Atrazine triggers developmental abnormality of ovary and oviduct in quails (Coturnix Coturnix coturnix) via disruption of hypothalamo-pituitary-ovarian axis

There has been a gradual increase in production and consumption of atrazine (ATR) in agriculture to meet the population rising demands. Female reproduction is necessary for growth and maintenance of population. However, ATR impact on females and particularly ovarian developmental toxicity is less clear. The aim of this study was to define the pathways by which ATR exerted toxic effects on ovarian development of ovary and hypothalamo-pituitary-ovarian (HPO) axis. Female quails were dosed by oral gavage from sexual immaturity to maturity with 0, 50, 250 and 500 mg ATR/kg/d for 45 days. ATR had no effect on mortality but depressed feed intake and growth and influenced the biochemical parameters. Notably, the arrested development of ovaries and oviducts were observed in ATR-exposed quails. The circulating concentrations of E2, P, LH and PRL were unregulated and FSH and T was downregulated in ATR-treated quails. The mRNA expression of GnRH in hypothalamo and LH in pituitary and FSH in ovary was downregulated significantly by ATR exposure and FSH and PRL in pituitary were upregulated. ATR exposure upregulated the level of P450scc, P450arom, 3β-HSD and 17β-HSD in ovary and downregulated ERβ expression in female quails. However, ATR did not change ERα expression in ovary. This study provides new insights regarding female productive toxicology of ATR exposure. Ovary and oviduct in sexually maturing females were target organs of ATR-induced developmental toxicity. We propose that ATR-induced developmental abnormality of ovary and oviduct is associated with disruption of gonadal hormone balance and HPO axis in female quails.

Embryonic growth of the yellow-spotted salamander (Ambystoma maculatum) is enhanced by the presence of the green alga Oophila amblystomatis, in the egg capsule. To further assess potential impacts of herbicides on this relationship, A. maculatum egg masses were exposed to atrazine (0–338 μg/L) until hatching (up to 66 days). Exposure to atrazine reduced PSII yield of the symbiotic algae in a concentration-dependent manner, but did not significantly affect visible algal growth or any metrics associated with salamander development. Algal cells were also cultured in the laboratory for toxicity testing. In the 96-h growth inhibition test (0–680 μg/L), ECx values were generally greater than those reported for standard algal test species. Complete recovery of growth rates occurred within 96-h of transferring cells to untreated media. Overall, development of A. maculatum embryos was not affected by exposure to atrazine at concentrations and durations exceeding those found in the environment.

Contaminant concentrations are reported for surface water, sediment, flora and fauna collected during 2010–2011 from the mangrove fringe along eastern Tampa Bay, Florida. Concentrations of trace metals, chlorinated pesticides, atrazine, total polycyclic aromatic hydrocarbons, and polychlorinated biphenyls were species-, chemical- and location-specific. Contaminants in sediments did not exceed proposed individual sediment quality guidelines. Most sediment quality assessment quotients were less than one indicating the likelihood of no inhibitory effect based on chemical measurements alone. Faunal species typically contained more contaminants than plant species; seagrass usually contained more chemicals than mangroves. Bioconcentration factors for marine angiosperms were usually less than 10 and ranged between 1 and 31. Mercury concentrations (ppm) in blue crabs and fish did not exceed the U.S. Environmental Protection Agency fish tissue criterion of 0.3 and the U.S. Food and Drug Administration action level of 1.0. In contrast, total mercury concentrations in faunal species often exceeded guideline values for wildlife consumers of aquatic biota.

This paper deals with the detection and quantification of APIs and other priority substances in the Arade River estuary (Portugal) providing the usefulness of Polar Organic Compound Integrative Samplers (POCIS). Thirteen APIs were detected whose variation was site and time dependent. Caffeine was at the highest concentration (804±209ng/L) followed by theophylline (184±44ng/L). Other APIs were analgesic, anticonvulsant, non-steroidal anti-inflammatory drugs, anti-lipidemic, anxiolytic and antidepressants. Twenty pesticides comprising atrazine, diuron, isoproturon, terbutryn and simazine included in the Water Framework Directive priority list were also site and time dependent. Carbendazim occurred at the highest concentration (45±18ng/L at site 1) but atrazine, diuron, isoproturon and simazine levels were below the Environmental Quality Standards. Although the summer impact was unclear, the results highlighted POCIS suitability for profiling these contaminants. This is to our knowledge the first study concerning APIs and pesticides in this area.

