Regioselective hydroxylation of carbendazim by mammalian cytochrome P450: A combined experimental and computational study

Carbendazim (CBZ), a broad-spectrum pesticide frequently detected in fruits and vegetables, could trigger potential toxic risks to mammals. To facilitate the assessment of health risks, this study aimed to characterize the cytochrome P450 (CYPs)-mediated metabolism profiles of CBZ by a combined experimental and computational study. Our results demonstrated that CYPs-mediated region-selective hydroxylation was a major metabolism pathway for CBZ in liver microsomes from various species including rat, mouse, minipig, dog, rabbit, guinea pig, monkey, cow and human, and the metabolite was biosynthesized and well-characterized as 6-OH-CBZ. CYP1A displayed a predominant role in the region-selective hydroxylation of CBZ that could attenuate its toxicity through converting it into a less toxic metabolite. Meanwhile, five other common pesticides including chlorpyrifos-methyl, prochloraz, chlorfenapyr, chlorpyrifos, and chlorothalonil could significantly inhibit the region-selective hydroxylation of CBZ, and consequently remarkably increased CBZ exposure in vivo. Furthermore, computational study clarified the important contribution of the key amino acid residues Ser122, and Asp313 in CYP1A1, as well as Asp320 in CYP1A2 to the hydroxylation of CBZ through hydrogen bonds. These results would provide some useful information for the metabolic profiles of CBZ by mammalian CYPs, and shed new insights into CYP1A-mediated metabolic detoxification of CBZ and its health risk assessment.

Polyurethane foam disks (PUF) passive air samplers (PAS) were deployed over 4 sampling periods of 3–5-months (≥ 1 year) at ten urban and rural locations throughout the Tuscany Region. The purpose was to assess the occurrence and seasonal variations of ten current-use pesticides (CUPs). PUF disk extracts were analyzed using GC–MS. The organophosphates insecticides; chlorpyrifos (3–580 pg m−3) and chlorpyrifos-methyl (below detection limit – to 570 pg m−3) presented the highest levels in air, and showed seasonal fluctuation coinciding with the growing seasons. The relative proportion urban/(urban + rural) ranged from 0.4 to 0.7 showing no differences between urban and rural concentrations. Air back trajectories analysis showed air masses passing over agricultural fields and potentially enhancing the drift of pesticides into the urban sites. This study represents the first information regarding CUPs in the atmosphere of Tuscany region using PAS-PUF disk.

Dissipations of three insecticides: chlorantraniliprole, chlorpyrifos-methyl and indoxacarb in apples were studied following their foliar application on apples intended for production of baby food. The apples were sprayed with formulations for control of codling moth (Cydia Pomonella L.) and leafrollers (Tortricidae). Six experiments were conducted; each insecticide was applied individually on dessert apples. A validated gas chromatography-based method with simultaneous electron capture and nitrogen–phosphorus detection (GC-ECD/NPD) was used for the residue analysis. The analytical performance of the method was satisfactory, with expanded uncertainties ≤36% (a coverage factor, k = 2, and a confidence level of 95%). The dissipations of insecticides were studied in pseudo-first-order kinetic models (for which the coefficient of determination, R ² , ranged between 0.9188 and 0.9897). Residues of studied insecticides were below their maximum residue limits of 0.5 mg/kg at an early stage of growth of the fruit. The half-lives of chlorantraniliprole, chlorpyrifos-methyl and indoxacarb were 16–17, 4–6 and 20–24 days, respectively. The initial residue levels declined gradually and reached the level of 0.01 mg/kg in 1 month for chlorpyrifos-methyl, 2 months for chlorantraniliprole and 2.5 months for indoxacarb. To obtain the insecticide residue levels below 0.01 mg/kg, which is the default MRL for food intended for infants and young children, the application of the studied insecticides should be carried out at recommended doses not later then: 1 month before harvest for chlorpyrifos-methyl, 2 months for chlorantraniliprole and 2.5 months for indoxacarb.

