Chronic exposure to microcystin-LR affected mitochondrial DNA maintenance and caused pathological changes of lung tissue in mice

Microcystin-LR (MC-LR), an important variant of cyanotoxin family, was frequently encountered in the contaminated aquatic environment and taken as a potent hepatotoxin. However, a little was known on the association between the long-term MC-LR exposure and lung damage. In this study, we investigated the changes of the pulmonary histopathology, mitochondrial DNA (mtDNA) integrity and the expression of mtDNA encoded genes in the mice with chronic exposed to MC-LR at different concentrations (1, 5, 10, 20 and 40 μg/L) for 12 months. Our results showed that the long-term and persistent exposure to MC-LR disturbed the balance of redox system, influenced mtDNA stability, changed the expression of mitochondrial genes in the lung cells. Notably, MC-LR exposure influenced the level of inflammatory cytokines and resulted in thickening of the alveolar septa. In conclusion, chronic exposure to MC-LR affected mtDNA maintenance, and caused lung impairment in mice.

Metformin is the most commonly used anti-diabetic drug in the world. When consumed, this unmetabolised pharmaceutical compound is excreted by the body and eventually enters the environment through a variety of pathways. Based on its high consumption and excretion rates, high concentrations of metformin have been detected in influents of wastewater treatment plants. Metformin and its transformation product, guanylurea, are also expected to be present in other aquatic environments based on their physico-chemical properties. Not surprisingly, guanylurea has also been detected in surface water, groundwater, and drinking water. Available information on ecotoxicological effects of metformin suggests that metformin is a potential endocrine disruptor and thus further emphasising the threat this drug could pose to our environment. This review provides a comprehensive overview of metformin and critically discusses available literature data with respect to its global use/demand, occurrence, fate and ecotoxicity in treatment facilities equipped with conventional and advanced treatment technologies, and its degradation/removal mechanisms. Final section highlights the existing knowledge gaps regarding its ultimate fate under the natural and engineered ecosystems and identifies some important research areas requiring urgent attention from regulatory makers and scientific community.

Antifouling paint applied to fishing vessels is the primary source of dichloro-diphenyl-trichloroethane (DDT) to the coastal marine environments of China. With the aim to provide science-based support of potential regulations on DDT use in antifouling paint, we utilized a fugacity-based model to evaluate the fate and impact of p,p′-DDT, the dominant component of DDT mixture, in Daya Bay and Hailing Bay, two typical estuarine bays in South China. The emissions of p,p′-DDT from fishing vessels to the aquatic environments of Hailing Bay and Daya Bay were estimated as 9.3 and 7.7 kg yr−1, respectively. Uncertainty analysis indicated that the temporal variability of p,p′-DDT was well described by the model if fishing vessels were considered as the only direct source, i.e., fishing vessels should be the dominant source of p,p′-DDT in coastal bay areas of China. Estimated hazard quotients indicated that sediment in Hailing Bay posed high risk to the aquatic system, and it would take at least 21 years to reduce the hazards to a safe level. Moreover, p,p′-DDT tends to migrate from water to sediment in the entire Hailing Bay and Daya Bay. On the other hand, our previous research indicated that p,p′-DDT was more likely to migrate from sediment to water in the maricultured zones located in shallow waters of these two bays, where fishing vessels frequently remain. These findings suggest that relocating mariculture zones to deeper waters would reduce the likelihood of farmed fish contamination by p,p′-DDT.

Plastic is ubiquitous in global oceans and constitutes a newly available habitat for surface-associated bacterial assemblages. Microplastics (plastic particles <5 mm) are especially susceptible to ingestion by marine organisms, as the size of these particles makes them available also to lower trophic levels. Because many marine invertebrates harbour potential pathogens in their guts, we investigated whether bacterial assemblages on polystyrene are selectively modified during their passage through the gut of the lugworm Arenicola marina and are subsequently able to develop pathogenic biofilms. We also examined whether polystyrene acts as a vector for gut biofilm assemblages after subsequent incubation of the egested particles in seawater. Our results showed that after passage through the digestive tract of A. marina, the bacterial assemblages on polystyrene particles and reference glass beads became more similar, harbouring common sediment bacteria. By contrast, only in the presence of polystyrene the potential symbiont Amphritea atlantica was enriched in the investigated biofilms, faeces, and water. Thus, especially in areas of high polystyrene contamination, this polymer may impact the bacterial composition of different habitats, with as yet unknown consequences for the respective ecosystems.