Suspended sediments can decrease the apparent bio-concentration factor of organic pollutants through adsorption. However, whether this process also weakens the toxicity of organic pollutants to non-target aquatic organisms is not clear. Therefore, natural sediments were chosen as suspended sediment examples in this research applying atrazine as the target pollutant and Brachydanio rerio (more recently, Danio rerio (Zebrafish)) as the target organism to conduct acute toxicity experiments. The concentration of atrazine in aqueous solution was measured as a time series. Results show that without suspended sediments, the 96-h LC₅₀of atrazine to Brachydanio rerio is 29.06 mg/l at 95 % confidence interval (24.41 to 40.70 mg/l). For suspended sediments of 7500 and 15,000 mg/l, the LC₅₀(i.e., concentration resulting in 50 % mortality) equates to 30.74 and 39.51 mg/l, respectively, and the corresponding confidence intervals are between 27.17 and 40.91 mg/l and between 30.43 and 126.93 mg/l in that order. Probit analysis, which is a type of regression used to analyze binomial response variables, was applied using the Statistical Package for the Social Sciences. For the series of no suspended solids (SS), 7500 and 15,000 mg/l SS, the so-called no-observed-effective concentrations were 3, 9, and 15 mg/l, correspondingly. The uptake quantity and uptake rate of atrazine by B. rerio according to atrazine concentrations in the aqueous solution were computed. The research indicates that suspended sediments can decrease the absorbed rate of atrazine by B. rerio. Thus, suspended sediments weaken the acute toxicity of atrazine to B. rerio.

Glyphosate applied in association with atrazine provides the best cost/benefit for weed control for genetically modified corn. Therefore, the aim of this work was to evaluate the effects of applying glyphosate in mixture with atrazine on soil microbial population and on herbicides fate in a representative Oxisol from Brazil. The treatments consisted in applying the recommended field rate of glyphosate in association with 0, 1, and 2 times the recommended field rate of atrazine (and vice versa), plus the control (without herbicides application). The presence of atrazine temporarily (21 days) decreased soil microbial biomass (SMB) and increased soil carbon mineralization (SCₘᵢₙ, up to 13 times) and microbial metabolic quotient (qCO₂) due to the stresses caused by its toxicity. When the mixture was applied independent of the rates, SMB was recovered and the amounts of extractable and non-extractable¹⁴C-residues were the same for both herbicides at 63 days. These results suggest that glyphosate may mitigate atrazine’s temporary impact on soil microbes by supplying them nutrients during their adaptation.

A series of column experiments was conducted to evaluate the effect of organic carbon fraction on long-term atrazine elution tailing for calcareous soil (foc = 0.97 %) and calcareous soil with 10 % by weight terra rossa amendment (foc = 1.20 %). Effluent atrazine concentrations were monitored for approximately 400 pore volume to understand the influence of controlling sorption–desorption kinetics on long-term tailing behavior. Laboratory studies showed that the sorption of atrazine was described by rate-limited, nonlinear reversible processes for both soils. Atrazine transport exhibited extensive elution tailing for all experiments due to the presence of hard carbon components such as black carbon and kerogen in both soils. This nonlinear sorption and extensive atrazine tailing behavior were more pronounced and extensive for soil with terra rossa amendment due to the addition of approximately 20 % organic carbon including 10 % hard carbon components from terra rossa soil. A mathematical model incorporating nonlinear, rate-limited sorption/desorption described by a continuous distribution function was used to successfully simulate atrazine transport early-time breakthrough and long-term concentration tailing for both porous media.

