When creating Triticum aestivum L. varieties in V. M. Remeslo Myronivka Institute of Wheat, intervarietal crossing was used as the basis for classical method of hybridization. Parental pairs were selected according to environmental and geographical principles. Crossing of lines selected by certain adaptive characteristics among themselves and with the local breeding varieties are widely used. The method of induced mutagenesis is one of the main factor of the evolutionary process which increases hundreds of times the frequency of altered forms occurrence. Thermal mutagenesis – selection of winter forms from spring varieties – is based on low temperature effect on spring wheat varieties (it was developed by V. M. Remeslo, the member of the Academy of Sciences). Method of intervarietal selection in winter wheat breeding provides creation of new varieties of this crop through mass selection of the best plants from local populations. At present, 10 varieties of winter wheat are included into the State Register of Plant Varieties Suitable for Dissemination in Ukraine, one variety – into the State Register of Breeding Achievements of the Russian Federation, seven varieties are passing state quality testing in these countries.

У результаті гібридизації сор­ту ‘Глесія’ із самозапиленою лінією шостого покоління сорту ‘Золотоніські 15’ і наступного добору на закріплення ознак високої продуктивності та поліпшення якісних ознак коноплепродукції створено новий сорт ‘Артеміда’. Сорт належить до середньостиглої групи: вегетаційний період до настання фази технологічної стиглості становить 94 доби, біологічної – 118 діб. За вирощування на зеленець забезпечує високі показники врожайності волокна (2,56 т/га) та виходу всього волокна (30,4%, зокрема довгого волокна – 27,6%). У разі вирощування на двобічне використання сорт ‘Артеміда’ формує вищі, порівняно із сортом-стандартом ‘Гляна’, урожаї волокна (2,01 т/га) та насіння (1,29 т/га) з підвищеним умістом в останньому олії (36,8%). При цьому він істотно поступається за висотою рослин, що позитивно для збирання насіння зернозбиральним комбайном. Аналіз відповідності емпіричного й теоретичного розподілу ознаки вмісту олії в насінні елітних рослин сорту ‘Артеміда’ свідчить про її високу стабільність. Примітною особливістю сорту є формування дружних сходів та інтенсивний ріст рослин на початку вегетації, що сприяє зменшенню забур’яненості посівів.Висновки. ‘Артеміда’ – новий сорт конопель універсального напряму господарського використання. Належить до середньоєвропейського еколого-географічного типу, хоча і створений у результаті сортолінійної гібридизації різних типів з наступним поліпшувальним селекційним добором за ознаками продуктивності. Характеризується пов­ною відсутністю канабіноїдних сполук, підвищеним умістом олії, поліпшеним її жирнокислотним складом та високою якістю волокна. Рекомендується для вирощування з метою отримання волокна й насіння. Завдяки високій потенційній урожайності продукції є конкурентоздатним на ринку промислових конопель. | Purpose. Creation of industrial hemp variety of multiple purposes with the absence of cannabinoid compounds, high oil content in seeds and fiber quality.Methods. Bree­ding (self-pollination, varietal-linear hybridization in the conditions of a vegetation house, selection), field, laboratory, instrumental-technological assessment of fiber quality, mathematical statistics.Results. The ‘Artemida’ variety was created as a result of hybridization of the ‘Hlesiia’ variety with the self-pollinated line of the sixth generation of the ‘Zolotoniski 15’ variety and selection for stabilization of high productivity traits and improvement of quality trait of hemp production. The variety belongs to the medium-ripe group; the growing season is 94 days before the phase of technological maturity and 118 days before the phase of biological maturity. When grown for fiber, the variety had a higher fiber yield (2.56 t/ha), the yield of all fiber (30.4% and including long fiber 27.6%). When grown for fiber and seeds, the plant is significantly inferior in height, which is positive for harves­ting seeds with a combine harvester, has a significantly higher seed yield (1.29 t/ha), oil content (36.8%) and fiber yield (2.01 t/ha) in comparison with the standard of the varie­ty ‘Hliana’. The analysis of the correspondence between the empirical and theoretical distribution of such a trait as the oil content in the seeds of elite plants of the ‘Artemida’ variety indicates its high stability. A notable feature of the variety is the formation of friendly seedlings and intensive plant growth at the beginning of the growing season, which helps to reduce the weediness of crops. Conclusions. The new variety of hemp ‘Artemida’ of multiple purposes belongs to the Central European ecological and geographical type, although created as a result of varietal-linear hybridization of different types with selection on the basis of productivity, is characte­rized by complete absence of cannabinoid compounds, high oil content and fiber quality. The variety is recommended for growing for fiber and seeds. Due to its high yield potential, it is competitive in the industrial hemp market.

