A qualitative investigation of animal-to-human brucellosis knowledge among health workers in Kajiado North Sub-County, Kenya Полный текст

Animal-to-human brucellosis transmission methods in Kajiado NorthSub - Count y, Kenya are inadequately documented. The Sub- County’s main livelihood is livestock-keeping, putting it at risk to brucellosis infections. Objective of this study was to determine the knowledge of brucellosis among animal and humanhealth workers. These workers were stationed in Kajiado North Sub-County and were the key informants of this study from whom qualitative data was collected. The data was analysed based on emerging themes, e.g. brucellosis significance in the community, livestock birthing and milk hygiene. Results revealed that brucellosis a significant disease in the community, informants were knowledgeable on the need for pre-purchase livestock examination, the need for proper disposal of livestock abortive material and on good milk hygiene measures. However, it was found that not all had adequate knowledge about brucellosis transmission through livestock birthing. The results indicated that measures to control and prevent brucellosis transmission to man, and between animals were inadequate as health workers were unable to adequately advise the community. It is recommendedthat health workers should be trained on all methods of brucellosis transmission, especially transmission during livestock births.

Septicaemic pasteurellosis is a fatal, sometimes epidemic, bacterial disease of domestic and wild animals including deer, bison, elk, and pronghorn antelope caused by Pasteurella multocida. This is the case report of septicaemic pasteurellosisin a captive sambar deer. The carcass was sent from Royal Endurance Stable, Bachok, Kelantan to the Kota Bharu RegionalVeterinary Laboratory for post-mortem. Gross examination of organs was followed by collection of specimens from lung, kidney,liver, spleen and heart for histopathology and bacterial examination. Pooled organ samples with rumen content were collected and sent to the nearest Chemistry Department for investigation. For histology, the liver, lung, spleen, kidney, and heart specimens were fixed in 10% neutral formalin, and routinely embedded in paraffin. Fivemicrometer sections were stained with H&E. Other tests such as worm and ectoparasiteidentification were conducted to identify the parasites. Post-mortem lesions revealed generalised haemorrhage in the organs.Pasteurella multocida serogroup B and E. coli were isolated from multiple tissues of the animal. Histological examination alsorevealed severe congestion and haemorhage of multiple tissues with infiltration of the inflammatory cells. The most likely mode of transmission of these bacteria is through an infected wound and into the bloodstream, thereby causing severe septicemia and death to the animal.

Infectious diseases of livestockare a major threat to global animal health and welfare and their effective control is crucialfor agronomic health, for safeguarding and securing national and international food supplies and for alleviating rural povertyin developing countries. Some devastating livestock diseases are endemic in many parts of the world and threats from old and new pathogens continue to emerge, with changes to global climate, agricultural practices and demography presenting conditions that are especially favourable for the spread of arthropod-borne diseases into new geographical areas. Zoonotic infections that are transmissible either directly or indirectly between animals and humans are on the increase and pose significant additional threats to human health and the current pandemic status of new influenza A (H1N1) is a topical example of the challenge presented by zoonotic viruses (Tomley and Shirley, 2009). Malaysia, being one of the members of the World Organisation forAnimal Health (OIE) which is responsible for setting standards for control of animal diseases. For year 2017, the list included 116 animal diseases, infections and infestations, many of which are zoonotic in nature. As such, this paper discusses the commonzoonotic infections diagnosed in the five Regional Veterinary Laboratories which are spread across the country and entrustedto carry out diagnostic tests to aid in the treatment and control of animal diseases. A total of almost half a million samples weretested comprising more than a million tests to help the Department of Veterinary Services control and eradicate economically important diseases to safeguard the animal population. Of these, zoonotic diseases comprise a small but significant entity which needs careful attention (Chandrawathani et al., 2017) Dora Tan (1981) reported that among the many zoonotic diseases prevalent in Malaysia, are leptospirosis, rabies, influenza, Japanese encephalitis, toxoplasmosis,ornithosis, Q fever and monkeypox which have been investigated at the lnstitute for Medical Research, Kuala Lumpur. The regional laboratories have full capability to conduct tests to confirm parasitic, viral and bacterial infections except for rabies andavian influenza, which was diagnosed in the Veterinary Research Institute. However, preliminary tests for avian influenza wascarried out in regional laboratories.

This study aimed at determining the current knowledge, attitude, and practices (KAP) of layer and pig farmers to antimicrobial usage and its consequences. Description of such KAP could provide insights useful for promoting the rational use of antimicrobials in livestock. From May 2014 to February 2016, a survey involving 251 respondents in Chiang Mai, Lamphun, and Chonburi provinces, Thailand, was conducted by using a validated questionnaire. More than half (51.0%) of the respondents incorrectly believed that antimicrobial drug efficacy could not be reduced by using sub-recommended dosages, 61.2% had misconceptions about non-therapeutic antimicrobial use, and 66.9% inaccurately felt that antimicrobials were also effective for virus and fungi. Over half (50.6 - 55.2%) did not see the need to follow instructions or advice of veterinarians. Moreover, only 10.4% regularly relied on responses to drugs sensitivity tests when evaluating the effectiveness of antimicrobials. Overall, assessment of KAP regarding antimicrobial usage indicated that the majority of respondents had low levels of knowledge of antimicrobials, neutral rather than positive attitudes, and employed poor practices in the use of antimicrobials. The results indicate improvements in KAP could be helpful in developing more effective interventions by farmers, reduce antibiotic usage, and slow the growth of antimicrobial resistance.