Chemical contamination of water resources on the planet generates a range of environmental disturbances which impair ecosystems. Humans ingest such chemicals often present in water. Conventional treatments fail to remove these contaminants from water, requiring complementary methods such as activated carbon filters, reverse osmosis, or distillation, which are expensive and seldom used in the public water supply. In recent years, there has been a search for alternative eco-friendly, low-cost methods which can effectively remove these contaminants. This study was conducted to test the effectiveness of biotite (black mica), an igneous mineral of the mica group, in removing pesticides from water. A trial was designed to assess the rate of pesticide removal using a methodology based on axes of variation of pH, temperature, concentration, and time. The pesticides tested were atrazine, fluazifop-p-butyl, lambda-cyhalothrin, chlorpyrifos, and lactofen. The results showed higher removal rates in acidic conditions (pH 3) and temperatures between 20 and 30 °C, requiring about 6 h to reach maximum adsorption. More than 80 % of all the pesticides were adsorbed. The best result was obtained for fluazifop (94.2 %) in 6 h, under pH 3, and temperature of 25 °C. The study revealed that biotite has a high absorption capacity of complex and varied compounds. These findings signal the need for further studies and tests. Due to the high cost of pesticide analysis, which can only be made using a chromatograph mass spectrometer, financial resources will be required.

Pyrolyzing municipal wastewater treatment sludge into biochar can be a promising sludge disposal approach, especially as the produced sludge-derived biochar (SDBC) is found to be an excellent sorbent for heavy metals and atrazine. The aim of this study was to investigate how and why the coexisting humic acids influence the sorption capacity, kinetic, and binding of these contaminants on SDBC surface. Results showed humic acids enhanced Pb(II)/Cr(VI) sorption binding, and increased the corresponding Pb(II) Langmuir sorption capacity at pH 5.0 from 197 to 233 μmol g⁻¹, and from 688 to 738 μmol g⁻¹ for Cr(VI) at pH 2.0. It can be mainly attributed to the sorbed humic acids, whose active functional groups can offer the additional sites to form stronger inner-sphere complexes with Pb²⁺, and supply more reducing agent to facilitate the transformation of Cr(VI) to Cr(III). However, humic acids reduced the atrazine adsorption Freundlich constant from 1.085 to 0.616 μmol g⁻¹. The pore blockage, confirmed by the decreased BET-specific surface area, as well as the more hydrophilic surface with more sorbed water molecules may be the main reasons for that suppression. Therefore, the coexisting humic acids may affect heavy metal stabilization or pesticide immobilization during SDBC application to contaminated water or soils, and its role thus should be considered especially when organic residues are also added significantly to increase the humic acid content there.

A multi-residue method, based on gas chromatography coupled to tandem mass spectrometry (GC-MS/MS), has been developed for the determination of 70 organic micropollutants from various chemical classes (organochlorinated, organophosphorous, triazines, carbamate and urea, polycyclic aromatic hydrocarbons, polychlorinated biphenyls, pharmaceuticals, phenols, etc.) in surface waters. A single-step SPE extraction using OASIS HLB cartridges was employed for the recovery of target micropollutants. The method has been validated according to monitoring performance criteria of the Water Framework Directive, taking into account the approved guidelines on quality assurance and quality control. The recoveries ranged from 60 to 110 %, the coefficient of variation from 0.84 to 27.4 %, and the uncertainty from 6 to 37 %. The LOD varied from 6.0 to 40 ng/L. The limits of quantification for the priority pollutants anthracene, alachlor, atrazine, benzo(a)pyrene, chlorfenvinphos, diuron, isoproturon, nonylphenol, simazine, and terbutryn fulfill the criterion of <30 % of the relevant environmental standards. The method was employed to investigate the water quality in the basin of a transboundary river, Strymonas, in NE Greece during three sampling campaigns conducted in the year 2013. Thirty-nine compounds were detected in the river water. Metolachlor, diuron, isoproturon, salicylic acid, chlorfenvinphos, 1,2-benzanthracene, pyrene, diflubenzuron, and carbaryl exhibited the highest detection frequencies.
