Prediction of biogas and pressure from rumen fermentation using plant extracts to enhance biodigestibility and mitigate biogases

Improving digestibility, fermentation characteristics, and reducing greenhouse biogases to protect the environment without the use of synthetic materials is an important goal of modern-day farming and nutritionist. Plant extracts are capable of solving these. This is due to the digestive enzymes and the bioactive components capable of performing antimicrobial functions inherent in these plants. This study was aimed to investigate the effect of standard maize substrate treated with selected herbs and spices extracts on ruminal environmental biogas production and pressure during fermentation via biogas production technique. Herbs (Azadirachta indica leaves (T1), Moringa oleifera leaves (T2), Ocimum gratissimum leaves (T3) and spices (Allium sativum bulb (T4), Zingiber officinale rhizome (T5)) were harvested, air dried, and milled using standard procedures. Methanolic extracts of the herbs and spices were prepared and used as additives at different concentrations (50, 100, and 150 μL) to the maize substrate for in vitro biogas production. Data were analyzed using regression analysis. There were significant (P < 0.05) differences across all the treatments on the volume and pressure of biogas. The pressure and volume of biogas when compared with the levels tested showed differences (P < 0.05) across all the treatments for the prediction of volume from pressure of biogas. The pressure and volume of gas produced in vitro increased (P < 0.05) and biogases decreased (P < 0.05) by the substrate treated with herbs and spices but for the drum stick leaves which was similar for the levels of concentration tested. This means that the level tested had a pronounced mitigation effect on pressure of biogas and volume of biogas produced. It was concluded that the herb and spice extracts have the potential to improve rumen fermentation and reduce the production of biogases in ruminant diet.

Preservation or curing of hides/skins is performed as the primary step of leather processing to conserve them from putrefaction. Normally preservation is carried out using common salt (NaCl), which is discharged in the soak liquor contributing to ~ 70%, of total dissolved solids (TDS) load of entire leather manufacturing. In an attempt to reduce the TDS and chlorides, phyto-based preservation using garlic peel (Allium sativum) and white onion peel (Allium cepa) was carried out. Different concentrations of salt in combination with garlic peel and white onion peel were applied on freshly flayed goat skins based on its green weight and compared to control (40% salt). Sensory evaluation of the preserved skin was done by assessing different parameters like hair slip, putrefaction and odour. Estimation of hydroxyproline (HP) release, moisture content and microbial load were carried out at regular intervals. Skins that remained in good condition for 14 days were further processed into leather and properties were examined which were found comparable to the conventionally cured skins. Hence, this cleaner curing technique helps in reducing the TDS and chlorides in the effluent, thus controlling the pollution caused by tanneries through sustainable leather processing.

In the present study, salt-tolerant strains, Dietzia sp. HRJ2, Corynebacterium variabile HRJ4, Dietzia cinnamea HRJ5 and Bacillus tequilensis HRJ6 were isolated from the Dagang oil field, China. These strains degraded n-alkanes and polycyclic aromatic hydrocarbons (PAHs) aerobically from heavy crude oil (HCO) in an experiment at 37 °C and 140 rpm. The GC/MS investigation for degradation of different chain lengths of n-alkanes (C8–C40) by individual strains showed the highest degradation of C8–C19 (HRJ5), C20–C30 (HRJ4) and C31–C40 (HRJ5), respectively. Moreover, degradation of 16 PAHs with individual strains demonstrated that the bicyclic and pentacyclic aromatic hydrocarbons (AHs) were mostly degraded by HRJ5, tricyclic and tetracyclic AHs by HRJ6 and hexacyclic AHs by HRJ2. However, the highest degradation of total petroleum hydrocarbons (TPHs), total saturated hydrocarbons (TSH), total aromatic hydrocarbons (TAH), n-alkanes (C8–C40) and 16 PAHs was achieved by a four-membered consortium (HRJ2 + 4 + 5 + 6) within 12 days, with the predominance of HRJ4 and HRJ6 strains which was confirmed by denaturing gradient gel electrophoresis. The abundance of alkB and nah genes responsible for catabolism of n-alkanes and PAHs was quantified using the qPCR. Maximum copy numbers of genes were observed in HRJ2 + 4 + 5 + 6 consortium (gene copies l⁻¹) 2.53 × 10⁴ (alkB) and 3.47 × 10³ (nah) at 12 days, which corresponded to higher degradation rates of petroleum hydrocarbons. The superoxide dismutase (SOD) (total SOD (T-SOD), Cu²⁺Zn²⁺-SOD), catalase (CAT) and ascorbate peroxidase (APX) activities in Allium sativum and Triticum aestivum were lower in the HRJ2 + 4 + 5 + 6-treated HCO as compared to the plantlets exposed directly to HCO. The present results revealed the effective degradation of HCO-contaminated saline medium using the microbial consortium having greater metabolic diversity.

