Understanding aquaporin regulation defining silicon uptake and role in arsenic, antimony and germanium stress in pigeonpea (Cajanus cajan)

Understanding of aquaporins (AQPs) facilitating the transport of water and many other small solutes including metalloids like silicon (Si) and arsenic (As) is important to develop stress tolerant cultivars. In the present study, 40 AQPs were identified in the genome of pigeonpea (Cajanus cajan), a pulse crop widely grown in semi-arid region and areas known to affected with heavy metals like As. Conserved domains, variation at NPA motifs, aromatic/arginine (ar/R) selectivity filters, and pore morphology defined here will be crucial in predicting solute specificity of pigeonpea AQPs. The study identified CcNIP2-1 as an AQP predicted to transporter Si (beneficial element) as well as As (hazardous element). Further Si quantification in different tissues showed about 1.66% Si in leaves which confirmed the predictions. Furthermore, scanning electron microscopy showed a higher level of Si accumulation in trichomes on the leaf surface. A significant alleviation in level of As, Sb and Ge stress was also observed when these heavy metals were supplemented with Si. Estimation of relative water content, H₂O₂, lipid peroxidation, proline, total chlorophyll content and other physiological parameters suggested Si derived stress tolerance. Extensive transcriptome profiling under different developmental stages from germination to senescence was performed to understand the tissue-specific regulation of different AQPs. For instance, high expression of TIP3s was observed only in reproductive tissues. Co-expression network developed using transcriptome data from 30 different conditions and tissues, showed interdependency of AQPs. Expression profiling of pigeonpea performed using real time PCR showed differential expression of AQPs after Si supplementation. The information generated about the phylogeny, distribution, molecular evolution, solute specificity, and gene expression dynamics in article will be helpful to better understand the AQP transport system in pigeonpea and other legumes.

Rice paddy fields are major sources of atmospheric methane (CH₄) and nitrous oxide (N₂O). Rice variety is an important factor affecting CH₄ and N₂O emissions. However, the interactive effects of rice metabolites and microorganisms on CH₄ and N₂O emissions in paddy fields are not clearly understood. In this study, a high greenhouse gas-emitting cultivar (YL 6) and a low greenhouse gas-emitting cultivar (YY 1540) were used as experimental materials. Metabolomics was used to examine the roots, root exudates, and bulk soil metabolites. High-throughput sequencing was used to determine the microbial community composition. YY 1540 had more secondary metabolites (flavonoids and isoflavonoids) in root exudates than YL 6. It was enriched with the uncultured members of the families Gemmatimonadanceae and Rhizobiales_Incertae_Sedis in bulk soil, and genera Burkholderia-Caballeronia-Paraburkholderia, Magnetospirillum, Aeromonas, and Anaeromyxobacter in roots, contributing to increased expression of pmoA and nosZ genes and reducing CH₄ and N₂O emissions. YL 6 roots and root exudates contained higher contents of carbohydrates [e.g., 6-O- acetylarbutin and 2-(3- hydroxyphenyl) ethanol 1′-glucoside] than those of YY 1540. They were enriched with genera RBG-16-58-14 in bulk soil and Exiguobacterium, and uncultured member of the Kineosporiaceae family in roots, which contributed to increased expression of mcrA, ammonia-oxidizing archaea, ammonia-oxidizing bacteria, nirS, and nirK genes and greenhouse gas emissions. In general, these results established a link between metabolites, microorganisms, microbial functional genes, and greenhouse gas emissions. The metabolites of root exudates and roots regulated CH₄ and N₂O emissions by influencing the microbial community composition in bulk soil and roots.

