Removal of the antiviral agent oseltamivir and its biological activity by oxidative processes

The antiviral agent oseltamivir acid (OA, the active metabolite of Tamiflu®) may occur at high concentrations in wastewater during pandemic influenza events. To eliminate OA and its antiviral activity from wastewater, ozonation and advanced oxidation processes were investigated. For circumneutral pH, kinetic measurements yielded second-order rate constants of 1.7 ± 0.1 × 10⁵ and 4.7 ± 0.2 × 10⁹ M⁻¹ s⁻¹ for the reaction of OA with ozone and hydroxyl radical, respectively. During the degradation of OA by both oxidants, the antiviral activity of the treated aqueous solutions was measured by inhibition of neuraminidase activity of two different viral strains. A transient, moderate (two-fold) increase in antiviral activity was observed in solutions treated up to a level of 50% OA transformation, while for higher degrees of transformation the activity corresponded to that caused exclusively by OA. OA was efficiently removed by ozonation in a wastewater treatment plant effluent, suggesting that ozonation can be applied to remove OA from wastewater.

Recently, an outbreak of a novel human coronavirus which is referred to as severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) (COVID-19) by the World Health Organization (WHO) was identified in Wuhan, China. To help combat the pandemic, a systematic review (SR) was performed to collect all available studies concerning inactivation methods, environmental survival, and control and prevention strategies. A comprehensive literature survey yielded 42 eligible studies which included in the SR. The results confirmed that the WHO recommended two alcohol-based hand rub formulations (ethanol 70–95% and 2-propanol 70–100%) had an efficient virucidal activity in less than 60 s by more and equal 4 log₁₀ (≥ 99.99) approximately and could be used for disinfection in public health and health-care facilities. The findings indicated that SARS-CoV-1 and SARS-CoV-2 can survive under different environmental conditions between 4 and 72 h approximately. The results also demonstrate that temperature and relative humidity are important factors in the survival of SARS-CoV-2. The main strategies recommended by the WHO to avoid contracting SARS-CoV-2 are hand washing several times in the day and maintaining social distancing with others. It is important to note that the more studies require addressing, the more possible airborne transmission due to the survival of SARS-CoV-2 in aerosols for 3 h approximately. We hope that the results of the present SR can help researchers, health decision-makers, policy-makers, and people for understanding and taking the proper behavior to control and prevent further spread of SARS-CoV-2.

Phycoremediation of swine wastewater is a promising treatment since it efficiently removes nutrients and contaminants and, simultaneously, its biomass can be harvested and used to obtain a wide range of valuable compounds and metabolites. In this context, biomass microalgae were investigated for the phycoremediation of swine wastewater, and biomass extracts for its virucidal effect against enveloped and non-enveloped viruses. Microalgae were cultivated in a pilot scale bioreactor fed with swine wastewater as the growth substrate. Hexane, dichloromethane, and methanol were used to obtain the microalgae extracts. Extracts were tested for virucidal potential against HSV-1 and HAdV-5. Virucidal assays were conducted at temperatures that emulate environmental conditions (21 °C) and body temperature (37 °C). The maximum production of microalgae biomass reached a concentration of 318.5 ± 23.6 mgDW L⁻¹. The results showed that phycoremediation removed 100% of ammonia-N and phosphate-P, with rates (k₁) of 0.218 ± 0.013 and 0.501 ± 0.038 (day⁻¹), respectively. All microalgae extract reduced 100% of the infectious capacity of HSV-1. The microalgae extracts with dichloromethane and methanol showed inhibition activities at the lowest concentration (3.125 µg mL⁻¹). Virucidal assays against HAdV-5 using microalgae extract of hexane and methanol inhibited the infectious capacity of the virus by 70% at all concentrations tested at 37 °C. At a concentration of 12.5 µg mL⁻¹, the dichloromethane microalgae extract reduced 50–80% of the infectious capacity of HAdV-5, also at 37 °C. Overall, the results suggest that the microalgae can be an attractive source of feedstock biomass for the exploration of alternative virucidal compounds.