Цель. Создать сорта пшеницы мягкой яровой при использовании отечественных и зарубежных сортов, носителей пшенично-ржаной транслокации (ПРТ). Методы. Полевые, лабораторные, статистические. Результаты. Перспективным направлением селекции, позволяющим существенно улучшить генофонд пшеницы мягкой яровой, является создание сортов с ПРТ 1АL.1RS. Представлено описание многолетних селекционных исследований, проведенных в Мироновском институте пшеницы имени В. Н. Ремесло НААН Украины (МИП), по получению сортов пшеницы мягкой з ПРТ 1АL.1RS. В качестве исходного селекционного материала использовали сорта и линии отечественной и зарубежной селекции, в частности сорт ‘ТАМ 107’ (США) с ПРТ 1АL.1RS. В ходе исполнения научной селекционной программы были созданы сорта пшеницы мягкой озимой ‘Експромт’ (1АL.1RS), мягкой яровой ‘Струна миронівська’ (1АL.1RS) и сорт ‘МІП Соломія’ (1АL.1RS). Как свидетельствуют результаты анализа спектра запасных белков, сор­та ‘Струна миронівська’ и ‘МІП Соломія’ в своих геномах имеют ПРТ 1АL.1RS. ‘МІП Соломія’ унаследовал ее от сорта пшеницы мягкой яровой ‘Струна миронівська’. Новый сорт пшеницы мягкой яровой ‘МІП Соломія’ характеризуется высокой урожайностью, адаптивностью, устойчивостью к мучнистой росе, фузариозу колоса, твердой головне, высокими показателями качества зерна. Выводы. Путем многолетних простых парных скрещиваний иностранных сортов и отечественного селекционного материала с использованием метода педигри созданы сорта пшеницы мягкой с ПРТ 1АL.1RS. Сорт пшеницы мягкой яровой ‘МІП Соломія’, переданный в 2017 р. на Государственное сортоиспытание, характеризуется высокой урожайностью, устойчивостью к полеганию, низким процентом поражения грибными болезнями, устойчивостью к твердой головне, адаптивностью. Наличие пшенично-ржаной транслокации у сортов яровой пшеницы мироновской селекции ‘Струна миронівська’ и ‘МІП Соломія’ подтверждено при помощи анализа спектра запасных белков при электрофорезе в полиакриламидном геле. | Мета. Creation of bread spring wheat varieties using domestic and foreign varieties as carriers of wheat-rye translocation. Меthods. Field, laboratory, statistical. Results. The creation of varieties with wheat-rye translocation 1AL.1RS is the perspective direction of breeding, which allows to significantly improve the gene pool of bread spring wheat. The paper covers outputs of long-term breeding studies conducted at the V. M. Remeslo Myronivka Institute of Wheat of the National Academy of Agrarian Sciences of Ukraine (MIW) for obtaining bread wheat varieties with wheat-rye translocation 1AL.1RS. As the initial breeding material varieties and lines of domestic and foreign breeding were used, in particular, the ‘TAM 107’ variety (USA) with wheat-rye translocation 1AL.1RS. During realization of the scientific breeding program, the bread winter wheat variety ‘Ekspromt’ (1AL.1RS), as well as the bread spring wheat varieties ‘Struna myronivska’ (1AL.1RS) and ‘MІP Solomіia’ (1AL.1RS) have been created. As evidenced by the analysis of the spectrum of storage proteins, the varieties ‘Struna myronivska’ and ‘MIP Solomiіa’ contain wheat-rye translocation 1AL.1RS in their genomes. The variety ‘MIP Solomiіa’ inherited it from the variety of bread spring wheat ‘Struna myronivska’. The bread spring wheat new variety ‘MІP Solomiia’ is characterized by high yielding capacity, adaptability, resistance to powdery mildew, fusarium head blight, common bunt and high indices of grain quality. Conclusions. By long-term single crossing foreign varieties and domestic breeding material when using the pedigree method, bread wheat varieties with wheat-rye translocation 1AL.1RS have been created. The bread spring wheat variety ‘MІP Solomiia’ submitted for the State Variety Testing in 2017, is characterized by high yielding capa­city, lodging resistance, low incidence of fungal diseases, resistance to common bunt, adaptability. The presence of wheat-rye translocation in spring wheats varieties of Myronivka breeding ‘Struna myronivska’ and ‘MІP Solomіia’ has been confirmed by analyzing the spectrum of storage proteins during polyacrylamide gel electrophoresis.