The present study evaluated the interactive effects of global change and heavy metals on the growth and development of three soybean [Glycine max (L.) Merrill] cultivars and the consequences on yield and food safety. Soybean cultivars (Alim 3.14 from Argentina, and ES Mentor and Sigalia, from Germany) were grown until maturity in heavy metals polluted soils from the Rhine Valley, Germany, at two CO₂ concentrations (400 and 550 ppm) and heat stress (HS) episodes (9 days with 10 °C higher than maximum regular temperature) during the critical growth period in controlled environmental chambers. Different morpho-physiological parameters, heavy metal concentration in aerial organs, seed quality parameters, and toxicological index were recorded. The results showed that no morphological differences were observed related to CO₂. Moreover, Alim 3.14 showed the highest yield under control conditions, but it was more sensitive to climatic conditions than the German cultivars, especially to heat stress which strongly reduces the biomass of the fruits. Heavy metals concentration in soil exceeds the legislation limits for agricultural soils for Cd and Pb, with 1.6 and 487 mg kg⁻¹ respectively. In all cultivars, soybeans accumulated Cd in its aerial organs, and it could be translocated to fruits. Cd concentration in seeds ranged between 0.6 and 2.4 mg kg⁻¹, which exceed legislation limits and with toxicological risk to potential Chinese consumers. Pb levels were lower than Cd in seeds (0.03–0.17 mg kg⁻¹), and the accumulation were concentrated in the vegetative organs, with 93% of the Pb incorporated. Moreover, pods accumulated 11 times more Pb than seeds, which suggests that they act as a barrier to the passage of Pb to their offspring. These results evidence that soybean can easily translocate Cd, but not Pb, to reproductive organs. No regular patterns were observed in relation to climatic influence on heavy metal uptake.

Clearfield® wheat (Triticum aestivum) have helped eliminate the toughest grasses and broadleaf weeds in Spain since 2005. This crop production system includes other tolerant cultivars to the application of imidazolinone (IMI) herbicides. However, the continuous use and off-label rates of IMI herbicides can contribute to the development of resistance in Lolium rigidum and other weed species. In this research, the main objectives were to study the resistance mechanisms to acetolactate synthase (ALS) and acetyl coenzyme A carboxylase (ACCase) inhibitors in a L. rigidum accession (LrR) from a Clearfield® wheat field, with a long history rotating these IMI-tolerant crops and compare them with those present in the IMI-tolerant wheat. The resistance to ACCase inhibitors in LrR was due to point mutations (Ile1781Leu plus Asp2078Gly) of the target site gene plus an enhanced herbicide metabolism (EHM), on the other hand, in wheat accessions was due only by EHM. Mechanisms involved in the resistance to ALS inhibitors were both point mutations of the target gene and EHM in the IMI-tolerant wheat, while only evidence of mutation (Trp574Leu) was found in the multiple herbicide resistant L. rigidum accession. This research demonstrates that if crop rotation is not accompanied by the use of alternative sites of action in herbicide-tolerant crops, resistant weeds to herbicide to which crops are tolerant, can easily be selected. Moreover, repeated and inappropriate use of Clearfield® crops and herbicide rotations can lead to the evolution of multiple resistant weeds, as shown in this study, and have also inestimable environmental impacts.

High cadmium (Cd) concentration in common wheat (Triticum aestivum L.) grains poses potential health risks. Several management strategies have been used to reduce grain Cd concentration. However, limited information is available on the use of ammonium-nitrogen (NH₄⁺-N) as a strategy to manage Cd concentration in wheat grains. In this study, NH₄⁺-N addition at the seedling stage unchanged the grain Cd concentration in the high-Cd accumulator, Zhoumai 18 (ZM18), but dramatically increased that in the low-Cd accumulator, Yunmai 51 (YM51). Further analysis revealed that the effects of NH₄⁺-N addition on whole-plant Cd absorption, root-to-shoot Cd translocation, and shoot-to-grain Cd remobilization were different between the two wheat cultivars. In ZM18, NH₄⁺-N addition did not change whole-plant Cd absorption, but inhibited root-to-shoot Cd translocation and Cd remobilization from lower internodes, lower leaves, node 1, and internode 1 to grains via the down-regulation of yellow stripe-like transporters (YSL), zinc transporters (ZIP5, ZIP7, and ZIP10), and heavy-metal transporting ATPases (HMA2). This inhibition decreased the grain Cd content by 29.62%, which was consistent with the decrease of the grain dry weight by 23.26%, leading to unchanged grain Cd concentration in ZM18. However, in YM51, NH₄⁺-N addition promoted continuous Cd absorption during grain filling, root-to-shoot Cd translocation and whole-plant Cd absorption. The absorbed Cd was directly transported to internode 1 via the xylem and then re-transported to grains via the phloem by up-regulated YSL, ZIP5, and copper transporters (COPT4). This promotion increased the grain Cd content by 245.35%, which was higher than the increased grain dry weight by 132.89%, leading to increased grain Cd concentration in YM51. Our findings concluded that the addition of NH₄⁺-N fertilizer at the seedling stage is not suitable for reducing grain Cd concentration in common wheat cultivars.