The greatest challenge the world is facing today is to win the battle against COVID-19 pandemic as soon as possible. Until a vaccine is available, personal protection, social distancing, and disinfection are the main tools against SARS-CoV-2. Although it is quite infectious, the SARS-CoV-2 virus itself is an enveloped virus that is relatively fragile because its protective fatty layer is sensitive to heat, ultraviolet radiation, and certain chemicals. However, heat and liquid treatments can damage some materials, and ultraviolet light is not efficient in shaded areas, so other disinfection alternatives are required to allow safe re-utilization of materials and spaces. As of this writing, evidences are still accumulating for the use of ozone gas as a disinfectant for sanitary materials and ambient disinfection in indoor areas. This paper reviews the most relevant results of virus disinfection by the application of gaseous ozone. The review covers disinfection treatments of both air and surfaces carried out in different volumes, which varies from small boxes and controlled chambers to larger rooms, as a base to develop future ozone protocols against COVID-19. Published papers have been critically analyzed to evaluate trends in the required ozone dosages, as a function of relative humidity (RH), contact time, and viral strains. The data have been classified depending on the disinfection objective and the volume and type of the experimental set-up. Based on these data, conservative dosages and times to inactivate the SARS-CoV-2 are estimated. In small chambers, 10–20 mg ozone/m³ over 10 to 50 min can be sufficient to significantly reduce the virus load of personal protection equipment. In large rooms, 30 to 50 mg ozone/m³ would be required for treatments of 20–30 min. Maximum antiviral activity of ozone is achieved at high humidity, while the same ozone concentrations under low RH could result inefficient. At these ozone levels, safety protocols must be strictly followed. These data can be used for reducing significantly the viral load although for assuring a safe disinfection, the effective dosages under different conditions need to be confirmed with experimental data.

Antiviral drugs have been recently recognized as one of the emerging contaminants in the environment. These are discharged after therapeutic use through human excretion. Effluent containing high concentration of antiviral drugs discharged from production facilities is also a cause of concern to nearby aquatic bodies. There is an increased interest in their removal because they are highly bioactive. Some antiviral drugs are resistant to conventional methods of degradation, and there is a risk of development of antiviral resistance in humans and animals if exposed repeatedly for long periods. To date, the potential human, animal, and ecological risks associated with the discharge of these antiviral compounds to the environment are not well documented. This study presents a brief summary on occurrence, ecotoxicological risks, and physicochemical properties of antiviral drugs in the environment. The needs regarding removal, disposal, and treatment of antiviral drugs are also addressed.

With increasing environment pollution and bacterial transmitted viral diseases globally, the development of new, effective, and low-cost materials/strategies is the current major challenge. To combat with this alarming problem, three new multi-functional and thermally stable SnO₂NPs@ZIF-8 composites (NC1, NC2, and NC3) were synthesized by a facile and sustainable approach involving in situ encapsulation of SnO₂NPs (150, 300, and 500 μL suspension in methanol) within zeolitic imidazole framework at room temperature. The morphology and crystallinity of ZIF-8 remained unchanged upon the proper encapsulation of SnO₂NPs in its matrix. Herein, for the first time, the antiviral potential of ZIF-8 and SnO₂NPs@ZIF-8 against chikungunya virus is reported by investigating their cytotoxicity against Vero cell line (employing MTT ((3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide)) assay). The maximum non-toxic doses were 0.04 mg mL⁻¹ for ZIF-8 and SnO₂NPs@ZIF-8 and 0.1 mg mL⁻¹ for SnO₂NPs. Further, NC1 exhibited (based on plaque assay) reduction in viral load/titers up to > 80% during post-treatment and > 50% during pre-treatment, greater than that of ZIF-8 and SnO₂NPs due to synergistic effect. Further, NC1 (10 mg) exhibited enhanced photocatalytic efficiency (≥ 96%) for degradation of methylene blue (0.5 × 10⁻⁵ M) at pH ˃ 7.0. The probable mechanism for their antiviral activity and photocatalytic activity has been discussed. The multi-functional composites can effectively be used to reduce water pollution and as remedy for mosquito/bacterial transmitted viral diseases.

Immunity represents a trait common to all living organisms, and animals and plants share some similarities. Therefore, in susceptible host plants, complex defence machinery may be stimulated by elicitors. Among these, chitosan deserves particular attention because of its proved efficacy. This survey deals with the antiviral activity of chitosan, focusing on its perception by the plant cell and mechanism of action. Emphasis has been paid to benefits and limitations of this strategy in crop protection, as well as to the potential of chitosan as a promising agent in virus disease control.

