Faecal indicator bacteria (FIB) are used for the assessment of faecal pollution and possible water quality deterioration. There is growing evidence that FIB used in temperate regions are not adequate and reliable to detect faecal pollution in tropical regions. Hence, this study evaluated the adequacy of FIB, including total coliforms (TC), Escherichia coli (EC), Enterococci (IEC), and Clostridium perfringens (CP) in the high-altitude, tropical country of Ethiopia. In addition to FIB, for microbial source tracking (MST), a ruminant-associated molecular marker was applied at different water types and altitudes, and faecal pollution risk mapping was conducted based on consensus FIB. The performances of the indicators were evaluated at 22 sites from different water types. The results indicate that EC cell enumeration and CP spore determination perform well for faecal contamination monitoring. Most of the sub-basins of Lake Tana were found to be moderately to highly polluted, and the levels of pollution were demonstrated to be higher in the rainy season than in the post-rainy season. Markers associated with ruminants (BacR) were identified in more than three quarters of the sites. A bacterial pollution risk map was developed for sub-basins of Lake Tana, including the un-gauged sub-basins. We demonstrate how bacterial pollution risk mapping can aid in improvements to water quality testing and reduce risk to the general population from stream bacteria.

Microbiological sea water quality is a public health problem that has serious repercussions in the tourism and economy of Colombia. This study determines the concentrations of Escherichia coli, Enterococcus faecalis and Clostridium perfringens at eleven beach water points and seven streams along the coast of the Department of Atlántico, Colombia. In seawater, total E. coli, E. faecalis and C. perfringens concentrations were found between 16 and 572 cfu/100 mL, 7–450 cfu/100 ml and 2–125 cfu/100 ml, respectively. The highest counts were observed mainly on urbanised beaches and in correspondence with streams whose waters had a high concentration of faecal origin microorganisms, which represent a serious health risk factor for bathers. Relevant efforts have to be addressed to improve the microbiological quality of these beaches by the establishment of efficient wastewater management programs aimed at enhancing the efficiency of the local treatment plant and the control of illegal sewage pouring onto the coast.

Non-point sources of sewage-related pollution in tropical marine waters are difficult to ascertain. Enterococci (ENT) are widely used as indicators of human waste but their efficacy in tropical waters is highly debated due to natural presence in tropical soils. Clostridium perfringens (CP) is often used as a secondary indicator of fecal contamination because its presence indicates sewage, and in tropical waters environmental sources are unlikely. We analyzed a 27-year dataset containing over 29,000 samples collected by the State of Hawaii, to determine a proposed CP standard for detecting human sewage, which has applicability throughout tropical marine waters globally. Measured ENT concentrations were highly correlated with turbidity. In three instances, sewage contamination was not detected by ENT samples alone, and impairments from non-point pollution may be highly misinformed in Hawaii. The EPA should examine relationships between CP and human health and implement CP as the primary FIB in tropical marine waters.

Three microorganisms were assayed to evaluate the microbiological quality in the seawater at a resort on the Mexican Pacific coast, and to test for possible associations among the titers of the various bacteria, their possible correlations with environmental conditions, and with the location of potential wastewater outflows. Significant microorganism levels were found (at Caletilla beach, Hornos beach, and Papagayo beach, respectively: for Enterococcus 157, 153, and 149, for C. perfringens 35, 89, and 56, for S. aureus 244,137, and 279CFU/100ml), often in excess of the presently set guideline values. In general, bacterial titers were higher during rainy season than in dry season. For S. aureus, in both seasons, highest concentrations were found at 3pm, the time of highest tourist presence at the beaches. Our results argue for the use of these three microorganisms as part of a set of indicators in the routine microbiological evaluation of Mexican beachwaters.

The Estuary of Bahía Blanca (EBB), Argentina, is an important wetland under intense sewage pollution. We investigated the occurrence of Clostridium perfringens (CP) in populations of two benthic crabs (Neohelice granulata and Cyrtograpsus angulatus) and in sediment from the EBB. CP was found in 49.1% of the crabs and all of the isolates were identified as type A. The alpha (cpa) and enterotoxin (cpe) encoding genes were identified. Genetic analyses identified 13 novel sequence types, and found no clustering among isolates, suggesting that CP is not part of the crabs’ commensal flora. CP carriage was 51 times more likely in crabs from the area nearest sewage outfalls compared with crabs from a reference site. Our in vitro experiments suggest that the carriage of CP in crabs is transient. The use of these benthic crabs as monitoring organisms of sewage pollution in coastal habitats is proposed.

Sewage is a major contributor to pollution problems involving human pathogens in tropical coastal areas. This study investigated the occurrence of intestinal protozoan parasites (Giardia and Cryptosporidium) in tropical recreational marine waters contaminated with sewage. The potential risks of Cryptosporidium and Giardia infection from recreational water exposure were estimated from the levels of viable (oo) cysts (DIC+, DAPI+, PI−) found in near-shore swimming areas using an exponential dose response model. A Monte Carlo uncertainty analysis was performed in order to determine the probability distribution of risks. Microbial indicators of recreational water quality (enterococci, Clostridium perfringens) and genetic markers of sewage pollution (human-specific Bacteroidales marker [HF183] and Clostridium coccoides) were simultaneously evaluated in order to estimate the extent of water quality deterioration associated with human wastes. The study revealed the potential risk of parasite infections via primary contact with tropical marine waters contaminated with sewage; higher risk estimates for Giardia than for Cryptosporidium were found. Mean risks estimated by Monte Carlo were below the U.S. EPA upper bound on recreational risk of 0.036 for cryptosporidiosis and giardiasis for both children and adults. However, 95th percentile estimates for giardiasis for children exceeded the 0.036 level. Environmental surveillance of microbial pathogens is crucial in order to control and eradicate the effects that increasing anthropogenic impacts have on marine ecosystems and human health.