Representing the staple crop for half of the world population, rice can accumulate high levels of cadmium (Cd) in its grain, posing concerns on food safety. Different soil amendments have been proposed to decrease Cd accumulation in rice grain by either decreasing soil Cd availability, introducing competitive ions on Cd uptake, or down-regulating the expression of transporters for Cd uptake. However, the effectiveness of soil amendments applied alone or in combinations needs to be tested under field conditions. Here, we present results of field trials with two rice cultivars differing in Cd accumulation grown at three field sites in southern China in two years, to investigate the effects of two Mn-containing soil amendments (MnO₂, Mn-loaded biochar (MB)), Si fertilizer (Si), limestone, and K₂SO₄, as well as interactions among MnO₂, Si, and limestone on decreasing Cd accumulation in rice grain. We found that single applications of MnO₂ or MB to acidic soils low in Mn decreased grain Cd concentrations by 44–53 % or 78–82 %, respectively, over two years without decrease in performance. These effects were comparable to or greater than those induced by limestone liming alone (45–62 %). Strong interactions between MnO₂ and limestone resulting from their influence on soil extractable Cd and Mn led to non-additive effects on lowering grain Cd. MB addition minimized grain Cd concentrations, primarily by increasing extractable and dissolved Mn concentrations, but also by decreasing Cd extractability in soil. In comparison, Si and K₂SO₄ amendments affected grain Cd levels only weakly. We conclude that the amendments that decrease labile Cd and increase labile Mn in soils are most effective at reducing Cd accumulation in rice grain, thus contributing to food safety.
Показать больше [+] Меньше [-]The role of root apoplastic barriers in cadmium translocation and accumulation in cultivars of rice (Oryza sativa L.) with different Cd-accumulating characteristics

The radial translocation of cadmium (Cd) from the root to the shoot is one of the major processes affecting Cd accumulation in rice (Oryza sativa L.) grains, but few studies have focused on Cd apoplastic transport in rice. The aim of this study was to determine how apoplastic barriers affect Cd translocation via the apoplastic pathway, Cd accumulation levels in upper parts (shoot and grains) of rice cultivars, and the possible mechanism involved. Hydroponic and soil pot trials were conducted to study the development and chemical constituents of apoplastic barriers and their permeability to bypass flow, and to determine Cd localization in the roots of rice cultivars with different Cd-accumulating characteristics. The Cd accumulation in upper parts was positively correlated with bypass flow in the root and the apparent Cd concentration in the xylem, indicating that the apoplastic pathway may play an important role in Cd root-shoot translocation in rice. Apoplastic barriers were deposited closer to the root tip and were thicker in low Cd-accumulating cultivars than in high Cd-accumulating cultivars. The amounts and rates of increase in lignin and suberin were significantly higher in ZD14 (a low Cd-accumulating cultivar) than in FYXZ (a high Cd-accumulating cultivar) under Cd stress, indicating that stronger barriers were induced by Cd in ZD14. The stronger and earlier formation of barriers in the low Cd-accumulating cultivar decreased bypass flow more efficiently, so that more Cd was retained in the root during apoplastic translocation. This was confirmed by localization analyses of Cd in root transverse sections. These results suggest that apoplastic barriers reduce Cd root-to-shoot translocation via the apoplastic pathway, leading to lower Cd accumulation in the upper parts of rice plants. Bypass flow may have the potential to be used as a rapid screening indicator for low Cd-accumulating rice cultivars.

