Genotoxic Testing of Titanium Dioxide Nanoparticles in Far Eastern Mussels, Mytilus Trossulus

Manufactured nanoparticles (NP) have caused extreme concern about their ecotoxicological effects on the marine systems. In this study, we investigated the biological effects (oxidative stress and genotoxic response) of TiO2-NP at predicted environmental relevant concentrations (0.2 mg/l and 1 mg/l) on marine mussel Mytilus trossulus a dominant member of the far eastern coastal community. The results of the experiment revealed that TiO2-NP when suspended in seawater, formed agglomerates ranging from 400 nm to several μm in diameter. However, TiO2-NP caused obviously oxidative damage on the mussel as evidenced by the significant elevated levels of malondialdehyde (MDA) in the gill and digestive gland. The genotoxic potential of TiO2-NP was assessed by comet assay, which detect primary DNA damage. The gill and digestive gland cells showed significantly enhanced DNA damage for both concentrations of TiO2-NP compared to the control group. These results propose that the TiO2-NP are entering the marine coastal waters can cause genotoxic effect on mollusks and comet assay can be successfully applied as an effective tool for risk assessment of NP on the marine invertebrates. The findings of this study demonstrate that the aggregation of TiO2-NP does not reduced of NP ecotoxicity, but only changes the biological responses.

The common mussel Mytilus galloprovincialis was selected as unique biomonitor species to implement a regional monitoring programme, the CIESM Mediterranean Mussel Watch (MMW), in the Mediterranean and Black Seas. As of today, and upon standardization of the methodological approach, the MMW Network has been able to quantify 137Cs levels in mussels from 60 coastal stations and to produce the first distribution map of this artificial radionuclide at the scale of the entire Mediterranean and Black Seas. While measured 137Cs levels were found to be very low (usually <1 Bq kg-1 wet wt) 137Cs activity concentrations in the Black Sea and North Aegean Sea were up to two orders of magnitude higher than those in the western Mediterranean Basin. Such effects, far from representing a threat to human populations or the environment, reflect a persistent signature of the Chernobyl fallout in this area. © 2007 Elsevier Ltd. All rights reserved.

International audience | Carbamazepine (CBZ) and Hg are widespread and persistent micropollutants in aquatic environments. Both pollutants are known to trigger similar toxicity mechanisms, e.g. reactive oxygen species (ROS) production. Here, their effects were assessed in the zebra mussel Dreissena polymorpha, frequently used as a freshwater model in ecotoxicology and biomonitoring. Single and co-exposures to CBZ (3.9 µg•L-1) and MeHg (280 ng•L-1) were performed for 1 and 7 days. Metabolomics analyses evidenced that the co-exposure was the most disturbing after 7 days, reducing the amount of 25 metabolites involved in protein synthesis, energy metabolism, antioxidant response and osmoregulation, and significantly altering cells and organelles' structure supporting a reduction of functions of gills and digestive glands. CBZ alone after 7 days decreased the amount of α-aminobutyric acid and had a moderate effect on the structure of mitochondria in digestive glands. MeHg alone had no effect on mussels' metabolome, but caused a significant alteration of cells and organelles' structure in gills and digestive glands. Single exposures and the co-exposure increased antioxidant responses vs control in gills and digestive glands, without resulting in lipid peroxidation, suggesting an increased ROS production caused by both pollutants. Data globally supported that a higher number of hyperactive cells compensated cellular alterations in the digestive gland of mussels exposed to CBZ or MeHg alone, while CBZ+MeHg co-exposure overwhelmed this compensation after 7 days. Those effects were unpredictable based on cellular responses to CBZ and MeHg alone, highlighting the need to consider molecular toxicity pathways for a better anticipation of effects of pollutants in biota in complex environmental conditions.

International audience | Because they are widely used, copper oxide nanoparticles (CuO NPs) are likely to enter the aquatic environment and then reach the sediment. We have examined the effect of CuO NPs in the freshwater endobenthic bivalve Corbicula fluminea. Some previous studies have investigated effects at biochemical and physiological levels, but molecular endpoints are still poorly studied despite they are sensitive in early detection of NPs effect. In the present study, we have investigated short-term effects (96 h) of CuO NP (12, 30 nm; 0, 20 and 100 mg/L) using molecular endpoints as well as more conventional biochemical and physiological markers. The expression of antioxidant (CuZnSOD, MnSOD, Cat, Se-GPx, Trxr) and antitoxic (GST-Pi, HSP70, MT, Pgp, MRP1) related genes was measured at the mRNA level while anti-oxidant (SOD, TAC) and antitoxic (GST, ACP) defenses, energetic reserves and metabolism (ETS, Tri, LDH), and cellular damages (LPO) were assessed using a biochemical approach. The filtration rate measured at 96 h provided information at the physiological scale. Gene expression and filtration rate were responsive to CuO NPs but the effects differed according to the NP size. The results suggest that defense mechanisms may have been set up following 30 nm-NP exposure. The response to 12 nm-NP was lower but still showed that exposure to 12 nm-NP led to activation of cellular elimination mechanisms. The lowering of the filtration rate may have protected the organisms from the contamination. However, this raised the question of further repercussions on organism biology. Together, the results (i) indicate that CuO NP may exert effects at different levels even after a short-term exposure and (ii) point out the precocity of molecular response.