The aim of this study was to analyze the impact of organophosphorus (OP) pollutants on oxidative stress and ultrastructural biomarkers in the midgut of the honeybee Apis mellifera collected from three locations that differ in their extent of spraying load with OP insecticides: a weakly anthropised rural site, Bolin which is considered as a reference site; moderately spraying site, El Kaza; and a strongly anthropised urban site, Tiba with a long history of pesticide use. Results showed that high concentrations of chlorpyrifos, malathion, diazinon, chlorpyrifos-methyl, and pirimiphos-methyl were detected in midgut at locations with extensive pesticide spraying. Reduced glutathione content, superoxide dismutase, catalase, and glutathione peroxidase displayed lowest activities in the heavily sprayed location (Tiba). Lipid peroxidation level in the midgut of honeybees in the sprayed locations was found to be significantly higher compared to the reference values. Meanwhile, various ultrastructural abnormalities were observed in the epithelial cells of midgut of honeybees collected from El Kaza and Tiba, included confluent and disorganized microvilli and destruction of their brush border, the cytoplasm with large vacuoles and alteration of cytoplasmic organelles including the presence of swollen mitochondria with lysis of matrices, disruption of limiting membranes, and disintegration of cristae. The nuclei with indented nuclear envelope and disorganized chromatin were observed. These investigated biomarkers indicated that the surveyed honeybees are being under stressful environmental conditions. So, we suggest using those biomarkers in the assessment of environmental quality using honeybees in future monitoring of ecotoxicological studies.

A simple method based on dispersive solid-phase extraction (DSPE) and dispersive liquid–liquid microextraction method based on solidification of floating organic droplets (DLLME-SFO) was developed for the extraction of chlorpyrifos (CP), chlorpyrifos-methyl (CPM), and their main degradation product 3,5,6-trichloro-2-pyridinol (TCP) in tomato and cucumber samples. The determination was carried out by high performance liquid chromatography with ultraviolet detection (HPLC-UV). In the DSPE-DLLME-SFO, the analytes were first extracted with acetone. The clean-up of the extract by DSPE was carried out by directly adding activated carbon sorbent into the extract solution, followed by shaking and filtration. Under the optimum conditions, the proposed method was sensitive and showed a good linearity within a range of 2–500 ng/g, with the correlation coefficients (r) varying from 0.9991 to 0.9996. The enrichment factors ranged from 127 to 138. The limit of detections (LODs) were in the range of 0.12–0.68 ng/g, and the relative standard deviations (RSDs) for 50 ng/g of each analytes in tomato samples were in the range of 3.25–6.26 % (n = 5). The proposed method was successfully applied for the extraction and determination of the mentioned analytes residues in tomato and cucumber samples, and satisfactory results were obtained.

Subsurface lateral flow in both texture-contrast soils and catchments with shallow bedrock is suspected to be a non-point source of contamination of watercourses by pesticides used in agriculture. As a case study, the north of the Beaujolais region (eastern France) provides a favorable environment for such contamination due to its agro-pedo-climatic conditions. Environments seen in the Beaujolais region include intense viticulture, permeable and shallow soils, steep hillslopes, and storms that occur during the periods of pesticide application. Watercourse contamination by pesticides has been widely observed in this region, and offsite pesticide transport by subsurface lateral flow is suspected to be involved in diffuse and chronic presence of pesticides in surface water. In order to confirm and quantify the potential role of such processes in pesticide transfer, an automated trench system has been designed. The trench was set up on a steep farmed hillslope in a texture-contrast soil. It was equipped with a tipping bucket flow meter and an automatic sampler to monitor pesticide concentrations in lateral flow at fine resolution, by means of a flow-dependent sampling strategy. Four pesticides currently used in vine growing were studied to provide a range of mobility properties: one insecticide (chlorpyrifos-methyl) and three fungicides (spiroxamine, tebuconazole, and dimethomorph). With this system, it was possible to study pesticide concentration dynamics in the subsurface lateral flow, generated by substantial rainfall events following pesticide applications. The experimental design ascertained to be a suitable method in which to monitor subsurface lateral flow and related transfer of pesticides.
