Uptake of waterborne Cd, Co, Mn and Zn was determined in laboratory experiments using radiotracer techniques (109Cd, 57Co, 54Mn and 65Zn). Labelled Zn was mainly accumulated in the digestive gland (65%) and Co in kidneys (81%); Cd and Mn were similarly distributed in digestive gland and gills. In a complementary field study, Ag, As, Cd, Co, Cr, Cu, Fe, Mn, Ni, and Zn were analysed in scallops collected at two stations showing different contamination levels. Digestive gland and kidneys displayed the highest concentrations. Ag, As, Cd, and Fe differed in soft tissues from the two stations, suggesting that Comptopallium radula could be a valuable local biomonitor species for these elements. Low Mn and Zn concentrations found in kidneys suggest that their content in calcium-phosphate concretions differs from the other pectinids. Preliminary risk considerations suggest that As would be the only element potentially leading to exposure of concern for seafood consumers. This study investigates metal accumulation behaviour in the tropical scallop Comptopallium radula and preliminary risk assessment for consumers.

Microplastics are emerging contaminants in the marine environment. They enter the ocean in a variety of sizes and shapes, with plastic microfiber being the prevalent form in seawater and in the guts of biota. Most of the laboratory experiments on microplastics has been performed with spheres, so knowledge on the interactions of microfibers and marine organisms is limited. In this study we examined the ingestion of microfibers by the sea anemone Aiptasia pallida using three different types of polymers: nylon, polyester and polypropylene. The polymers were offered to both symbiotic (with algal symbionts) and bleached (without algal symbionts) anemones. The polymers were introduced either alone or mixed with brine shrimp homogenate. We observed a higher percentage of nylon ingestion compared to the other polymers when plastic was offered in the absence of shrimp. In contrast, we observed over 80% of the anemones taking up all types of polymers when the plastics were offered in the presence of shrimp. Retention time differed significantly between symbiotic and bleached anemones with faster egestion in symbiotic anemones. Our results suggest that ingestion of microfibers by sea anemones is dependent both on the type of polymers and on the presence of chemical cues of prey in seawater. The decreased ability of bleached anemones to reject plastic microfiber indicates that the susceptibility of anthozoans to plastic pollution is exacerbated by previous exposure to other stressors. This is particularly concerning given that coral reef ecosystems are facing increases in the frequency and intensity of bleaching events due to ocean warming.

The study of the uptake and distribution of elements in marine environments is of great interest for understanding their pathways and accumulation. Here, we investigated in laboratory experiments the accumulation behavior of Cd in the sea anemone Bunodosoma caissarum and the mussel Perna perna. Specimens were incubated with isotopically enriched 116Cd in aquariums. Cd concentrations in the seawater and in the tissues of B. caissarum and P. perna were followed by inductively coupled plasma-mass spectrometry (ICP-MS) by means of isotope dilution analysis. Bioconcentration factors for B. caissarum and P. perna exposed to 0.9μg·L−1 of 116Cd were determined to be 80.5 and 850, respectively. P. perna specimens exposed to 4.5μg·L−1 of 116Cd reached 530. Cytosolic proteins associated with Cd from the tissues were extracted and further analyzed by size-exclusion chromatography coupled to ICP-MS. Cd accumulation could be detected in both organisms ranging from high-molecular to low-molecular species.

Mercury (Hg) has a complex biogeochemical cycle in aquatic environments. Its most toxic form, methylmercury (MeHg), is produced by microorganisms. This study investigated how the sea anemone Bunodosoma caissarum affects Hg distribution, methylation and volatilization in laboratory model systems. 203Hg was added to microcosms and its distribution in seawater, specimens and air was periodically measured by gamma spectrometry. MeHg was measured by liquid scintillation. After the uptake period, specimens had a bioconcentration factor of 70 and in microcosms with and without B. caissarum, respectively 0.05% and 0.32% of the initial spike was found as MeHg. After depuration, MeHg in specimens ranged from 0.2% to 2.4% of total Hg. Microcosms with B. caissarum had higher Hg volatilization (58%) than controls (17%), possibly due to Hg2+ reduction mediated by microorganisms associated with its tissues and mucus secretions. Marine organisms and their associated microbiota may play a role in Hg and MeHg cycling.

In this study, the physiology of symbiotic ‘peacock-tail’ shrimp Ancylocaris brevicarpalis and its host ‘Haddon's carpet’ sea anemone Stichodactyla haddoni were tested under lowered pH (7.7) and control (8.1) conditions. The biochemical responses such as digestive enzyme (AP), organic acids (lactate and succinate), oxidative damages (MDA), antioxidants metabolites/enzymes (ASC, GSH, SOD, CAT, APX, GPX, GR, POX, and PHOX), and detoxification enzyme (GST) were measured. The AP showed insignificantly reduced values in both the organisms in lowered pH conditions compared to control indicating the effect of abiotic stress. The hierarchical clustering analysis indicated low MDA in sea anemone can be explained by higher POX, APX, GR, ASC, and GSH levels compared to shrimps. However, the detoxification enzyme GST showed less activity in sea anemones compared to shrimps. The results suggest that A. brevicarpalis and sea anemone S. haddoni may have deleterious effects when exposed to short-term acidification stress.

