the systemic circulation and further into organs and tissues was proved. After topical administration, moxidectin is absorbed and distributed systemically and is slowly removed from the plasma, which manifests itself in detectable concentrations of moxidectin in the blood for 1 month. Conclusion: The present results of pharmacokinetic investigations may promote to the determination of effective therapy strategy and prophylaxis of parasitic diseases in dogs and cats. Objective: The purpose of our research was to evaluate the effect of mechanochemical technology on the efficacy of supramolecular complex of fenbendazole (SMCF) with polyvinylpyrrolidone (PVP) polymer against some helminthosis of animals. Materials and Methods: The SMCF samples with PVP were synthesized using a solid-state mechanochemical technology in activators of impact-abrading type and their physicochemical properties were analyzed. The efficacy of SMCF was studied on the laboratory model of Hymenolepis nana and Trichinella spiralis infection of mice and helminthosis of sheep. Results: In the trials conducted on laboratory models, the supramolecular complex showed 93.94% and 98.56 % efficacy at the dose of 1 mg/kg of body weight (b/w), while the substance of fenbendazole showed 7.97% and 8.33% efficacy at the same dose. A high efficacy (>94%) of the SMCF was revealed at the dose of 2.0 mg/kg of b/w at oral administration against nematodes in naturally infected sheep by the results of the fecal examination, while the substance of fenbendazole was active at the dose of 5.0 mg/kg at single oral administration. Moreover, the SMCF demonstrated 97.37% efficacy at the dose of 2 mg/kg against Moniezia spp. infection of sheep. Physicochemical studies confirmed the increase in solubility of the complex, reducing of particle sizes, amorphization of fenbendazole substance, and incorporating it with micelles of PVP. Conclusion: According to the results, supramolecular complex of fenbendazole with PVP was more active than the basic substance of fenbendazole and its anthelmintic properties were expanded. Now-a-days, various types of antibiotics are being used worldwide in veterinary sector indiscrim¬inately for promotion of growth and treatment of the livestock. Significant portions of antibiotics are released through milk of dairy animals unaltered and exert serious harmful effects on human health. This review evaluates and compare researches on antibiotic residues in milk in published literatures from Pubmed, CrossRef, CAB direct, DOAJ, JournalTOCs, AGRICOLA, ScientificGate, Electronic Journals Library, CAB abstracts, Global Health Databases, Global Impact Factor, Google Scholar, Park Directory of Open Access Journals, BanglaJOL and ISC E-Journals. Antibiotics resi¬due in milk was first detected in 60s and then with an increasing trend with highest after 2,000 (188). The highest no. of works, 49 (21.87%) were accomplished in China, followed by Spain, 30 (13.39%); Germany, 11 (4.91%); and USA, 10 (4.46%). Continent-wise highest researches are published from Europe, 105 (46.88%), followed by Asia, 77 (34.38%); South America, 18 (8.04%); North America, 16 (7.14%); and Africa, 8 (3.57%). For detection, Bovine milk sample is mostly used, 193 (86.16%), followed by ovine, 19 (8.48%); and caprine, 14 (6.25%). Acetonitrile was used in maximum cases (77) for processing the samples. Chromatographic technique was the highest, 115 (51.34%) for detection. Residue of β-lactam group have been detected mostly 133 (36.54%), followed by tetracyclines, 51 (14.01%); fluoroquinolones, 49 (13.46%); sulfonamides, 46 (12.64%); and aminoglycosides, 38 (10.44%). This review observe that antibiotics residues are more common in milk samples that are being manifested in increasing researches on antibiotic detection and measures should adopt to cease this residue. Objective: The objective of this study was to identify the multi-drug resistance (MDR) Klebsiella sp. from mastitis milk samples. Materials and Methods: In the current research, 48 clinical mastitis milk samples were collected from Rangpur division, Bangladesh. Confirmation of bovine mastitis (BM) was done by the California Mastitis Test (CMT). All the CMT positive isolates were subjected for the identification of Klebsiella sp. using through a series of cultural and biochemical tests. MDR Klebsiella sp. isolates were determined using the disk diffusion method, and minimum inhibitory zones were measured by following Clinical and Laboratory Standards Institute. MDR patterns of the isolates were also subjected to study by using housefly (Musca domestica). Results: Among the isolates, 62.5% (n = 30/48) revealed the presence of Klebsiella sp. Eight antimicrobial agents including Amoxicillin, Novobiocin, Erythromycin, Vancomycin, Cephradine, Tetracycline, Bacitracin, Methicillin, and housefly (M. domestica) showed complete resistance to Klebsiella sp. On the other hand, Chloramphenicol, Gentamicin, Ciprofloxacin, Azithromycin, Norfloxacin, Levofloxacin, and Nalidixic acid showed sensitivity. Conclusion: This study helps to treat BM with effective antibiotics and helps in an epidemiological study in Rangpur division as well as helps to create public health awareness. Objective: The study was performed to determine