The present investigation was undertaken to evaluate the ameliorative activity of Allium sativum against lead-induced oxidative stress in the brain, liver, and kidney of male rats. Four groups of male Wistar strain rats (100–120 g) were taken: group 1 received 1000 mg/L sodium acetate and group 2 was given 1000 mg/L lead acetate through drinking water for 2 weeks. Group 3 and 4 were treated with 250 mg/kg body weight/day of A. sativum and 500 mg/kg body weight/day of A. sativum, respectively, by oral intubation for a period of 2 weeks along with lead acetate. The rats were sacrificed after treatment and the brain, liver, and kidney were isolated on ice. In the brain, four important regions namely the hippocampus, cerebellum, cerebral cortex, and brain stem were separated and used for the present investigation. Blood was also drawn by cardiac puncture and preserved in heparinized vials at 4 °C for estimation of delta-aminolevulinic acid dehydratase (ALAD) activity. The results showed a significant (p < 0.05) increase in reactive oxygen species (ROS), lipid peroxidation products (LPP), total protein carbonyl content (TPCC), and lead in the selected brain regions, liver, and kidney of lead-exposed group compared with their respective controls. Blood delta-ALAD activity showed a significant (p < 0.05) decrease in the lead-exposed rats. However, the concomitant administration of A. sativum resulted in tissue-specific recovery of oxidative stress parameters namely ROS, LPP, and TPCC. A. sativum treatment also restored the blood delta-ALAD activity back to control. Overall, our results indicate that A. sativum administration could be an effective antioxidant treatment strategy for lead-induced oxidative insult.

Formulation and characterization of garlic (Allium sativum L.) essential oil nanoemulsion and its acaricidal activity on eriophyid olive mites (Acari: Eriophyidae)

Green and nanoacaricides including essential oil (EO) nanoemulsions are important compounds to provide new, active, safe acaricides and lead to improvement of avoiding the risk of synthetic acaricides. This study was carried out for the first time on eriophyid mites to develop nanoemulsion of garlic essential oil by ultrasonic emulsification and evaluate its acaricidal activity against the two eriophyid olive mites Aceria oleae Nalepa and Tegolophus hassani (Keifer). Acute toxicity of nanoemulsion was also studied on male rats. Garlic EO was analyzed by gas chromatography–mass spectrometry (GC-MS), and the major compounds were diallyl sulfide (8.6%), diallyl disulfide (28.36%), dimethyl tetrasulfide (15.26%), trisulfide,di-2-propenyl (10.41%), and tetrasulfide,di-2-propenyl (9.67%). Garlic oil nanoemulsion with droplet size 93.4 nm was formulated by ultrasonic emulsification for 35 min. Emulsification time and oil and surfactant ratio correlated to the emulsion droplet size and stability. The formulated nanoemulsion showed high acaricidal activity against injurious eriophyid mites with LC₅₀ 298.225 and 309.634 μg/ml, respectively. No signs of nanoemulsion toxicity were noted in treating rats; thus, it may be considered non-toxic to mammals. Stability of garlic oil nanoemulsion, high acaricidal activity, and the absence of organic toxic solvents make the formulation that may be a possible acaricidal product. Results suggest the possibility of developing suitable natural nanoacaricide from garlic oil.
