Isolation and molecular characterization of bacteria to heavy metals isolated from soil samples in Bokaro Coal Mines, India

In recent years, environmental pollution by coal mining is a long-established human activity affecting all levels of life with various environmental impacts by generating heavy metals. The presence of heavy metals even in trace amount is toxic and detrimental to all living organisms. The coal mine area in Bokaro is one of the “Toxic Hotspot” in India. Bacteria have evolved uptake and efflux mechanisms to adapt in heavy metals contaminated environments and thus represent a potential source for bioremediation processes. In the present study, we isolated and characterized eight heavy metal resistant bacteria (NK-1 to 8) from soil sample in Bokaro coal mines, India. Isolates were selected based on high level of heavy metal resistance and its biochemical characterization. The following bacteria were identified based on 16S rRNA gene sequencing Enterobacter ludwigii (KM029957; NK-1), Klebsiella pneumonia (KM029958; NK-2), Enterobacter ludwigii (KM029959; NK-3), Enterobacter ludwigii (KM029960; NK-4), Klebsiella oxytoca (KM029961; NK-5), Enterobacter cloacae (KM029962; NK-6), Acinetobacter gyllenbergii (KM029963; NK-7), Enterobacter cloacae (KM029964; NK-8). A high degree of metal resistance associated with multiple antibiotic resistances was also detected in the selected isolate which was confirmed by the presence of plasmid. These isolates can further be used for bioremediation of heavy metals from contaminated site.

In this study, the effectiveness of bioremediating 1,2-dichloroethane (DCA)-contaminated groundwater under different oxidation–reduction processes was evaluated. Microcosms were constructed using indigenous bacteria and activated sludge as the inocula and cane molasses and a slow polycolloid-releasing substrate (SPRS) as the primary substrates. Complete DCA removal was obtained within 30 days under aerobic and reductive dechlorinating conditions. In anaerobic microcosms with sludge and substrate addition, chloroethane, vinyl chloride, and ethene were produced. The microbial communities and DCA-degrading bacteria in microcosms were characterized by 16S rRNA-based denatured-gradient-gel electrophoresis profiling and nucleotide sequence analyses. Real-time polymerase chain reaction was applied to evaluate the variations in Dehalococcoides spp. and Desulfitobacterium spp. Increase in Desulfitobacterium spp. indicates that the growth of Desulfitobacterium might be induced by DCA. Results indicate that DCA could be used as the primary substrate under aerobic conditions. The increased ethene concentrations imply that dihaloelimination was the dominate mechanism for DCA biodegradation.

The potential arsenite bioteansformation activity of arsenic was investigated by examining bacterial arsenic arsenite-oxidizing gene such as aoxS, aoxR, aoxA, aoxB, aoxC, and aoxD in high arsenic-contaminated drinking water produced from the surface water of floating houses. There is a biogeochemical cycle of activity involving arsenite oxidase aox system and the ars (arsenic resistance system) gene operon and aoxR leader gene activity in Alcaligenes faecalis SRR-11 and aoxS leader gene activity in Achromobacter xylosoxidans TSL-66. Batch experiments showed that SRR-11 and TSL-66 completely oxidized 1 mM of As (III) to As (V) within 35–40 h. The leaders of aoxS and aoxR are important for gene activity, and their effects in arsenic bioremediation and mobility in natural water has a significant ecological role because it allows arsenite oxidase in bacteria to control the biogeochemical cycle of arsenic-contaminated drinking water produced from surface water of floating houses.

A non invasive sampling and remediation strategy was developed and implemented at shoreline contaminated with spilt diesel. To treat the contamination, in a practical, cost-effective, and safe manner (to personnel working on the stockpiles and their ship loading activity), a non-invasive sampling and remediation strategy was designed and implemented since the location and nature of the impacted geology (rock fill) and sediment, precluded conventional ex-situ and any in-situ treatment where drilling is required. A bioremediation process using surfactant, and added N & P and increased aeration, increased the degradation rate allowing the site owner to meet their regulatory obligations. Petroleum hydrocarbons decreased from saturation concentrations to less than detectable amounts at the completion of treatment.

Arsenic contamination is turning out to be a major problem these days with its area coverage and the number of people affected directly or indirectly. Now, the level of the contaminant has spread over the soil and sediments from groundwater and other natural sources. Arsenic poisoning in groundwater events is familiar to the world, but the consequences of soil contamination are still unrevealed to the community, specially the people of contaminated counties. Arsenic is a serious instantaneous concern for the people and other life forms regarding the poisoning through crops and vegetables. Many remediation technologies that mainly include physical, chemical, and a few biological methods have been evolved with time to check its effects. The physical and chemical methods for this purpose are often inefficient and/or very expensive, mainly limited to application in aqueous systems, and produce toxic sludge, which again becomes a matter of concern. But bioremediation relies on the fact that biological organisms have the ability to degrade, detoxify, and even accumulate harmful chemicals and offers attractive perspectives for biomonitoring (via biosensors), treatment of wastewater, and the recycling of polluted soils.