Passive sampling devices (PS) are widely used for pollutant monitoring in water, but estimation of measurement uncertainties by PS has seldom been undertaken. The aim of this work was to identify key parameters governing PS measurements of metals and their dispersion. We report the results of an in situ intercomparison exercise on diffusive gradient in thin films (DGT) in surface waters. Interlaboratory uncertainties of time-weighted average (TWA) concentrations were satisfactory (from 28% to 112%) given the number of participating laboratories (10) and ultra-trace metal concentrations involved. Data dispersion of TWA concentrations was mainly explained by uncertainties generated during DGT handling and analytical procedure steps. We highlight that DGT handling is critical for metals such as Cd, Cr and Zn, implying that DGT assembly/dismantling should be performed in very clean conditions. Using a unique dataset, we demonstrated that DGT markedly lowered the LOQ in comparison to spot sampling and stressed the need for accurate data calculation.

Pollution by Organic Contaminants (OC) in aquatic environments is a relevant issue at the global scale. Lipids comprised of Fatty Acids (FA) play many important roles in the physiology and life history of fishes. Toxic effects of OC are partly dependent on its bioaccumulation in the lipids of aquatic organisms due its physicochemical properties. Therefore, there is an increasing interest to investigate the gene expression as well as the presence and activity of proteins involved in FA metabolism. The attention on Peroxisome Proliferation Activate Receptors (PPARs) also prevails in fish species exposed to OC and in the transport, biosynthesis and β-oxidation of FA. Several studies have been conducted under controlled conditions to evaluate these biological aspects of fish species exposed to OC, as fibrates, endocrine disrupting compounds, perfluoroalkyl acids, flame retardants, metals and mixtures of organic compounds associated with a polluted area. However, only fibrates, which are agonists of PPARs, induce biological responses suitable to be considered as biomarkers of exposure to these pollutants. According to the documented findings on this topic, it is unlikely that these physiological aspects are suitable to be employed as biomarkers with some noticeable exceptions, which depend on experimental design. This emphasises the need to investigate the responses in fish treated with mixtures of OC and in wild fish species from polluted areas to validate or refute the suitability of these biomarkers for environmental or fish health monitoring.

With the increasing environmental application and discharge of iron-based nanoparticles (NPs), a comprehensive understanding of their fate and ecotoxicological effect in the aquatic environment is very urgent. In this study, toxicities of 4 zero-valent iron NPs (nZVI) of different sizes, 2 Fe2O3 NPs of different crystal phases, and 1 type of Fe3O4 NPs to a green alga (Chlorella pyrenoidosa) were investigated, with a focus on the effects of particle size, crystal phase, oxidation state, and environmental aging. Results show that the algal growth inhibition of nZVI increased significantly with decreasing particle size; with similar particle sizes (20–30 nm), the algal growth inhibition decreased with oxidation of the NPs with an order of nZVI > Fe3O4 NPs > Fe2O3 NPs, and α-Fe2O3 NPs presented significantly higher toxicity than γ-Fe2O3 NPs. The NP-induced oxidative stress was the main toxic mechanism, which could explain the difference in algal toxicity of the NPs. The NP-cell heteroagglomeration and physical interactions also contributed to the nanotoxicity, whereas the effect of NP dissolution was negligible. The aging in distilled water and 3 surface water samples for 3 months increased surface oxidation of the iron-based NPs especially nZVI, which decreased the toxicity to algae. These findings will be helpful for the understanding of the fate and toxicity of iron-based NPs in the aquatic environment.