the level of knowledge, attitude, and prac¬tice among different professionals toward zoonotic diseases in selected coastal areas of Barguna district, Bangladesh. Materials and Methods: A total of 485 respondents were randomly selected from different upazilas (sub-districts) of Barguna district, Bangladesh. A questionnaire-based survey was conducted to collect data about awareness of zoonosis, hygienic management, zoonotic disease transmission from different species of domestic animals and consumption of their products, consciousness on management of pet animals, disease transmission from wild animals, effects of natural disaster on zoonosis, and extension works on zoonosis provided by government or private sector. Results: Based on the level of knowledge of the different respondents, meat (43.92%) is the prime way for transmission of zoonotic disease followed by egg (18.14%) and milk (13.61%). The aware¬ness regarding management of pet animals (23.71%) and zoonotic disease from wild animals (26.69%) were more or less similar. It has been observed that 33.81% respondents were conscious about natural disaster causing zoonotic infection. The respondents also mentioned that extension services about zoonotic infection provided by government or private sector was 34.22%. Among all the respondents, the awareness of zoonotic infection was high in employee of livestock depart¬ment followed by employee of health department and teachers. Conclusion: The awareness of zoonoses was high in employee of livestock department followed by the employee of health department, teachers, and other professionals. The present study observed that low educational background of professionals or non-health educated professionals is not conscious on zoonotic diseases. Objective: The purpose of this study was to evaluate the clinical outcome of surgical decompression and rehabilitation therapy in dogs with thoracolumbar intervertebral disk herniation (IVDH). Materials and Methods: After surgery, physiotherapeutic rehabilitation was performed by a combination of electrotherapy, infrared therapy, training for standing, deep tendon reflex, and aquatic treadmill exercise. A total of 186 dogs were selected from the hospital records and included in two groups: the rehabilitated group (RG, n = 96) and non-rehabilitated group (NRG, n = 90). Dogs in each group were subdivided into three groups based on a pre-operative clinical severity grading system and those in grades 24 were included in this study. Post-operative neurologic functions, unassisted standing, walking, and the success rate of both groups were evaluated and compared Results: Overall, 86.46% (83/96) of dogs had a successful neurologic outcome in the RG group, which was significantly (p < 0.01) higher than the NRG group 52.22% (47/90). Interestingly, the success rate differed when the preoperative grading system was considered. The success rates of grades 2, 3, and 4 were 97.14% (34/35), 97.33% (42/45), and 43.75% (7/16), respectively, in the rehabilitated groups, whereas in the non-rehabilitated groups, success rates were 82.35% (28/34), 51.85% (14/27), and 17.24% (5/29), respectively. The differences in success rates among the groups according to grading were 14.79%, 41.48%, and 26.51%, respectively, indicating that the proposed rehabilitation therapy is remarkably advantageous for increasing the success rate. Conclusion: Rehabilitation therapy after surgical decompression of thoracolumbar IVDH improves neurologic functions and increases the success rate, especially when the preoperative pathological condition is severe. Objective: The present study was carried out to appraise the antibiotic resistance and to detect some of the target resistant genes in Escherichia coli (E. coli) isolated from apparently healthy broilers. Materials and Methods: Cloacal swab samples (n = 60) were collected from apparently healthy broilers (n = 60) sold at two different live bird markets (LBMs) of Chattogram, Bangladesh. Isolation and identification of the Escherichia coli was done by the following standard bacteriological techniques followed by biochemical tests. The antibiotic susceptibility of E. coli isolates was determined by the disk diffusion method. The antibiotic resistant genes were detected by polymerase chain reaction (PCR) using specific primers. Results: The overall prevalence of E. coli in broilers was 61.67% (n = 37/60) (95% CI = 4972.93). The antibiogram study showed that the isolates were 100% resistant to ampicillin and tetracycline followed by sulfomethoxazole-trimethoprim (94.59%, n = 35/37) and nalidixic acid (91.89%, n = 34/37). To the contrary, 56.76% (n = 21/37) isolates were sensitive to both ceftriaxone and gentamicin followed by colistin (48.65%, n = 18/37). All of E. coli isolates were multidrug resistant (MDR) and carried blaTEM, tetA, and Sul2 genes. Objective: This study was aimed at evaluating the effect of fermentation using Chrysonillia crassa and Monascus purpureus on nutritional qualities, antioxidant, and antimicrobial activities of the used rice as a poultry feed ingredient. Materials and Methods: The used rice was soaked, steamed, and spread on a tray to cool. Suspension of M. purpureus or C. crassa was inoculated on the steamed used rice, and then