The bioremediation efficiency of China’s largest scale Porphyra yezoensis cultivation for removing dissolved nutrients and controlling harmful algae was studied in the radial sandbanks waters of Jiangsu Province in the year 2012–2013. Mean nutrient concentration values in the P. yezoensis cultivation area were significantly lower than those in the non-cultivation area, especially during the cultivation season (p<0.05). Tissue nitrogen and phosphorus contents of seaweeds were 5.99–0.80% (dry weight (DW)) and 0.16–0.19% (DW), respectively. Production of P. yezoensis was 58950.87tons DW. Based on these values, 3688.15tons of tissue nitrogen and 105.61tons of tissue phosphorus were removed by harvesting P. yezoensis. The richness index of the red tide species Skeleton emacostatum declined from 0.32 to 0.05 during the P. yezoensis cultivation season. These results indicate that large-scale cultivation of P. yezoensis can be used to efficiently alleviate eutrophication and control harmful algae blooms in open sea.

This study analyzed the microbial diversity colonizing the surface of an oil sample during its contact with water, off the Trindade Island coast and simulated the efficiency of eight different bioremediation strategies for this environment. The diversity analysis was performed using acrylic coupons that served as the support for an oil inclusion at sea. The coupons were sampled over 30days, and T-RFLP multiplex was employed to access the diversity of fungi, Bacteria and Archaea present on the oil surface. The bioremediation strategies were simulated in a respirometer. The results showed that the bacterial domain was the most dominant in oil colonization and that the richness of the species attached to the oil gradually increases with the exposure time of the coupons. The combination of biostimulation and bioaugmentation with a native population was proven to be an effective strategy for the remediation of oil off the Trindade Island shoreline.

A microbiological survey of the Priolo Bay (eastern coast of Sicily, Ionian Sea), a chronically polluted marine coastal area, was carried out in order to discern its intrinsic bioremediation potential. Microbiological analysis, 16S rDNA-based DGGE fingerprinting and PLFAs analysis were performed on seawater and sediment samples from six stations on two transects. Higher diversity and variability among stations was detected by DGGE in sediment than in water samples although seawater revealed higher diversity of culturable hydrocarbon-degrading bacteria. The most polluted sediment hosted higher total bacterial diversity and higher abundance and diversity of culturable HC degraders. Alkane- and PAH-degrading bacteria were isolated from all stations and assigned to Alcanivorax, Marinobacter, Thalassospira, Alteromonas and Oleibacter (first isolation from the Mediterranean area). High total microbial diversity associated to a large selection of HC degraders is believed to contribute to natural attenuation of the area, provided that new contaminant contributions are avoided.

The present study aims to optimize the slow release biostimulant ball (BSB) for bioremediation of contaminated coastal sediment using response surface methodology (RSM). Different bacterial communities were evaluated using a pyrosequencing-based approach in contaminated coastal sediments. The effects of BSB size (1–5cm), distance (1–10cm) and time (1–4months) on changes in chemical oxygen demand (COD) and volatile solid (VS) reduction were determined. Maximum reductions of COD and VS, 89.7% and 78.8%, respectively, were observed at a 3cm ball size, 5.5cm distance and 4months; these values are the optimum conditions for effective treatment of contaminated coastal sediment. Most of the variance in COD and VS (0.9291 and 0.9369, respectively) was explained in our chosen models. BSB is a promising method for COD and VS reduction and enhancement of SRB diversity.

Although microbes play important roles during the bioremediation process using macrobenthos in degraded environments, their response to macrobenthos bioturbation remains poorly understood. This study used 16S rRNA gene-Illumina Miseq sequencing to investigate the microbial communities and their response to bioturbation by artificially introducing macrobenthos to the mudflat of Sansha Bay, southeast China. A total of 56 phyla were identified, dominated by δ- and γ-Proteobacteria, with a total percentage of over 50%. Others, such as Acidobacteria, Chloroflexi, Bacteroidetes, Planctomycetes and Alphaproteobacteria occupied 4–7% respectively. Eighteen genera indicating the microbial communities response to bioturbation and seasonal change were identified. Bioturbated samples contained more ecologically important genera, and untreated samples contained more genera ubiquitous in marine environments. The physicochemical characteristics did not change significantly probably due to the short time of bioremediation and low survival rate of macrobenthos, confirming that microbial communities are more sensitive and can serve as sentinels for environmental changes.