This paper presents the results of an experiment carried out for the first time in situ to select a treatment to devitalize mosses for use in active biomonitoring of water pollution. Three devitalizing treatments for the aquatic moss Fontinalis antipyretica were tested (i.e. oven-drying at 100 °C, oven-drying with a 50-80-100 °C temperature ramp, and boiling in water), and the effects of these on loss of material during exposure of the transplants and on the accumulation of different heavy metals and metalloids were determined. The suitability of using devitalized samples of the terrestrial moss Sphagnum denticulatum to biomonitor aquatic environments was also tested. The structure of mosses was altered in different ways by the devitalizing treatments. Devitalization by boiling water led to significantly less loss of material (p < 0.01) than the oven-drying treatments. However, devitalization by oven-drying with a temperature ramp yielded more stable results in relation to both loss of material and accumulation of elements. With the aim of standardizing the moss bag technique, the use of F. antipyretica devitalized by oven-drying with a temperature ramp is recommended, rather than other devitalization treatments or use of S. denticulatum.

Many heavy metals in sediments and water have potential adverse effects on aquatic organisms such as Corbicula fluminea (O.F. Müller, 1774), a bivalve species frequently used as a biomonitor for metal pollution. Studies over the past decades examining the heavy metal uptake by C. fluminea, very few has investigated the effect of hydrodynamic conditions on accumulation of heavy metal by C. fluminea. Therefore, in this study, to investigate the mechanism of intracellular and extracellular accumulation of metal, individuals of C. fluminea were exposed to cadmium (Cd)-treated water under three different hydrodynamic conditions. These included exposures in two set ups: three rates of rotation (500, 350, 200 r/min) in beakers for 10 days, and then exposure to Cd-treated sediment under two naturally turbulent water conditions (14 cm/s and 3.2 cm/s) in experimental flumes for 23 days. Hydrodynamic force increased the burrowing rate but decreased the activity of C. fluminea. After 10 days of exposure, the extracellular concentrations of Cd in the tissues of C. fluminea in the sand group were significantly higher than that in the gravel groups. The intracellular and extracellular concentrations of Cd in the tissues of C. fluminea dramatically increased in the Cd-treated sediment test. Moreover, the concentration of the extracellular Cd adsorbed on the tissues of C. fluminea in the 14 cm/s and 3.2 cm/s groups was significantly higher than that in the control group, whereas the effect of hydrodynamic force on absorption of Cd by C. fluminea was not obvious. These results suggest that hydrodynamic condition plays an important role in extracellular accumulation of Cd by C. fluminea. In future study, when using C. fluminea to assess Cd pollution in aquatic environment, extracellular Cd adsorbed on the tissue should be removed to avoid the influence of hydrodynamics.

Harbours, as strategic places in tourism and transportation, are exposed to many sources of contamination. Assessing the quality of harbours sediment by guidelines and regulations does not reflect the actual level of contamination and the risk posed to aquatic ecosystems. Selection of an appropriate management technique for contaminated sediments in those strategic locations is crucial for the aquatic environment. The purpose of this study is to show that insufficient information, provided by sediment quality guidelines (SQGs) to identify the actual contaminants, could lead to a destructive or potentially ineffective decision for risk reduction in contaminated harbours. A comprehensive evaluation on physicochemical characteristics of sediment and water samples of a shallow harbour in St. Lawrence River was performed. Results of trace metal fractionation and risk assessment indicated that Cd and Pb were the contaminants that could pose a threat to aquatic ecosystem, although the SQG outcomes implied that Cu and Zn may cause an adverse effect on the benthic organisms. The results of multivariate statistical analysis demonstrated that the locations in the vicinity of the maintenance area contained the most contaminated sediment samples and require appropriate management. Antifouling paint particles and probably the runoff entering the harbour were the main sources of pollution. Among the diverse range of management strategies, the resuspension technique is suggested as a viable alternative in this specific case for shallow locations with contaminated sediments. A suitable management strategy could reduce the cost of remediation process by identifying the actual contaminated spots and also reduce the risk of remobilization of trace metals.