Purpose. To reveal breeding and genetic peculiarities of modern spring barley varieties for the “number of grains per main ear” trait and identify genetic sources of increased combining ability for involving in hybridization. Methods. Investigations were carried out at the V. M. Remeslo Myronivka Institute of Wheat of NAAS of Ukraine. Modern varieties of domestic (‘Virazh’, ‘Talisman Myronivskyi’, ‘Komandor’) and foreign (‘KWS Aliciana’, ‘KWS Bambina’, ‘Zhana’, ‘Explorer’) breeding were involved in crossing for a full (7´7) diallel scheme. Parents and F1 were studied in field conditions during 2014–2016. Results. The analysis of variance of combining ability has shown a significant advantage in varying of general combining ability (GCA) effects. The mean square of specific combining ability (SCA) was significantly less than the GCA, but reliable throughout the years. The reciprocal effect was reliable only in 2014. Stably high effects of GCA during all years of investigations were noted in the varieties ‘KWS Aliciana’ (1.18–1.62) and ‘Virazh’ (1.33–1.48). The variety ‘KWS Bambina’ was characterized by lower but reliable positive effects of GCA (0.43–0.99) as compared to mentioned above. Non-allelic gene interaction was not found, that allowed to calculate the basic parameters of genetic variation. During all years of investigations, dominant effects of genes (H1 and H2) prevailed over the additive (D) ones in phenotypic expression of grain number per main ear. Mean degree of dominance in the experiment (H1/D) has shown overdominance. The same pattern was also distinctive for the index of mean degree of dominance in the loci . The dominance was reliably directed. Dominant effects of genes increased grain content, and recessive ones reduced it. At least 3–4 genes (groups of genes) have been revealed which determined the effects of dominance. At the same time, recessive genes (F<0) or gene effects were prevailed quantitatively in the varieties investigated. A high coefficient of heritability in broad sense (H2 = 0.98) has shown a significant determination of phenotypic variability with genetic factors. The coefficient of heritability in narrow sense (h2 = 0.66–0.68) confirmed that despite the advantage of dominant effects over the additive ones, the contribution of the latter was also significant. Conclusions. The prevalence of dominant effects of genes in the phenotypic expression of the number of grains per main ear causes the need for sufficient sample size of hybrid material and points to the expediency of conducting a more “rigid” selection for phenotype in later generations. At the same time, the considerable contribution of additive effects and high values of heritability indices give reason to predict the efficiency of selections aimed at increasing the trait in created hybrid material. The varieties ‘Virazh’, ‘KWS Aliciana’, ‘KWS Bambina’ should be used as effective genetic sources to increase grain content in combination breeding.

Purpose. To assess introduced samples of drought-resistant species of perennial grasses, select a promising parent material and create on its base high-yielding varie­ ies with economic characters. Methods. Field experiment, laboratory testing. Results. The results of studies on introduction and breeding were given aimed to improve drought tolerance of non-traditional perennial grasses under the conditions of the Right-Bank Forest-Steppe zone of Ukraine. Based on the selected parent material, varieties were created by the use of hybridization and ecotype breeding methods and then entered into the State Register of plant varieties suitable for dissemination in Ukraine, among them: intermediate wheatgrass (Elytrigia intermedia (Host) Nevski) – ‘Hors’, crested wheatgrass (Agropyron cristatum (L.) Gaertn.) – ‘Petrivskyi’; meadow brome (Bromus riparia Rehm.) – ‘Boian’; slender wheatgrass (Roegneria trachycaulon (Link) Nevski) – ‘Co­umb’. As compared with conventional, relatively drought-tolerant species of smooth brome (Bromopsis inermis (Leyss.) Holub) – ‘Mars’, increment of dry matter content of these species in the extreme drought conditions of 2011 was increased by 1,52–3,73 t/ha. Under more sufficient moistening conditions of 2012, slender wheatgrass ‘Columb’ was at the level of the сheck variety in terms of this indicator. Other varieties exceeded it by 1.44–3.22 t/ha. The data was given including seed productivity and sowing quality indicators, after-ripening duration and economic fitness of seeds. Conclusions. The use of the recommended varieties of drought-resistant species of perennial grasses as part of grass mixtures will increase significantly the productivity of grasslands and pastures in the current context of climate change.