Since the Industrial Revolution, the global ambient O3 concentration has more than doubled. Negative impact of O3 on some common crops such as wheat and soybeans has been widely recognized, but there is relatively little information about maize, the typical C4 plant and third most important crop worldwide. To partly compensate this knowledge gap, the maize cultivar (Zhengdan 958, ZD958) with maximum planting area in China was exposed to a range of chronic ozone (O3) exposures in open top chambers (OTCs). The O3 effects on this highly important crop were estimated in relation to two O3 metrics, AOT40 (accumulated hourly O3 concentration over a threshold of 40 ppb during daylight hours) and POD6 (Phytotoxic O3 Dose above a threshold flux of 6 nmol O3 m−2 s−1 during a specified period). We found that (1) the reduced light-saturated net photosynthetic rate (Asat) mainly caused by non-stomatal limitations across heading and grain filling stages, but the stomatal limitations at the former stage were stronger than those at the latter stage; (2) impact of O3 on water use efficiency (WUE) of maize was significantly dependent on developmental stage; (3) yield loss induced by O3 was mainly due to a reduction in kernels weight rather than in the number of kernels; (4) the performance of AOT40 and POD6 was similar, according to their determination coefficients (R2); (5) the order of O3 sensitivity among different parameters was photosynthetic parameters > biomass parameters > yield-related parameters; (6) Responses of Asat to O3 between heading and gran filling stages were significantly different based on AOT40 metric, but not POD6. The proposed O3 metrics-response relationships will be valuable for O3 risk assessment in Asia and also for crop productivity models including the influence of O3.

The emission of mercury (Hg) from cropland soil greatly affects the global Hg cycle. Combinations of different crop cultivars and planting densities will result in different light transmittance under canopies, which directly affects the solar and heat radiation flux received by the soil surface below crops. In turn, this might lead to differences in the soil–air total gaseous mercury (TGM) exchange under different cropping patterns. However, soil–air TGM exchange fluxes in croplands under differing canopies have been poorly investigated. Here, a one-year observation of TGM exchange flux was conducted for cropland soils covering five different crop cultivars and three planting densities in North China Plain using the dynamic flux chamber method. The results showed that light transmittance under the canopies was the key control on soil–air TGM exchange fluxes. High light transmittance can enhance soil TGM emission rates and increase the magnitude of diurnal variations in soil–air TGM exchange fluxes. Furthermore, we found that there were piecewise–function relationships (Peak function–constant equation) between light transmittance under the different canopies and the numbers of days after crop sowing. The soil–air TGM exchange fluxes showed a parabolic response to changes in light transmittance under the different canopies. A second-order model was established for the response relationship between soil–air TGM exchange flux and soil Hg concentration, total solar radiation above the canopy, and numbers of days after sowing. The estimated annual average soil–air TGM exchange flux was 5.46 ± 21.69 ng m⁻² h⁻¹ at corn–wheat rotation cropland with 30 cm row spacing using this second-order model. Our results might a data reference and a promising foundation for future model development of soil–air TGM exchange in croplands under different crop cultivars and planting densities.

There is a lack of understanding about the potential accumulation of antibiotics in plants exposed to low-dose contaminated soil. 12 Brassica rapa subsp. chinensis cultivars were used to investigate the different accumulation capacities of sulfamethoxypyridazine, tetracycline, ofloxacin, norfloxacin and difloxacin from the soil. The results showed a significant variation (p < 0.05) among the 12 cultivars in the accumulation of antibiotics. Cultivars Y1 and Y2 had the highest accumulation capacity with average concentrations of 3.26 and 3.00 μg kg⁻¹, respectively, while cultivars Y4 and Y9 had the lowest accumulation capacity with average concentrations of 0.83 and 0.89 μg kg⁻¹. The average antibiotic concentration in all edible part samples (2.74 μg kg⁻¹) of the treatment group was about 3.0-fold of that of the control group (0.93 μg kg⁻¹). The average bioconcentration factors of sulfamethoxypyridazine, tetracycline, ofloxacin, norfloxacin and difloxacin were 0.051, 0.031, 0.017, 0.036 and 0.034, respectively, indicating a higher uptake of sulfamethoxypyridazine compared to ofloxacin. And the mobility of antibiotics in soil is a main factor affecting the bioavailability for plants. The average concentration of antibiotics in edible parts of cultivar Y12 on the 25th and 45th day were 1.52 and 1.73 μg kg⁻¹ and that of the roots were 3.73 and 6.61 μg kg⁻¹, respectively. The concentrations of tetracycline and difloxacin in the edible parts and roots significantly increased with growing time, while the concentration of sulfamethoxypyridazine and ofloxacin changed little throughout the growing period. The potential risks of antibiotics in vegetables on human health cannot be ignored. Overall, attention should be paid to the translocation of antibiotics from soil to plants.
