Biomarker responses reveal that food quality affects cadmium exposure in the soil collembolan Folsomia candida

Food quality affects the food consumption rate, flux through the gut, and exposure to contaminants in animals. This study evaluated the effects of food quality on cadmium exposure in the soil collembolan Folsomia candida. Animals were exposed to constant concentrations of cadmium for 38 h via artificial food consisting of an agar medium with various concentrations of sugar (glucose), total nutrients (baker's yeast), or fungal odour (1-octen-3-ol). The expression of the gene encoding a deduced metallothionein-like motif containing protein was used as a biomarker of cadmium exposure. Glucose concentrations of 2% or higher reduced the expression levels of the biomarker. Within the range of 0.1–8% yeast, medium concentrations led to higher biomarker levels. At high concentrations of 1-octen-3-ol (2000 mg/l), feeding and the biomarker response were reduced. These results suggest that even at equivalent cadmium concentrations, food quality affects cadmium exposure by altering food consumption rates.

In this study, we isolated and characterized Shewanella putrefaciens from shellfish harvested from the West Sea in Korea. For the initial isolation of S. putrefaciens, LB agar plates supplemented with ferrous sulfate and sodium thiosulfate were inoculated with shellfish homogenates, incubated for 24h, and then black colonies were selected. Gram-negative and catalase-positive colonies were subsequently confirmed by PCR assays and API 20E kit test system. The Shewanella-specific 16S rRNA and gyrB genes were used to design S. putrefaciens-specific PCR primers. From 6 species of shellfish tested, 24 S. putrefaciens strains were isolated. These 24 isolates had the following profiles of resistance against 16 antibiotics: all the isolates were resistant to cephalothin and vancomycin and 95.8% were resistant to ampicillin. Here, we report the isolation of S. putrefaciens from shellfish and our results point to a new possible route for exposing healthy individuals to S. putrefaciens.

Ratios of sucrose-negative to sucrose-positive vibrios on TCBS agar (suc−/suc+) indicate the abundance of potential human pathogenic non-cholera vibrios in coastal mariculture environments of the Lingayen Gulf (Philippines. In guts of adult maricultured milkfish (Chanos chanos) of suc− vibrios reached extreme peak values ranging between 2 and 545millionperg wet weight. Suc− vibrios outnumbered suc+ vibrios in anoxic sediments, too, and were rarely predominant in coastal waters or in oxidized sediments. Suc−/suc+ ratios in sediments increased toward the mariculture areas with distance from the open sea at decreasing redox potentials. There is circumstantial evidence that suc− vibrios can be dispersed from mariculture areas to adjacent environments including coral reefs. An immediate human health risk by pathogenic Vibrio species is discounted, since milkfish guts contained mainly members of the Enterovibrio group. A representative isolate of these contained proteolytic and other virulence factors, but no genes encoding toxins characteristic of clinical Vibrio species.

A total of 69 bacteria were isolated from crude oil enrichments of the polychaetes Megalomma claparedei, Sabella spallanzanii and Branchiomma luctuosum, and screened for biosurfactant (BS) production by conventional methods. Potential BS-producers (30 isolates) were primarily selected due to the production of both interesting spots on thin layer chromatography (TLC) plates and highly stable emulsions (E24⩾50%). Only few strains grew on cetyltrimethylammonium bromide and blood agar plates, indicating the probable production of anionic surfactants. The 16S rRNA gene sequencing revealed that selected isolates mainly belonged to the CFB group of Bacteroidetes, followed by Gammaproteobacteria and Alphaproteobacteria.A number of BS-producers belonged to genera (i.e., Cellulophaga, Cobetia, Cohaesibacter, Idiomarina, Pseudovibrio and Thalassospira) that have been never reported as able to produce BSs, even if they have been previously detected in hydrocarbon-enriched samples. Our results suggest that filter-feeding Polychaetes could represent a novel and yet unexplored source of biosurfactant-producing bacteria.

Aquaculture wastewaters, with oleic acid (C18:1 ω9) as the most representative contaminant fatty acid, were used as inoculum to perform enrichment cultures in mineral medium in the presence of oleic acid as the sole carbon and energy source, allowing isolation of four bacterial strains named DG1a, DG2a, DG1b and DG2b. 16S rRNA gene sequencing analysis assigned the four isolates to the species Pseudomonas aeruginosa. Among the isolates, P. aeruginosa strain DG2a showed degradation of fatty acids, including oleic acid (C18:1 ω9). The hydrophobicity features were investigated in strain DG2a, and a constitutive hydrophobicity in the bacterial cells was highlighted. The capability to produce biosurfactants by cells of the bacterial strain P. aeruginosa strain DG2a was evidenced both in the presence of oleic acid and of aquaculture wastewaters by revealing emulsifying activity, oil spreading tests, haemolytic and cetyltrimethylammonium bromide agar tests. Bacterial cultures containing raw biosurfactant were added to native wastewaters, showing a depletion of the oleic acid content. The use of the isolated bacterial strain P. aeruginosa strain DG2a and of the produced biosurfactant in bioremediation of aquaculture wastewaters is proposed, and the valorization of aquaculture wastewaters as raw material for biosurfactant production by using the isolate is moreover suggested.

Phosphogypsum (PG) is a by-product of the phosphorus–fertiliser industry and represents an environmental concern since it contains pollutants such as cadmium (Cd). We have recently shown that the overexpression of a proton pump gene (TaVP1) in transgenic tobacco (Nicotiana tabacum) led to an enhanced Cd tolerance and accumulation. The aim of this study was to evaluate the potential of transgenic Arabidopsis thaliana plants harbouring the TaVP1 gene to phytoremediate phosphogypsum. A pot experiment was carried out under greenhouse conditions. Transgenic A. thaliana plants harbouring the TaVP1 gene were grown on various substrates containing phosphogypsum (0, 25, 50 and 100 %) for 40 days. At the end of the growth period, we examined the growth (germination, root length, fresh weight) and physiological parameters (chlorophyll and protein contents, catalase activity and proteolysis) as well as the cadmium, Mg, Ca, and P contents of the A. thaliana plants. In order to evaluate Cd tolerance of the A. thaliana lines harbouring the TaVP1 gene, an in vitro experiment was also carried out. One week-old seedlings were transferred to Murashige and Skoog agar plates containing various concentrations of cadmium; the germination, total leaf area and root length were determined. The growth and physiological parameters of all A. thaliana plants were significantly altered by PG. The germination capacity, root growth and biomass production of wild-type (WT) plants were more severely inhibited by PG compared with the TaVP1 transgenic A. thaliana lines. In addition, TaVP1 transgenic A. thaliana plants maintained a higher antioxidant capacity than the WT. Interestingly, elemental analysis of leaf material derived from plants grown on PG revealed that the transgenic A. thaliana line accumulated up to ten times more Cd than WT. Despite its higher Cd content, the transgenic A. thaliana line performed better than the WT counterpart. In vitro evaluation of Cd tolerance showed that TaVP1 transgenic A. thaliana lines were more Cd-tolerant than the WT plants. These results suggested that ectopic expression of a vacuolar proton pump in A. thaliana plants can lead to various biotechnological applications including the phytoremediation of industrial wastes.