The search for alternative indicators of disease-risk from non-enteric pathogens at the beach revealed high densities of targeted bacteria. To explain the high numbers of potential non-enteric pathogens, Staphylococcus aureus and Pseudomonas aeruginosa, in beach sand, we investigated factors affecting their survival and distribution, as well as those of a potential fecal indicator, Clostridium perfringens. Results indicated greater S. aureus and P. aeruginosa survival and proliferation in sterile beach sand, than seawater, with diminished numbers upon exposure to natural micro-predators. C. perfringens remained relatively consistent with initial numbers. Intermediate sand particles (850μm–2mm) constituted the major micro-niche; creating implications for beach classification programs. Colonization of sterile sand boxes at the beach by S. aureus and P. aeruginosa confirmed the filtering action (>100×) of beach sand. The use of these potential pathogens in periodic sanitary evaluation of beach sand quality is indicated, regardless of the factors influencing their abundance.

Spatial and temporal patterns of coastal microbial pollution are not well documented. Our study examined these patterns through measurements of fecal indicator bacteria (FIB), nutrients, and physiochemical parameters in Hilo Bay, Hawai'i, during high and low river flow. >40% of samples tested positive for the human-associated Bacteroides marker, with highest percentages near rivers. Other FIB were also higher near rivers, but only Clostridium perfringens concentrations were related to discharge. During storms, FIB concentrations were three times to an order of magnitude higher, and increased with decreasing salinity and water temperature, and increasing turbidity. These relationships and high spatial resolution data for these parameters were used to create Enterococcus spp. and C. perfringens maps that predicted exceedances with 64% and 95% accuracy, respectively. Mapping microbial pollution patterns and predicting exceedances is a valuable tool that can improve water quality monitoring and aid in visualizing FIB hotspots for management actions.

While interest in and adoption of graywater reuse for irrigation has rapidly grown in recent years, little is known about the long-term effects of graywater irrigation. Concerns exist in relation to the presence of pathogenic organisms, fate of personal care products, and accumulation of salts. The purpose of this research was to evaluate the long-term effects of graywater irrigation to soil quality under real conditions where homeowners may not always apply graywater in a highly controlled manner. Four households from different climatic and geological conditions were selected for sampling (AZ, CA, CO, and TX) where graywater was applied for irrigation for a minimum of 5 years. Soil samples were collected in areas irrigated with graywater and areas irrigated with freshwater within the same yard. Soil cores were taken at depths of 0–15, 15–30, and 30–100 cm and analyzed separately for surfactants, antimicrobials, sodium adsorption ratio (SAR), electrical conductivity (EC), extractable boron, fecal indicator organisms (E. coli, enterococci, and Clostridium perfringens), and soil dehydrogenase activity. In surface soil samples (0–15 cm), the average total surfactant concentration (over all sites) was higher in graywater-irrigated soil (0.078 ± 0.033) compared to freshwater-irrigated soil (0.030 ± 0.025 mg kg⁻¹). This difference was not found to be significant (P > 0.05). Triclosan and triclocarban were detected in surface soil samples at some locations (3.8–6.3 and 3.5–9.1 μg kg⁻¹, respectively), but not in samples deeper than 15 cm. Among the sampling locations, the TX household appeared to be most impacted by graywater, as evidenced by elevated SAR, potentially toxic levels of B, and relatively high numbers of E. coli and enterococci due to 30 years of graywater application for irrigation.

A comparative study of indicator bacteria concentrations obtained by laboratory analysis of grab samples of storm water, which were collected manually or by automatic samplers, was carried out in two urban catchments. Samples were analyzed for four types of indicator bacteria, total coliforms, Escherichia coli (E. coli), enterococci, and Clostridium perfringens and further documented by measurements of total suspended solids (TSS) and turbidity. Analysis of complete data sets (N = 198) indicated no statistically significant differences in the geometric means of all the constituent samples collected automatically or manually, but there were some small differences between the results produced by the two sampling methods applied. Total coliform concentrations were positively biased in samples collected by automatic samplers, but for the three remaining indicator bacteria (E. coli, enterococci, and C. perfringens), the opposite was true. Risk of sample cross-contamination in automatic samplers was assessed in the laboratory by sampling consecutively synthetic storm water with high and low concentrations of E. coli and enterococci. The first low-concentration samples preceded by high-concentration samples were cross-contaminated and the measured concentrations were positively biased. This cross-contamination was explained by storm-water residue in the sampling line. Such a residue remained in place even after line purging by compressed air, and its mass depended on the sampling line length (tested up to 5 m), as verified by measurements in the laboratory. The study findings should be helpful for improving field protocols for indicator bacteria sampling.