The results of studies on introduction and breeding were given aimed to improve drought tolerance of non-traditional perennial grasses under the conditions of the Right-Bank Forest-Steppe zone of Ukraine. Based on the selected parent material, varieties were created by the use of hybridization and ecotype breeding methods and then entered into the State Register of plant varieties suitable for dissemination in Ukraine, among them: intermediate wheatgrass (Elytrigia intermedia (Host) Nevski) – ‘Hors’, crested wheatgrass (Agropyron cristatum (L.) Gaertn.) – ‘Petrivskyi’; meadow brome (Bromus riparia Rehm.) – ‘Boian’; slender wheatgrass (Roegneria trachycaulon (Link) Nevski) – ‘Co­umb’. As compared with conventional, relatively drought-tolerant species of smooth brome (Bromopsis inermis (Leyss.) Holub) – ‘Mars’, increment of dry matter content of these species in the extreme drought conditions of 2011 was increased by 1,52–3,73 t/ha. Under more sufficient moistening conditions of 2012, slender wheatgrass ‘Columb’ was at the level of the сheck variety in terms of this indicator. Other varieties exceeded it by 1.44–3.22 t/ha. The data was given including seed productivity and sowing quality indicators, after-ripening duration and economic fitness of seeds. Conclusions. The use of the recommended varieties of drought-resistant species of perennial grasses as part of grass mixtures will increase significantly the productivity of grasslands and pastures in the current context of climate change.

Purpose. Creation of experimental sugar beet hybrid combinations of high sugar yield values and defining gene­tic determination of their heterotic effect. Methods. Diallel crossing and topcrossing, genetic analysis of quantitative traits. Results. The authors have studied the frequency of occurrence of sugar beet heterotic hybrid combinations for «sugar yield» trait created on the basis of two pollinator lines to be genetically valuable for productivity elements, CMS lines and single-cross sterile hybrids with the use of diallel and topcrossing system of controlled hybridization. The share of parental components’ effect and their interaction in CMS hybrids variability for productivity was determined. Expediency of heterotic forecasting based on high combining ability lines was substabtiated. Promising high-yielding sugar beet combinations were selected that exceeded the group standard by 4.1–16.3%. Conclusions. The theory of genetic balance by M. V. Turbin was confirmed. Such hybrids as [CMS 5OT 4]MGP 1 (116.3%), [CMS 1OT 2]MGP 1 (112.5%) and [CMS 3OT 5]MGP 1 (113.2%) were recognized as the best for their productivity, MGP 1 and MGP 2 lines – as the best for their combining ability.

The article presents the results of studies to determine the parameters of the polymerase chain reaction (PCR) in order to develop a multiplex system for identification of the structural elements of the design of transgenic gene cp 4 epsps, which provides tole­rance to glyphosate. For amplicon target DNA sequences, the following values of temperature conditions of PCR were determined: step 1 (initial denaturation) 95 °C – 3 min; step 2 (specific reaction products accumulation): denaturation 95 °C – 45 s; hybridization of primers 55 °C – 50 s; elongation 72 °C – 1 min; number of cycles – 40; step 3 (final elongation) 72 °C – 6 min. A series of PCR were carried out for the purpose of selecting the optimal amount of DNA matrix for efficient estimate of transgenic sugar beet plants for the presence of specific sequences. Conclusions. To identify transgenic glyphosate-tolerant sugar beet, it is advisable to determine 35S promoter and gene cp 4 epsps in individual genotypes. It was found that during the selection of temperature parameters of multiplex reaction a 5 °C rise in primer hybridization temperature did not affect the identification of gene als that allowed to include specific primers for determination of this sequence as an internal control. Based on the results of test multiplex reactions, concentrations of dNTPs and Mg2+ ions were determined that allowed to exclude the possibility of non-specific fragments and false-negative results. The optimum amount of matrix DNA (100–150 ng) for an efficient estimate of transgenic sugar beet plants for the presence of specific sequences was determined. Obtained results allowed to develop a multiplex test system for identification of transgenic glyphosate-to­lerant sugar beet which can be used for simultaneous determination of the 35S promoter, cp 4 epsps gene and als gene as an internal reaction control.