Bivalves are useful bioindicators of microplastic contamination in the marine environment for several reasons, such as extensive filter feeding activity, broad geographical distribution, and limited movement capability. This study conducted a nationwide monitoring of microplastic pollution along the Korean coasts using filter-feeding bivalves (including oyster, mussel, and Manila clam) as bioindicators to identify the national contamination level and characteristics of microplastics. Seawater sample was collected from the same sampling stations of oyster and mussel for comparison. Microplastics were widely distributed in both coastal bivalves and waters with mean concentrations of 0.33 ± 0.23 n/g (1.21 ± 0.68 n/individual) in oyster/mussel, 0.43 ± 0.32 n/g (2.19 ± 1.20 n/individual) in Manila clam, and 1400 ± 560 n/m³ in seawater. Despite the lack of significant relationship in the abundance of microplastics, their dominant features such as size, shape, color and polymer type were similar between bivalves and seawater. Fragments (69% for oyster/mussel, 72% for Manila clam, and 77% for seawater), particles smaller than 300 μm (96% for oyster/mussel, 83% for Manila clam, and 84% for seawater) and colorless (79% for oyster/mussel, 85% for Manila clam, 75% for seawater) were the dominant shape, size and color, respectively. The major polymer types were polypropylene, polyethylene, and polyester. The microplastic level in bivalves was relatively high in urbanized areas with a wide diversity of polymer types compared with those in non-urbanized areas, and the proportion of polystyrene in the Korean samples was abundant compared with other regions due to wide use of polystyrene products in Korea. Our result suggests that microplastic contamination is widespread in the Korean coastal environment, and bivalves can reflect the microplastic pollution characteristics of the surrounding waters where they live.

Diet is an important route of mercury (Hg) uptake in marine organisms. Trophic transfer of Hg throughout the food webs may be influenced by various factors, including diet and Hg speciation. Bivalves such as oysters are widely used as bioindicators of trace element pollution such as Hg. Nevertheless, our current knowledge regarding their ability to accumulate Hg from their diet is mainly based on experiments performed using phytoplankton. In their natural environment, oysters feed on a variety of food items including ciliates, detritus, in addition to phytoplankton. The present study aimed at examining the influence of diet composition on the trophic transfer of inorganic Hg (iHg) and methylmercury (MeHg) in the Pacific cupped oyster Crassostrea gigas. The pulse-chase feeding method was used with two radiolabeled food items: a heterotrophic protist (Uronema marinum) and a phytoplanktonic diatom (Thalassiosira pseudonana). Depuration of dietary Hg in the oysters was followed for 50 d. Kinetic parameters including assimilation efficiency (AE) and efflux rate constant (kₑ) were calculated. Our results showed that oysters fed on ciliates assimilated 96 ± 1% and 31 ± 2% of the ingested MeHg and iHg, respectively whereas these elements were similarly assimilated in the oysters fed on phytoplankton (78 ± 3% and 86 ± 4% for MeHg and iHg, respectively). Mercury assimilation in oyster is thus diet dependent (significant differences in AE, p < 0.05), metal species-dependent and likely resulting from variations in Hg bioavailability in the two food items tested and a gut passage time-dependent of the ingested matrix.

Organophosphate flame retardants (OPFRs), as alternatives to polybrominated biphenyl ethers (PBDEs), are frequently detected in various environmental matrices. Owing to urbanization and industrial pollution, co-contamination of OPFRs and heavy metals is ubiquitous in the environment. The toxicity of OPFRs in aqueous phase is a significant concern, but uncertainty still exists regarding the co-toxicity to benthic organisms of OPFRs and metals in sediments. Hence, we explored the physiological response of Corbicula fluminea to OPFRs and Cd in sediments. The results indicated that the antioxidant system in the clams was stimulated in the presence of OPFRs and Cd, and the oxidative stress increased with increasing concentrations of OPFRs. In contrast, the cytochrome P450 (CYP450) content and acetylcholinesterase (AChE) activity were reduced by exposure to both OPFRs and Cd. The cytochrome P450 4 family (CYP4) mRNA expression and OPFR toxicity were lower than those in previously reported experiments conducted in the water phase. Moreover, the expression levels of metallothionein (MT) and AChE mRNA decreased when OPFRs and Cd were present together. The highest integrated biomarker response (IBR) index (IBR = 15.41) was observed in the presence of 45 mg kg⁻¹ Cd + 200 mg kg⁻¹ OPFRs, rather than the 45 mg kg⁻¹ Cd + 400 mg kg⁻¹ OPFRs treatment (IBR = 9.48). In addition, CYP450 and AChE in the digestive glands of C. fluminea exhibited significant correlations with the concentration of the OPFR/Cd mixture (p < 0.01) and could be effective biomarkers for OPFR and Cd co-contamination. The results potentially contribute to more realistic predictions and evaluations of the environmental risks posed by OPFRs in aquatic sediments contaminated with heavy metals, particularly with respect to the risk to benthic organisms.