mixed thoroughly. Afterward, the mixture was spread out on the tray, which was then covered with an aluminum foil. It was aerobically incubated for 7 and 4 days for the M. purpureus- and C. crassa-inoculated used rice, respectively. Subsequent to sun drying, the fermented used rice was grounded and analyzed. Results: Crude protein and ash contents were higher (p < 0.05) in the used rice fermented with C. crassa or M. purpureus than in the unfermented. Conversely, carbohydrate content was lower (p < 0.05) in the fermented compared with the unfermented. Gross energy and energy from fat were higher (p < 0.05) in the used rice fermented with M. purpureus than the unfermented. Amino acids L-methionine, L-serine, L-glutamic acid, L-valine glycine, L-leucine, L proline, L-threonine, L-histidine, and L-Sistine were higher (p < 0.05) in M. purpureus-fermented used rice than in C. crassa-fermented and the unfermented used rice. However, amino acids L-isoleucine, L- alanine, L-lysine, and L-tryptophan were higher (p < 0.05) in the used rice fermented with both C. crassa and M. purpureus, compared with the unfermented. L-tyrosine content was higher (p < 0.05) in M. purpureus-fermented used rice than in the unfermented. Furthermore, the antioxidant activities of the fermented products were higher (p < 0.05) than that of the unfermented. In addition, the antimicrobial activities of the fermented products against Staphylococcus aureus were higher (p < 0.05) than that of the unfermented used rice. Conclusion: In conclusion, the used rice fermented using C. crassa and M. purpureus improved the nutritional quality, as well as the antioxidant and antimicrobial activities of the products. Objective: Parasitic infestation is a major cause of losses in livestock production in tropical regions. A cross-sectional study was conducted to determine the prevalence of Gastro-intestinal (GI) parasites of dromedary camel (Camelus dromedarius) and fat-tailed sheep (dhumba), and the prevalence of hemoparasites in camel from Dhaka, Bangladesh. Materials and Methods: A total of 87 fecal samples (32 dhumba and 55 camel) and 55 camel blood samples were collected during SeptemberOctober 2015. Fecal samples were examined by direct smear, sedimentation method, flotation technique, and McMaster technique for GI parasite. Giemsa stained blood smears were examined under microscope for hemoparasite detection. Results: 62% camel (n = 34; 95% confidence interval (CI): 47.774.6) were infected with at least one genus of parasite. 15% camel were harboring more than one genus of parasite. The prevalence of GI parasite and hemoparasite in camel were recorded as Trichuris spp. (n = 16; 29%; 95% CI: 17.642.9), Balantidium coli (n = 12; 22%; 95% CI: 11.835.0), Trichostrongylus spp. (n = 7; 13%; 95% CI: 5.324.5), Strongyloides spp. (n = 5; 9%; 95% CI: 3.020.0), Anaplasma spp. (n = 5; 9%; 95% CI: 3.0220.0), Paragonimus spp. (n = 1; 2%; 95% CI: 0.059.7), Schistosoma spp. (n = 1; 2%; 95% CI: 0.059.7), Hymenolepis spp. (n = 1; 2%; 95% CI: 0.059.7), Moniezia spp. (n = 1; 2%; 95% CI: 0.059.7), and Babesia spp. (n = 1; 2%; 95% CI: 0.059.7). Mean EPG feces of camel was 291.76 ± 42.03 with a range of 01,400. Total 59.4% dhumba (n = 19; 95% CI: 4176) were positive for GI parasite, including Trichostrongylus spp. (n = 10; 31.3%; 95% CI: 16.150), Strongyloides spp. (n = 9; 28%; 95% CI: 13.846.8), B. coli(n = 5; 15.6%; 95% CI: 5.332.8), and Trichuris spp. (n = 4; 12.5%; 95% CI: 3.528.9). Conclusions: High percentage of parasitic infestation in camel and dhumba in the present study refers to the necessity of use of anthelmintic for health and production improvement and to prevent zoonotic parasite transmission to animal handler and workers. Objective: House musk shrew (Suncus murinus), a small experimental animal with low body fat, may be a possible model for human lipodystrophy. Leptin is an adipocyte-derived hormone thought to have an important role in the pathophysiology of lipodystrophy. The objectives of this study were to clarify the structure and distribution of suncus leptin. Materials and methods: To determine the primary structure of suncus leptin, we cloned the suncus Lep cDNA using the rapid amplification of cDNA ends method. The obtained amino acid (aa) sequence was compared with other mammals and the protein structure prediction was performed. Results: The suncus Lep cDNA encodes 170 aa. The putative suncus leptin precursor has a predicted signal peptide of 21 aa, and the mature leptin comprises 149 aa. The mature leptin is 75%82% homologous to that of other species. Insertion of the three aa, VPQ, not seen in other mammals was found. This VPQ insertion is thought to be due to a nucleotide insertion of nine bases by slippage-like microindels. The predicted 3D structure of suncus leptin exhibited a typical four a-helix structure, however, the VPQ region protruded compared with human leptin. LepmRNA expression was observed only in white and brown adipose tissues. Conclusion: This study revealed the structure and distribution of suncus leptin. Because the addition of VPQ, which is not found in other mammals, was observed, suncus leptin attracts attention to its physiological action, and to the possibility of being a model of human lipodystrophy.