Benthic surveys of mesophotic reefs in the Gulf of Mexico post Deepwater Horizon (DWH) showed that Swiftia exserta octocorals exhibited significantly more injury than in years before the spill. To determine the vulnerability of S. exserta to oil and dispersants, 96h toxicity assays of surrogate DWH oil water-accommodated fractions (WAF), Corexit® 9500 dispersant, and the combination of both (CEWAF) were conducted in the laboratory. Fragment mortality occurred within 48h for some fragments in the dispersant-alone and oil-dispersant treatments, while the WAF group remained relatively unaffected. The 96h LC50 values were 70.27mg/L for Corexit-alone and 41.04mg/L for Corexit in CEWAF. This study provides new information on octocoral sensitivity to toxins, and indicates that combinations of oil and dispersants are more toxic to octocorals than exposure to oil alone. These results have important implications for the assessment of effects of the DWH spill on deep-water organisms.

Polychlorinated biphenyls (PCBs) are persistent and bioaccumulable organic compounds. The occurrence of PCBs was assessed in two populations of the intertidal sea anemone Bunodosoma zamponii living under different anthropogenic strains. One location was in vicinity to a wastewater plant (Las Delicias, LD), and the other was a preserved location chosen as a reference site (Punta Cantera, PC). Anemone populations were sampled 4 times (spring, summer, autumn and winter) throughout a year, in addition to seawater and sediment from those areas. PCB loadings ranged from 2.89 to 79.41 ng L⁻¹ in seawater samples and from 0.07 to 6.61 ng g⁻¹ dry weight in sediment samples. Total PCB levels ranged from 0.22 to 14.94 and 2.79 to 24.69 ng g⁻¹ wet weight in anemones from PC and LD, respectively. PCBs concentrations in anemones from LD were significantly greater than PC during spring, summer and winter. The congeners 18 and 44 prevailed in seawater samples, 44 and 52 in sediment and 18 and 132+153 in anemones. Redundancy analysis integrated PCB levels from all matrixes and bolstered contrast between sampling sites. Seasonality was also a relevant factor since during winter PCB loading was greater in sediment and anemone samples, coincident with the rainiest season. Disparity between sites could be due to LD’s proximity to the wastewater plant, effect of littoral drift direction and/or asymmetries in anemones physiological condition.

The sea anemone Exaiptasia pallida (formally Aiptasia pulchella) has been identified as a valuable test species for tropical marine ecotoxicology. Here, the sensitivities of newly developed endpoints for E. pallida to two unidentified whole effluents were compared to a standard suite of temperate toxicity test species and endpoints that are commonly used in toxicological risk assessments for tropical marine environments. For whole effluent 1 (WE1), a 96-h lethal concentration 50 % (LC50) of 40 (95 % confidence intervals, 30–54) % v/v and a 12-day LC50 of 12 (9–15) % v/v were estimated for E. pallida, exhibiting a significantly higher sensitivity than standard sub-lethal endpoints in Allorchestes compressa (96-h effective concentration 50 % (EC50) of >100 % v/v for immobilisation) and Hormosira banksii (72-h EC50 of >100 % v/v for germination), and a similar sensitivity to Mytilus edulis galloprovincialis larval development with a 48-h LC50 of 29 (28–30) % v/v. Sub-lethal effects of whole effluent 2 (WE2) on E. pallida pedal lacerate development resulted in an 8-day EC50 of 7 (3–11) % v/v, demonstrating comparable sensitivity of this endpoint to standardised sub-lethal endpoints in H. banksii (72-h EC50 of 11 (10–11) % v/v for germination), M. edulis galloprovincialis (48-h EC50 for larval development of 12 (9–14) % v/v) and Heliocidaris tuberculata (1-h EC50 of 13 (12–14) % v/v for fertilisation; 72-h EC50 of 26 (25–27) % v/v for larval development) and a significantly higher sensitivity than A. compressa immobilisation (96-h EC50 of >100 % v/v). The sensitivity of E. pallida compared to a standard test species suite highlights the value in standardising the newly developed toxicity test methods for inclusion in routine toxicological risk assessment of complex whole effluents. Importantly, this species provides an additional taxonomic group to the test species that are currently available for tropical marine ecotoxicology and, being a cnidarian, may represent important tropical marine environments including coral reefs.

Polycyclic aromatic hydrocarbons (PAHs) are the most persistent organic pollutants in worldwide aquatic environments. The extensive isolation of genes responsive to PAH pollution in soft coral (Scleronephthya gracillimum) is described herein. Soft coral colonies were exposed to 100 μg/L of a standard mixture of PAHs. Gene candidates with transcript levels that changed in response to PAH exposure were identified by differential display polymerase chain reaction (DD-PCR). There were 37 types of candidate genes identified, of which 20 were upregulated in expression and 17 were downregulated. The functions of the genes identified included oxidative stress response, ribosomal structure maintenance, molecular chaperone activity, protein kinase activation and tumorigenesis, defense mechanisms, transcription, and other biological responses. mRNA quantification was carried out using real-time quantitative PCR in eight selected genes: cytosolic malate dehydrogenase, protein disulfide isomerase, ribosomal protein L6, ral guanine nucleotide dissociation stimulator-like 1, poly(ADP-ribose) polymerase 4, peptidylglycine α-hydroxylating monooxygenase, a disintegrin and metalloproteinase (ADAM) metallopeptidase protein, and eukaryotic initiation factor 4 gamma 3. Changes in transcript levels were consistent with DD-PCR results. The gene candidates isolated in this study were differentially expressed and therefore have potential as molecular biomarkers for understanding coral responses to environmental stressors.