Background, aim, and scope Polysaccharides are renewable resources representing an important class of polymeric materials of biotechnological interest, offering a wide variety of potentially useful products to mankind. Exopolysaccharides (EPSs) of microbial origin with a novel functionality, reproducible physico-chemical properties, stable cost and supply, became a better alternative to polysaccharides of algal origin. EPSs are believed to protect bacterial cells from desiccation, heavy metals or other environmental stresses, including hostimmune responses, and to produce biofilms, thus enhancing the cells chances of colonising special ecological niches. One of the most important stress factor is salt stress for microorganisms. The present investigation is aimed to determine correlation between salt resistance and EPS production by three cyanobacterial isolates (Synechocystis sp. BASO444, Synechocystis sp. BASO507 and Synechocystis sp. BASO511). It is also aimed to investigate the effect of salt concentrations on EPS production by cyanobacteria and effect of salt on monosaccharide composition of EPS. Materials and methods Cyanobacterial isolates were identified by 16 S rRNA analysis. Its salt (NaCl) tolerance and association with exopolysaccharides (EPSs) production in three cyanobacterial isolates were investigated. Also, EPS was analysed by HPLC for monomer characterization. Results Increased EPS production was associated with NaCl tolerance. The most tolerant isolate, Synechocystis sp. BASO444, secreted the most EPS (500 mg/L). EPS production by Synechocystis sp. BASO444, Synechocystis sp. BASO507 and Synechocystis sp. BASO511 was investigated following exposure to 0.2 and 0.4 M NaCl. Also, flasks containing medium without NaCl were inoculated in the same manner to serve as controls. The monosaccharide compositions of EPS produced by the three isolates following exposure to 0.2 M NaCl were analysed by HPLC. Control EPS of BASO444 was composed of glucose (97%) and galacturonic acid (3%). The composition of BASO511 (control) was glucose (95%), xylose (4.80%), arabinose (0.13%), glucuronic acid (0.03%) and galacturonic acid (0.04%). However, the composition of BASO507 (control) was glucose (0.98%), xylose (98.00%), arabinose (1.00%), glucuronic acid (0.01%) and galacturonic acid (0.01%). In the presence of 0.2 M NaCl, EPS compositions and ratios of three cyanobacterial isolates changed. Discussion Although hyperproduction of EPS in response to starvation, antiviral activity, thickening agent and cosmetic industry for product formulations has been reported for cyanobacteria, the effect of NaCl on EPS production in cyanobacteria is not a popular area of study. There are no clear reports correlating EPS production and NaCl tolerance. The gap in the data about the effect of NaCl on cyanobacterial EPS production was filled by this investigation, and the results of our study have important implications in both the industrial and environmental arenas. Conclusions Our results indicate that 1) exposure to elevated concentrations of NaCl affects the composition of EPS produced by Synechocystis sp. BASO444, Synechocystis sp. BASO507 and Synechocystis sp. BASO511, and 2) there is a correlation between NaCl tolerance and EPS production in some cyanobacteria. Recommendations and perspectives Differences in the monosaccharide composition and ratios of EPS may promote NaCl tolerance in these microorganisms. As well, these alternative composition polysaccharides may be important for industrial applications.

Propolis is a natural product of bees with biological activities that are mainly associated with bee type and geographic origin. Propolis extract has been proposed with several applications in environmental health. The ethanol extracts have shown good antimicrobial activity. The association of this technique with ultrasound-assisted extraction has been studied to improve the characteristics of the obtained extracts. Thus, the objective of this work is to verify the antiviral action against two strains of bacteriophages of two extracts of Brazilian propolis (green and red) obtained by conventional extraction and ultrasonic extraction. The activities of the propolis red and green extracts were confirmed by the significant ~3 and ~4.5 Log 10 PFU/mL reduction in the concentrations of the MS2 and Av-08 bacteriophages, respectively. It was found that ultrasound-assisted extraction is comparable to the maceration process and demonstrated the best antiviral activities. Brazilian red propolis was more effective than green propolis in viral reduction in all treatments.