Canine coronavirus is a single-stranded RNA virus that causes enteritis in dogs of any age. Coronaviral enteritis is seldom definitively diagnosed, since it is usually much less severe than many other types of enteritis and is self-limiting. Conventional diagnostics for the canine coronaviral enteritis such as polymerase chain reaction (PCR), virus isolation, and electron microscopic examination are inappropriate for small animal clinics due to the complicated experimental processes involved. Therefore, a commercially available lateral flow test kit based on chromatographic immunoassay techniques was tested to evaluate its performance as a first-line diagnostic test kit that could be used in clinics. The coronavirus antigen test kit detected canine coronavirus-infected dogs with 93.1% sensitivity and 97.5% specificity. The detection limit of the test kit was between 1.97 × 104 /mL and 9.85 × 103 /mL for samples with a 2- fold serial dilution from 1.25 × 106 TCID50 (TCID50, 50% tissue culture infectious dose). Additionally, the test kit had no cross-reactivity with canine parvovirus, distemper virus, or Escherichia coli. Overall, the commercially available test kit showed good diagnostic performance in a clinical setting, with results similar to those from PCR, confirming their potential for convenient and accurate use in small animal clinics.

The efficacy of using a bacteriophage (phage) to control Flavobacterium psychrophilum (F. psychrophilum) infection of ayu (Plecoglossus altivelis altivelis) was evaluated in this study. Intramuscular challenge failed to induce sufficient infection levels; therefore, a newly designed net-scratch challenge method was also used to induce bacterial infection. Administration of phage PFpW-3 in F. psychrophilum-infected ayu showed notable protective effects, increased survival rates and mean times to death. Additionally, the fate of inoculated bacteria and phage in ayu were investigated. Our results suggest that the phage PFpW-3 could be considered an alternative biocontrol agent against F. psychrophilum infections in ayu culture.

The helicase genes and hypervariable regions (HVRs) of three avian hepatitis E viruses (HEVs) detected at three different farms were sequenced and characterized. Two isolates (DW-L and GI-B2) were classified as genotype 2 and one isolate (GR-B) was classified as genotype 1. A phylogenetic tree, based on the helicase gene and HVR nucleotide sequences, revealed the newly detected viruses and other avian HEVs were classified similarly. Unlike previously reported avian HEVs, the DW-L isolate detected in broiler breeders with characteristic lesions of avian HEV had no prolinerich motif in its HVR, suggesting that the proline-rich motif is non-essential for viral replication and infection.

An 8-year-old, castrated, male Rottweiler was referred for evaluation of chronic right thoracic limb lameness and a progressively growing mass surrounding the right elbow joint. On admission, the dog’s general health was good, without abnormalities detected on physical examination. The dog was diagnosed with periarticular histiocytic sarcoma. Although draining lymph nodes and lung metastases were suspected, palliative amputation was performed. Localized histiocytic sarcomas, with destructive lesions involving multiple bones of a joint and periarticular soft-tissue masses, are uncommon in dogs. This case report presents clinical findings, imaging characteristics, and histopathologic and immunohistochemical features of a periarticular joint histiocytic sarcoma.

The present study observed the effects of a green tea (Camellia sinensis) flower extract (GTFE) on melanin synthesis in B16-F10 melanoma cells. GTFE exhibited antioxidant activity on 2,2-diphenyl-1-picrylhydrazyl and inhibited mushroom tyrosinase activity in a dose-dependent manner. Furthermore, GTFE significantly diminished alphamelanocyte stimulating hormone (alpha-MSH) stimulated cellular melanin content and tyrosinase activity throughout the concentration range evaluated. Based on RNA sequencing analysis, differential gene expression patterns observed in alpha-MSH stimulated B16-F10 melanoma cells were normalized by the addition of GTFE. In particular, the expression levels of melanoregulin and tyrosinase genes which are key regulating genes in melanin synthesis were up-regulated by 3.5 and 3 fold respectively by alpha-MSH, and were normalized to control levels by the addition of GTFE. The results suggest that GTFE inhibits melanin synthesis in alpha-MSH stimulated B16-F10 melanoma cells by normalizing expression of genes that are essential for melanin synthesis. Overall, the results suggest that GTFE could be applied in the development of a whitening agent for the treatment of dermal hyperpigmentation.

Outbreaks of porcine reproductive and respiratory syndrome virus (PRRSV) in vaccinated sow herds from occurrence to stabilization were monitored and analyzed in terms of serology and reproductive performance. Three different conventional pig farms experienced severe reproductive failures with the introduction of a type 1 PRRSV. These farms had adopted mass vaccination of sows using a type 2 PRRSV modified live vaccine (MLV). Therefore, to control the type 1 PRRSV, an alternative vaccination program utilizing both type 1 and type 2 MLV was undertaken. Following whole herd vaccinations with both types of MLV, successful stabilization of PRRS outbreaks was identified based on serological data (no viremia and downward trends in ELISA antibody titers in both sows and suckling piglets) and recovery of reproductive performance. Additionally, through comparison of the reproductive parameters between outbreak and non-outbreak periods, it was identified that PRRSV significantly affected the farrowing rate and the number of suckling piglets per litter at all three pig farms. Comparison of reproductive parameters between periods when the different vaccination strategies were applied revealed that the number of piglets born in total and born dead per litter were significantly increased after the introduction of the type 1 PRRS MLV.
